Nusantara Bioscience 4: 16-21. Carica pubescens Lenne & K. Koch is a species of fruit plant firstly cultivated in South America and has adapted to the highland environment, such as Dieng Plateau, Central Java (±2000 m asl). It has a narrow habitat range and limited or unknown intraspecies variation. Therefore, important information about the characters of the plants at various altitudes is needed, so that it is possible to extend its distribution through transplantation to other areas. Characterization can be performed based on morphological characters, chemical content, and protein banding patterns. This study aimed to describe the morphological characters, the chemical content (antioxidant capacity), and the pattern of protein bands by staining, using Coomassie brilliant blue on C. pubescens in the Dieng Plateau. The research was conducted in the villages of Kejajar (1400 m asl), Patak Banteng (1900 m asl.), and Sembungan (2400 m asl.). The observations of morphological characters were conducted in the field and continued in the laboratory. Morphological characters, the chemical content (antioxidant capacity), and the banding pattern of protein of C. pubescens were analyzed descriptively. The results showed that the morphological characters of C. pubescens in Dieng Plateau varied in stems, leaves, and fruits. The antioxidant capacity decreased with decreasing habitat altitude, 2400 m asl. altitude> 1900 m altitude> 1400 m asl.. The Protein banding patterns did not vary, but the pattern in C. Papaya was different. The uniformity of the pattern of protein bands showed that genetic stability in C. pubescens was not affected by environmental factors.

Abstract. Trimanto, Sajidan, Sugiyarto. 2011. Characterization of taro (Colocasia esculenta) based on morphological and isozymic patterns markers. Nusantara Bioscience: 7-14. The aims of this research were to find out: (i) the variety of Colocasia esculenta based on the morphological characteristics; (ii) the variety of C. esculenta based on the isozymic banding pattern; and (iii) the correlation of genetic distance based on the morphological characteristics and isozymic banding pattern. Survey research conducted in the Karanganyar district, which include high, medium and low altitude. The sample was taken using random purposive sampling technique, including 9 sampling points. The morphological data was elaborated descriptively and then made dendrogram. The data on isozymic banding pattern was analyzed quantitatively based on the presence or absence of bands appeared on the gel, and then made dendrogram. The correlation based on the morphological characteristics and isozymic banding pattern were analyzed based on the product-moment correlation coefficient with goodness of fit criterion. The result showed : (i) in Karanganyar was founded 10 variety of C. esculenta; (ii) morphological characteristics are not affected by altitude; (iii) isozymic banding pattern of peroxides forms 14 banding patterns, esterase forms 11 banding patterns and shikimic dehydrogenase forms 15 banding patterns; (iv) the correlation of morphological data and the isozymic banding pattern of peroxidase has good correlation (0.893542288) while esterase and shikimic dehydrogenase isozymes have very good correlation (0.917557716 and 0.9121985446); (v) isozymic banding pattern of data supports the morphological character data.

Abstract. Muna L, Astirin OP, Sugiyarto. 2010. Teratogenic test of Pandanus conoideus var. yellow fruit extract to development of rat embryo (Rattus norvegicus). Nusantara Bioscience 2: 126-134. This experiment was performed to examine the effect of Pandanus conoideus Lam. var. yellow fruit extract on the percentage of the living fetus, the death intrauterus, heavy and long of fetus, fetus morphology, and skeleton structure of fetus. The experiment was used by 25 pregnant mice that randomly were divided into 5 groups, they contained 5 mice. Each group was given with a different dose. The P1 group (control) was given 1 mL sesame oil, the group P2, P3, P4 and P5 were respectively given the yellow fruit extract 0,02 mL, 0,04 mL, 0,08 mL and 0,16 mL. The P. conoideus var. yellow fruit extract was given orally on day 5 to 17 of gestation (organogenesis period). Observation was carried out on day 18 of gestation by cesarean section to take the fetus from the uterus. Fetus morphology was observed after taking fetus from uterus, whereas observation of skeleton structure was made wholemount preparation with dual colorization, they are Alcian blue and Alizarin Red-S. The result was analyzed with one way ANOVA. Results showed that giving yellow fruit extract didn't influence to the percentage of the living fetus, the death intrauterus, heavy and long of fetus. The effect of giving yellow fruit extract to the maternal was abnormality skeleton (lordosis) of fetus in the dose of 0.16 mL and obstacles to the ossification of fetus.

