Piper is widely used as source of essential oils from which many bioactive compounds with medicinal properties were used in traditional medicine. The objectives of this study were to explore the chemical composition of essential oils of eight Piper species and to identify its role in determining taxonomic relationships. Chemical composition of essential oils was analyzed using GC-MS analysis on petroleum ether extracts from stems of eight Piper species, while taxonomic relationship was determined using cluster analysis and principal component analysis. Results showed that 21 terpenoid compounds were identified as constituents of the essential oils, consisted of 4 monoterpenes, 14 sesquiterpenes, and 3 diterpenes. The composition of essential oils varied between species. Cluster analysis and principal component analysis showed that differences in the composition of essential oil compounds determined the grouping of species into two clusters. Five compounds were identified as having major role in the grouping of species. These compounds were ?-selinene, ?-caryophyllene, ?-caryophyllene, farnesyl acetone, and ?-amorphene. The results of this study offer opportunities for the use of Piper stems as source of essential oils. This study also confirms the interspecies variability in composition of essential oils, and at the same time supports the use of essential oils in chemotaxonomic studies.

Abstract. Nugroho LH, Dewatisari WF, Retnaningrum E, Purwestri YA. 2021. The potency of Sansevieria trifasciata and S. cylindrica leaves extracts as an antibacterial against Pseudomonas aeruginosa. Biodiversitas 22: 408-415. Sansevieria trifasciata and Sansevieria cylindrica are the major herbs in Indonesia, which contain several bioactive compounds as potential sources of antibacterial agents. This study aims to evaluate the antibacterial activity of S. trifasciata and S. cylindrica leaves extract and fraction against Pseudomonas aeruginosa and to identify its bioactive compounds. Crude ethanolic extract of S. trifasciata and S. cylindrica leaves were tested for their antimicrobial activity by disk diffusion method against P. aeruginosa. S. trifasciata showed strong antibacterial activity with an inhibition zone of 18.3 mm compared to S. cylindrica. Different concentrations of extract i.e. 4 mg/mL, 8 mg/mL, 16 mg/mL, 32 mg/mL, 64 mg/mL, 128 mg/mL and 256 mg/mL were tested for their minimum inhibitory concentrations (MIC). The MIC results showed that the S. trifasciata extract was able to inhibit bacterial growth at a concentration of 32 mg/mL. Results of vacuum liquid chromatography (VLC) and thin-layer chromatography (TLC) revealed that only fraction 3 showed the highest antibacterial activity at 16 mg/mL. In TLC bioautography analysis, fraction 3 showed a clear zone at Rf 0.93. The phytochemical analysis showed that terpenoid, phenolic, triterpenoid, and flavonoid compounds were found in S. trifasciata extract that were associated with antibacterial activity.

Abstract

Abstract. To’bungan N, Pratiwi R, Widyarini S, Nugroho LH. 2021. Cytotoxicity extract and fraction of knobweed (Hyptis capitata) and its effect on migration and apoptosis of T47D cells. Biodiversitas 23: 572-580. Hyptis capitata Jacq. has an ethnobotanical history. The leaves and stems of H. capitata have been reported to have potential anticancer properties. This study was conducted to determine the anticancer potential of the roots, stems, leaves, and flowers of H. capitata. Organs of H. capitata were extracted in stages using chloroform and continued with methanol to obtain eight extracts. Crude extracts were tested for cytotoxicity on T47D and WiDr cells with the MTT method. Potential inhibition of cell migration by potential fraction was tested with scratch wound healing assay and the content of secondary metabolites was analyzed with gas chromatography-mass spectrometry (GC-MS). An Annexin V-Pl apoptosis test was performed to determine the number of living cells, necrosis, and apoptosis. The results showed that the root chloroform extract showed the highest cytotoxic activity against T47D cells (34.90 ± 4.78µg/mL) and WiDr (44.65 ± 12.07 µg/mL), and it was referred to as a potential extract. Fractionation of the root chloroform extract resulted in four fractions (F1, F2, F3, and F4). Fraction F2 had the highest cytotoxic activity (13.8 ± 0.65 µg/mL) and was selective against T47D cells (selectivity index 3.71). Fraction F2 showed its antimetastatic and apoptotic potential against T47D cells. Gas chromatography-mass spectrometry analyses combined with library showed that the root chloroform extract and fraction F2 contain ferruginol, campesterol, and stigmasterol.
