Abstract

Abstract. Infectious diseases are still the main cause of death in several developing countries, one of which is Indonesia. One of the bacteria that causes infections in humans is Escherichia coli . Butterfly pea flowers (Clitoria ternatea L.) contain various secondary metabolites including alkaloids, flavonoids, saponins, tannins, steroids, and terpenoids. Flavonoids are secondary metabolites that have the potential as antibacterials. This study aims to determine the antibacterial power and optimal concentration of ethanol extract of blue butterfly pea leaves that can inhibit the growth of Escherichia coli  bacteria. The extraction method used was soxhletation with 70% ethanol solvent and antibacterial testing used the disc diffusion method with several variations in the concentration of blue butterfly pea leaf extract, namely 10%, 20%, 30%, 40%, and 50%.. Amoxicillin was used as a positive control while DMSO was used for the negative control. Antibacterial activity is indicated by the diameter of the inhibition zone which is then measured and grouped according to the categories very strong, strong, moderate, and weak. Research data on 70% ethanol extract of blue butterfly pea leaves has antibacterial activity against Escherichia coli  bacteria which is included in the weak category. The optimum concentration of ethanol extract of blue butterfly pea leaves that can inhibit Escherichia coli  bacteria is at a concentration of 50% with an inhibition zone diameter of 3.33 mm. Abstrak. Penyakit infeksi masih menjadi penyebab utama kematian di sejumlah negara berkembang, salah satunya adalah Indonesia. Salah satu bakteri yang menyebabkan infeksi pada manusia adalah Escherichia coli. Bunga telang (Clitoria ternatea) mengandung berbagai metabolit sekunder termasuk alkaloid, flavonoid, saponin, tanin, steroid, dan terpenoid. Flavonoid merupakan metabolit sekunder yang memiliki potensi sebagai antibakteri. Penelitian ini bertujuan mengetahui daya antibakteri serta konsentrasi optimal dari ekstrak etanol daun telang biru yang dapat menghambat pertumbuhan bakteri Escherichia coli.. Metode ekstraksi yang digunakan secara soxhletasi dengan pelaur etanol 70% serta pengujian antibakteri menggunakan metode difusi cakram dengan beberapa variasi konsentrasi ekstrak daun telang biru yaitu 10%, 20%, 30%, 40%, dan 50%. Sebagai kontrol positif digunakan Amoxicillin sedangan untuk kontrol negatif digunakan DMSO. Aktivitas antibakteri ditunjukkan dengan diameter zona hambat kemudian diukur dan dikelompokkan berdasarkan kategori sangat kuat, kuat, sedang, dan lemah. Data penelitian ekstrak etanol 70% daun telang biru memiliki aktivitas antibakteri terhadap bakteri Escherichia coli yang termasuk dalam kategori lemah. Konsentrasi optimum ekstrak etanol daun telang biru yang mampu menghambat bakteri Escherichia coli yaitu pada konsentrasi 50% dengan diameter zona hambat sebesar 3,33 mm.

Abstract

The immune system is a complex mechanism consisting of cells, tissues, and organs that work in coordination to protect the body from exposure to pathogens. However, pathogens also possess strategies to counteract immune defenses, thereby necessitating external substances to support immune enhancement. One such substance is antioxidants. Butterfly pea flower (Clitoria ternatea) is a potential candidate as an immunomodulator due to its high antioxidant content, which functions to prevent lipid peroxidation and combat free radicals. This study aims to investigate the immunomodulatory potential of the ethanol extract of butterfly pea flower using the carbon clearance test in mice. The research method was experimental, employing 30 BALB/c mice as test animals, which were divided into six groups: Butterfly Pea Flower Extract (EBTB) at doses of 150, 300, and 600 mg/kg BW, levamisole 25 mg and methylprednisolone 4 mg as comparison groups, and sodium CMC as the negative control group. The substances were administered orally for seven consecutive days, followed by intravenous induction of carbon ink on day 8. The obtained serum was measured for absorbance using UV-Vis spectrophotometry at 650 nm. Parameters observed included the phagocytic index (PI) and lymphoid organ index (liver, spleen, and thymus). Based on the phagocytic index calculation, EBTB at doses of 150 and 300 mg/kg BW yielded values of 2.990 and 2.929, respectively, indicating strong immunostimulant activity, while BPFE at 600 mg/kg BW showed moderate immunostimulant activity with a PI value of 1.423. Results for the lymphoid organ index in the BPFE 150 group did not show significant differences (P > 0.05) compared to the control group. In conclusion, the ethanol extract of butterfly pea flower demonstrates potential as an immunomodulator for nonspecific immune responses.