Abstract. Nurmiyati, Sugiyarto, Sajidan. 2009. Kimpul (Xanthosoma spp.) characterization based on morphological characteristic and isozymic analysis. Nusantara Bioscience 1: 138-145. This research is aimed: (i) to know the variety of kimpul (Xanthosoma spp.) based on morphological characteristics and isozymes analysis; (ii) to know the correlation between its genetic space based on morphological characteristics and its genetic resemblance based on isozymes-banding pattern. These research results were analyzed and described by descriptive qualitative methods. Morphological observation was carried out in sub-District of Galur, Lendah, and Girimulyo, Kulonprogo District, Yogyakarta. Morphological data of the kimpul plant was explored descriptively and then made dendrogram. Data of isozymic banding pattern were analyzed quantitatively based on the appearance of the band on the gel, and qualitatively based on the thickness of the band formed, and then made dendrogram. The correlation, between its genetic distance based on morphological characteristics and its genetic resemblance based on isozymes-banding pattern, were then analyzed grounded on coefficient correlation between product-moment and goodness of it criteria based on correlation. The results pointed out that morphologically, on eight observed samples which were consist of four different types (species), each Xanthosoma from different locations did not indicate obvious differences. Esterase was formed four different banding-patterns, Glutamate Oxaloacetate Transaminase indicated eight different banding-patterns, and Peroxidase indicated seven different banding-patterns. Correlation between morphological data and data from EST and GOT isozymic banding pattern was very good (0.967918 and 0.937113), While, the correlations between morphological data and POD isozymes were good (0.892721). goodness of it criteria based on correlation. The results pointed out that morphologically, on eight observed samples  which were consist of four different types (species), each Xanthosoma from different locations did not indicate obvious differences. Abstract. Maryati KT, Sugiyarto. 2010. Characterization of white grub (Melolonthidae; Coleoptera) in salak plantation based on morphology and protein banding pattern. Nusantara Bioscience 1: 72-77. This research aims to find out the white grub (Melolonthidae; Coleoptera) variability based on the morphological characteristic and protein banding pattern found in "salak pondoh" farm in Regencies of Sleman, Yogyakarta and Magelang, Central Java. Each area has five sampling points. Morphological analysis on white grub was conducted using descriptive method and analysis on protein banding pattern was conducted using qualitative analysis based on the presence or absence of band pattern on the gel, and qualitatively based on the relative mobility value (Rf) of protein. The result indicated that the white grub in Sleman and Magelang, based on morphology characteristic is only one species, namely Holothricia sp. Based on the protein banding pattern, the white grub sample has differences in protein band number and molecular protein weight.

Abstract. Wardani S, Sugiyarto. 2009. Characterization of white grubs (Melolonthidae: Coleoptera) at salak pondoh agroecosystem in Mount Merapi based on isozymic banding patterns. Nusantara Bioscience 1: 38-42. The aim of this research was to know the characteristics of white grubs (Melolonthidae: Coleoptera) based on isozyme banding patterns. This research was conducted at Sleman, Yogyakarta, and Magelang-Central Java for morphological purposes. The sample was taken from 5 places with different heights in which five samples were taken from each location. The method used in this research was polyacrylamide gel electrophoresis (PAGE) using the vertical type. The enzyme system used in this research were peroxidase and esterase to detect the isozyme banding patterns. The results showed a variation in isozyme banding patterns of white grubs (Melolonthidae: Coleoptera) at salak pondoh agroecosystem in Mount Merapi’s slope (peroxidase in station II and IV while esterase in station III and V). It means that genetic variation on the white grubs population at salak pondoh agroecosystem in Mount Merapi’s slope was found. The environmental condition also influenced the isozyme banding pattern’s appearance because each location had a different state.

Abstract

Abstract. Fathonah D, Sugiyarto. 2009. Effect of IAA and GA3 toward the growing and saponin content of purwaceng (Pimpinella alpina). Nusantara Bioscience 1: 17-22. The aims of this research are to examine (i) the effect of IAA and GA3 in different concentrations on the growth of the plants and (ii) the saponin contained inside the P. alpina leaves. The research was done in Sikunang Village, Kejajar Subdistrict, Wonosobo District, Central Java, from July to November 2007. The Completely Random Design used the experimental methods with two factors to analyze this experiment. The first treatment gives IAA and GA3; the second provides different IAA and GA3 concentrations. These experiments were repeated three times. Variables measured in this research were plant growth consisting of the number of leaves, their height, width, wet weight, and dry weight. The chemical compound of the secondary metabolite in the form of leave saponin was employed. Analysis of Variance (ANOVA) analyzed the result, then continued to Duncan Multiple Range Test in 5% level to analyze the real difference between those treatments. The result showed that giving IAA and GA3 differently affects the growth of P. alpina. In variable of the height, the optimal wet weight and dry weight in GA3 treatment was 50 ppm; an optimum number of leaves in GA3 treatment was 50 ppm, whereas the leaves width in IAA treatment was 200 ppm and GA3 treatment was 75 ppm. Optimum saponin treatment was IAA 200 ppm and GA3 25 ppm.
