Abstract

Microbial resistance to antibiotics in skin infections has created a need for new sources of antibacterial compounds. Physalis angulata L. leaves contain various bioactive compounds, making them potential source of endophytic bacteria capable of producing antimicrobial substances. This study aimed to isolate, characterize, and identify the bioactive compounds produced by endophytic bacteria from ciplukan leaves and evaluate their antimicrobial activity. Methods included direct plating for isolation, phenotypic characterization, and antimicrobial testing of single isolates and combinations of endophytic bacteria against Staphylococcus aureus, Cutibacterium acnes, Pseudomonas aeruginosa, Escherichia coli, dan Candida albicans. Bioactive compounds were identified using Gas Chromatography–Mass Spectrometry (GC-MS). Six endophytic bacterial isolates were obtained. The bacterial consortium supernatant produced larger inhibition zones than single isolates, showing the highest activity against C. acnes (13.9 mm), S. aureus (13.6 mm), E. coli (10.6 mm), and P. aeruginosa (9.8 mm). GC-MS analysis identified Bis(2-ethylhexyl) phthalate (39.33%) and 1,4-Benzenedicarboxylic acid, bis(2-ethylhexyl) ester (11.25%) as the dominant compounds likely responsible for the antimicrobial activity. These results indicate that endophytic bacteria from ciplukan leaves have potential as natural antibacterial agents. This study contributes scientific knowledge that can serve as a basis for developing natural antibacterial compounds derived from ciplukan leaf endophytic bacteria.
