Abstract

Bakteri endofit dapat ditemukan dari semua jaringan tanaman tetapi tidak menyebabkan gangguan atau kerusakan pada tanaman tersebut. Tujuan penelitian untuk mengeksplorasi dan mengkarakterisasi morfologi bakteri endofit dari akar tanaman gambir. Sampel akar gambir diambil dari Sumatera Barat, Kabupaten Lima Puluh Kota dari 5 titik Lokasi yang berbeda. Isolasi dilakukan untuk memperoleh isolat tunggal bakteri endofit. Karakterisasi meliputi pengamatan morfologi koloni, uji gram dengan KOH 3% serta uji patogenesitas pada daun tembakau. Hasil isolasi diperoleh 23 isolat tunggal bakteri endofit, dimana dari pengamatan morfologi didominasi oleh bentuk koloni yang tidak beraturan, permukaan yang halus tidak mengkilap, elevasi datar dan warna koloni putih susu. Dari 23 isolat yang diuji gram dengan KOH 3%, sebanyak 11 isolat tergolong gram negatif dan 12 isolat lainnya kelompok gram positif. Uji patogenesitas pada daun tembakau diperoleh hasil sebanyak 7 isolat menunjukkan reaksi hipersensitif yang ditandai dengan gejala nekrosis, sedangkan 16 isolat lainnya tidak menunjukkan gejala nekrosisi pada daun tembakau

Abstract

Endophytic bacteria in a plant tissue do not cause disturbance or damage to the plant. Phytohormones produced by endophytic bacteria can increase plant growth and function as antimicrobials. The study aims to explore, select, and characterize the morphology of endophytic bacteria originating from the stems and roots of coffee plants from Solok Radjo. Morphological observations were carried out after obtaining a single isolate of endophytic bacteria, then a Gram test with 3% KOH and a hypersensitive reaction test on water henna plants were carried out to determine the pathogenicity of the endophytic bacteria. A total of 35 isolates were obtained from the isolation results, namely 23 isolates from the stem and 12 isolates from the roots. The results of morphological observations of endophytic bacterial colonies on Nutrient Agar (NA) media were dominated by irregular colony shapes, smooth and shiny surfaces, flat elevations, and yellowish-white colony colors. Gram testing with 3% KOH obtained 17 isolates classified as Gram-positive and 18 isolates as Gram-negative. Hypersensitive observations that showed symptoms of necrosis (positive reaction) were 19 isolates, and no symptoms of necrosis (negative reaction) were 16 isolates.

Abstract

Jagung merupakan tanaman pangan yang menjadi salah satu komoditas penting di Indonesia. Produksi jagung di Indonesia belum dapat dikatakan memenuhi semua kebutuhan penduduk Indonesia, impor adalah salah satu cara untuk menanggulanginya. Salah satu penghambat produksi jagung di Indonesia ialah adanya organisme pengganggu tanaman. Tujuan penelitian ini adalah untuk mengetahui perbedaan insidensi penyakit tanaman jagung dengan pola tanam secara monokultur dan tumpangsari. Penelitian ini dilaksanakan di kebun percobaan Kampus III Universitas Andalas, Sumatera Barat di bulan Desember 2024. Metode yang dilakukan dalam penelitian ini dengan cara pengamatan tanaman secara langsung dalam suatu lahan dengan menghitung insidensi penyakit pada lahan monokultur dan tumpangsari. Hasil penelitian menunjukkan bahwa penyakit tanaman jagung yang ditumpangsari dengan tanaman kedelai dan lahan monokultur jagung yang diamati ditemukan penyakit yang menyerang antara lain penyakit hawar daun, karat daun, serta penyakit bercak daun. Insidensi penyakit hawar daun di lahan monokultur yaitu 100% dan pada lahan tumpangsari yaitu 83%. Pada penyakit karat daun, insidensi sebesar 45% di lahan monokultur, 30% di lahan tumpangsari. Intensitas penyakit bercak daun sebesar 10% di kedua lahan. Insidensi dan keparahan penyakit lebih besar di lahan monokultur dibandingkan dengan tumpangsari. Hal ini menunjukkan bahwa budidaya tanaman jagung lebih baik di lahan tumpangsari dibandingkan monokultur.

Abstract

Gambir is one of Indonesia’s superior commodities in the international market. However, problems that causes a decrease in the quality and productivity of gambir plants is disease attacks. The percentage and intensity of disease attacks on gambir plants in Lima Puluh Kota Regency have never been reported. This studi aimed termine the level of attack occurrence and intensity of diseases that attack gambir plants as basic information related to plant health status in an effort to manage diseases effectively. The study was conducted in three gambir land locations, using a survey method with Simple Random Sampling. Direct observation includes symptoms of attack, percentage and intensity of disease. The results of observations on land 1 and 2 found five main diseases namely red rust, brown spots, black spots, black mosaic and chlorosis, while on land 3 brown spot disease, dieback and black mosaic were found. The highest incidence of disease on land 1 was red rust which reached 100%, on land 2 red rust and brown spots were 100%, and on land 3 brown spot disease was 96%. The highest intensity of disease attack was red rust reaching 82% which was included in the category of very severe attacks. The high incidence and intensity of attacks of this disease are influenced by the age of the plants and the cultivation techniques used by gambir farmers, such as the lack of weed and disease control and fertilization. Abstract

Soil insects play an important role in maintaining the balance of the ecosystem in plantation areas, both as decomposers of organic matter and as biological control agents. Their presence can be an indicator of soil health and the stability of the agricultural environment. On the other hand, soil insects can be detrimental, namely as pests on plants. This study was conducted with the aim of inventorying the types of soil insects found in coffee plantations (Coffea sp.) Solok Radjo Cooperative, Solok Regency. Sampling was carried out from July to September 2025 using the pitfall trap method with a diagonal transect pattern of 100 meters. Identification of soil insects was carried out at the Integrated Pest Management Laboratory, Andalas University, Dharmasraya Campus. Identification based on morphological characteristics of soil insects with the help of a microscope and insect identification keys. The results based on research conducted in the Solok Radjo coffee plantation, Solok Regency, identified consist of 5 orders of soil insects, namely Coleoptera, Hymenoptera, Blattodea, Dermaptera, and Orthoptera. Also found 8 families of soil insects including Scarabaeidae, Carabidae, Formicidae, Ectobiidae, Blattidae, Anisolabididae, Gryllidae, and Gryllotalpidae. From the results of the study, there were 8 genera found, namely Onthophagus, Pheropsophus, Odontomachus, Blattella, Eurycotis, Euborellia, Lepidogryllus, and Gryllotalpa. Insects that act as predators are Pheropsophus, Odontomachus, and Euborellia, insects that act as decomposers are Onthopghagus and Eurycotis. Insects that act as pests are Blattella and Gryllotalpa, and Lepidogryllus.
