Abstract

Triploid banana breeding could be achieved by crossing tetraploid to diploid banana cultivars. The availaibility of tetraploidbananas is naturally very limited. One of methods to produce tetraploid bananas is inducing tetraploid plants usingchemicals that inhibit spindle fiber formation during mitosis such as oryzalin or colchicine. The present research is aimed toproduce and characterize tetraploid Pisang Mas Lumut (Musa acuminata, AAAA genome) induced by in-vitro oryzalintreatment from diploid Pisang Mas Lumut. Fifteen in vitro-cultured shoots were treated wih oryzalin at of 60μM for 7days in a liquid MS basal medium with addition of 2 mg/l BA. After 6 sub-cultured, 54 shoots and 104 plantlets wereproduced. Forty seedlings were then survived and grew well and healthy. Ploidy identification of induced Pisang MasLumut was conducted using Flowcytometer at seedling stage and repeated at reproductive phases for 2 cycles. The resultsshowed that oryzalin treatments produced tetraploids at a frequency of 30%, diploid at 65% and mixoploid at 5%. Thetetraploid plants have longer and thicker pseudostems, produce fewer suckers, drooping and larger leaves, and broader fruitscompared to its diploid control at harvest time. The tetraploid plants have similar genetic properties with their diploidcontrols as shown by genetic identity of 0.90-0.97.Key words: Pisang Mas Lumut, Musa acuminata, AA, oryzalin, tetraploid

Abstract

Propagation of Amorphophallus titanum Becc (Araceae) via In Vitro Technology. Amorphophallus titanum is a unique plant with gigantic inflorescence and radiating corpselike odorl and therefore has become a botanical flagship and attract visitors. Its existence innature may be endangered due to habitat conversion that reduce population size required for cross pollination to occur. Propagation via non-conventional technique is much needed for the resulting planting material to be exchanged among botanical garden or even traders. Mass propagation using in vitro technology has been successful. An improvement of the propagation method has been developed by incorporating high level of cyitokinin BA of 1-2 mg/l in the medium for shoot proliferation and lowering it to 0-0,1 mg/l for shoot elongation. Shoot elongation is effectively influenced by age and the phase of medium whereas the liquid phase induced the enlargement of the shoots significantly without even promoting any hyperhidricity. The plantlets produced survive acclimatization.Key words: Amorphophallus titanum, mass propagation, in vitro, mediurm cair, BA.

Abstract

ABSTRACTEarly detection for Fusarium wilt resistant of banana plants can be done at seedling phase under greenhouse condition. The purpose of early detection was to screen and evaluate some number of local and hybrid accession of banana plants for resistance to Fusarium oxysporum f. sp. cubense (Foc VCG 01213/16 TR4 or Foc TR4). The treatments were done by immersing the root seedling in Fusarium inoculant at concentration of 106 conidia mL-1, for 2 hours, then planted in sterile sand media and placed in container with "double tray system". Experiments were performed using 15 accessions of banana i.e. 1 accession of wild musa (Musa acuminata var. malaccesis), 7 local accessions and 7 hybrid bananas. Based on greenhouse bioassay, banana Rejang#2 (AA) showed resistance to Foc TR4. The results were consistent in the field condition. Tetraploid plants for pisang Rejang#2 (AAAA) also showed resistant to Foc TR4. Cultivar Cavendish (AAA) and Madu (AA) showed highly susceptible; Ustrali (AAAB) and Goroho (AAA) showed susceptible to Foc TR4. Wild banana (Musa acuminata var. malaccensis) and Mas Jambe 4x (AAAA) showed moderate susceptible. Of the total 7 hybrids accession; 3 hybrids showed moderate susceptible, and 4 other hybrids showed susceptible to Foc TR4.Keywords: banana plants, early detection, Foc TR4, hybrid, local ABSTRAK Deteksi awal tingkat ketahananan tanaman pisang terhadap penyakit layu Fusarium oxysporum f. sp. cubense (Foc) dapat dilakukan pada masa pembibitan di rumah kaca.
