Paparan sinar ultraviolet (UV) berperan dalam terjadinya penuaan dini, eritema, dan kerusakan kulit. Buah merah (Pandanus conoideus L.) merupakan tanaman khas Papua yang mengandung senyawa bioaktif berpotensi sebagai antioksidan. Penelitian ini bertujuan untuk mengevaluasi aktivitas antioksidan dan kemampuan perlindungan terhadap sinar UV dari ekstrak etanol biji buah merah. Ekstraksi dilakukan dengan metode maserasi menggunakan pelarut etanol, diikuti skrining fitokimia. Aktivitas antioksidan diuji menggunakan metode DPPH dengan spektrofotometri UV-Vis, dengan vitamin C sebagai kontrol positif, dan dinyatakan sebagai nilai IC50. Penentuan nilai Sun Protection Factor (SPF) dilakukan secara in vitro pada rentang panjang gelombang 290–320 nm. Hasil menunjukkan bahwa ekstrak etanol biji buah merah memiliki aktivitas antioksidan sangat lemah dengan nilai IC50 sebesar 4364,61 ppm, sedangkan vitamin C menunjukkan aktivitas sangat kuat dengan nilai IC50 sebesar 10,66 ppm. Nilai SPF ekstrak pada konsentrasi terendah 200 ppm sebesar 2,63 yang termasuk dalam kategori proteksi minimal, sedangkan pada konsentrasi tertinggi 1000 ppm diperoleh nilai SPF sebesar 6,76 yang termasuk dalam kategori perlindungan ekstra terhadap sinar ultraviolet. ABSTRACT Ultraviolet (UV) exposure contributes to premature aging, erythema, and skin damage. Red fruit (Pandanus conoideus L.) is a plant native to Papua that contains bioactive compounds with potential as antioxidants. This study aimed to evaluate the antioxidant activity and ultraviolet protection capability of the ethanol extract of red fruit seeds. Extraction was carried out by maceration using ethanol as the solvent, followed by phytochemical screening. Antioxidant activity was tested using the DPPH method with UV-Vis spectrophotometry, with vitamin C as the positive control, and expressed as an IC50 value. The Sun Protection Factor (SPF) value was determined in vitro over a wavelength range of 290–320 nm. The results showed that the ethanol extract of red fruit seeds had very weak antioxidant activity with an IC50 value of 4364.61 ppm, whereas vitamin C exhibited very strong activity with an IC50 value of 10.66 ppm. The SPF value of the extract at the lowest concentration of 200 ppm was 2.63, classified as minimal protection, while at the highest concentration of 1000 ppm an SPF value of 6.76 was obtained, classified as extra protection against ultraviolet rays.
