Abstract

Chitinases hydrolyzing chitin are produced by various organisms and their physiological functions depend on their sources.Chitinases and its hydrolysis products have a broad range of applications. Increasing commercial interest in chitinase and its products has led to the need of inexpensive, reliable sources of active and stable chitinase preparations.The activity of chitinase produced by Aspergillus rugulosus 501 in liquid culture and the parameters that controlled the enzyme synthesis were studied to find optimal conditions for the enzyme production. Hydrolysis products of chitinase were detected by Reissig method. The results showed that fungal chitinase activity in media containing organic nitrogen and phosphate was higher than that in media containing trace element, organic and anorganic nitrogen but lack of phosphate. The maximum enzyme activity reached in the second day incubation.Chitinase production was affected by the initial pH of media, chitin consentration and source of nitrogen, while the consentrasion of inoculum used in this study did not affect enzyme production. High enzyme activity was found in media with initial pH 5.0-6.5. When using different consentration of colloidal chitin, chitinase activities were also found high in concentration from 0.75% tol .5%. The organic nitrogen sources gave the better results compared to the anorganic ones.

Abstract

Marosatherina ladigesi is one of the famous rainbow fish species from Sulawesi.This endemic fish species from Sulawesi is one of the Indonesian export commodity since more than 30 years ago.All of the export specimens come from the wild habitat.The anxiousness of the extinction of this species stated in the redlist of IUCN since 1994. Two field work of Maros Karst Project conducted in 2006, 2007 and an international expedition in 2007 showed the decreasing population of this species.The results of the three field trips showed the difficulties to get M. ladigesi in the streams.Taxonomical status and classification, coloration, sex dimorphism and distribution discussed.

Abstract

Study in attempt to substitute commercial imported strychnine for elimination wild dogs in rabies disease program with Kimalakian (Croton tiglium) seeds has been conducted. This research was aimed to examine the destructive effect of croton seed petroleum extract (oil) in mice. There were 50 mice divided into 5 treatment groups. Each group was given (orally) 0.05, 0.10, 0.20, 0.40 and 0.80 ml oil/mice respectively and 1 group (10 mice) was blank. After the animal died necropsy was performed and gross pathological and hystopathological features were observed. The results showed that LD!0: 1.66 ml of oil/kg b.w. on mice. Furthermore, it was applied to dogs (b.w. 3kg) with dose: 2.5 ml, 5.0 ml and 10 ml oil/dog respectively. All dogs became hyperactive within few minutes, then dead after dosing 5.0 and 10.0 ml oil. Gross lesion in mices and dogs showed general congestion and hemorrhage in the lung, heart and liver. Histopathologically, the gastric mucosa was atropic resulted from gastric malfunction as leading to death.

Abstract

tRNA " UAA (trnL) intron have been widely utilised for phylogenetic reconstruction in angiosperm families. Two main helical elements of secondary structure of trnL intron (stem loop regions P6 and P8) contained the most variable sequences across taxa.Detailed examination on the insertion-deletion events in trnL intron sequences of 110 species of Dipterocarpaceae has identified 4 stem loop structures within both loops.The first stemJoqp was found in P6 loop and three other were discovered in P8 loop.These structures featuring characteristic for genericJinfra-generic level depended upon which taxonomic classifications are foJJowed.One of these structures formed a large loop of 72 nucleotides and was a unique major stem loop in Dipterocarps family whose loop was the major structural mutation of trnL intron in this family.

Abstract

Photorhabdus luminescens HJ is an entomopathogenic bacterium that has a high toxicity against Tenebrio molitor larvae.Toxicity assay of crude extra cellular protein precipitated using ammonium sulphate showed that the highest toxin activity was found in 70 % saturation. Purification of the toxin using Hi Prep 16/60 Sephacryl S-200 HR column exhibited one fraction of toxic protein and three fractions of non-toxic protein. Mortality of T. molitor larvae treated with 19.2 nanogram of toxic fraction was up to 80%. Denatured protein analysis using sodium dodecyl sulphate-polyacrylamide gel electrophoresis revealed that the toxic fraction was composed of three proteins, which were 19.5, 42, and 66 kDa respectively. Based on toxin activity bioassay, this toxin type was an injectable toxin and presumably classified as Mcf toxin.
