Abstract

As part of the Advanced Indonesia 2045 vision, this study explores the potential of milk-based nutraceuticals enriched with purple sweet potato (Ipomoea batatas var. Ayumurasaki) and rosella (Hibiscus sabdariffa L.) extracts. This research aimed to evaluate the immunomodulatory and antidiabetic activities of three drink formulations (FI, FII, FIII). Male mice were divided into groups (n=5 per group). Immunomodulation was assessed using the carbon clearance method (0.2 ml/20 g BW dosage for 7 days) compared to Stimuno as a positive control. Antidiabetic activity was tested on alloxan-induced hyperglycemic mice (150 mg/kg BW) for 7 days, with glibenclamide as the reference. Data were analyzed using One-Way ANOVA followed by the Duncan test (p < 0.05). Proximate analysis confirmed all formulations met nutritional standards for protein and fat. In the immunomodulatory assay, all formulations significantly increased the phagocytosis constant (K) and reduced the carbon elimination half-life (t1/2) compared to the healthy control (p < 0.05), with FIII showing the highest activity. For the antidiabetic test, FI, FII, and FIII significantly reduced fasting blood glucose (FBG) levels starting from day 3, reaching near-normal levels by day 7, comparable to glibenclamide. The synergistic combination of anthocyanins and vitamin C in these formulations effectively enhances macrophage activity and controls glycemic levels, making them viable nutraceutical candidates for health promotion.
