Abstract

Komunitas perifiton sungai telah merespons kondisi lingkungan. Mereka berguna dalam menggambarkan keadaan ekologis, kinerja, dan keberlanjutan ekosistem karena kemampuan untuk mengukur berbagai parameter lingkungan dan menghubungkannya dengan keanekaragaman, kemerataan, dan kekayaan. Studi ini menyelidiki keanekaragaman perifiton Sungai Bango di Malang dan karenanya menghasilkan daftar periksa kekayaan perifiton sungai, terutama perifiton epilitik. Pengambilan sampel perifiton dan identifikasi dilakukan di Laboratorium Hidrobiologi Fakultas Perikanan dan Ilmu Kelautan Universitas Brawijaya Malang. Penelitian ini dilakukan seminggu sekali dalam sebulan (Mei 2020) antara jam 9 dan 13 (GMT + 7). Identifikasi dan sampel Perifiton dikumpulkan dengan cara mengikis substrat (batuan/batuan) di dalam dan di sekitar titik pengambilan sampel air masing-masing. Hasil penelitian menunjukkan kelimpahan total perifiton berkisar antara 938.900 Ind/mm2 sampai dengan 1.598.125 Ind/mm2 dengan 30 genera yang ditemukan. Kami berasumsi bahwa titik sampling 1 sampai 3 memiliki indeks keanekaragaman sedang; Sebaran jumlah individu tiap spesies tergolong sedang; stabilitas komunitas sedang dan organisme akuatik dapat mentolerir kondisi lingkungan. Namun, titik pengambilan sampel 4 memiliki keragaman yang rendah; sebaran jumlah masing-masing spesies rendah; stabilitas komunitasnya rendah dan hanya dapat ditoleransi oleh organisme akuatik tertentu.

Abstract

The development of the fisheries sector has a significant contribution in improving the economy, especially in Indonesia, which has great potential but has not been utilized optimally. In Tawangargo Village, Malang Regency, training and assistance was carried out for alternative feed formulations for Dumbo catfish (Clarias sp.). This activity aims to empower the community by increasing skills and knowledge in cultivating freshwater fish. The methods used include a community approach, outreach through seminars and direct practice, as well as assistance in alternative feed formulation. The activity stages include initial surveys, outreach and training, which were responded positively by the local community and youth. The cultivation medium is made from a mixture of chicken and cow manure, EM4, molasses and fermented water, then dried in the sun and sprayed with EM4 again before use. Fermented feed is made from cow dung, tofu dregs, corn and rice bran, pellets, EM4, molasses, tape yeast, vitamin C, and water which is fermented for 7 days. The results of the activity show that catfish farming business actors in Tawangargo Village can formulate alternative feed with low costs, easy methods, and efficient results, thereby reducing losses. This training also increases the community's knowledge and ability to make maximum use of land for cultivating freshwater fish, supporting the sustainability of the local economy

Abstract

Pesticides are widely applied in the agriculture sector to protect crops and pest control. The poisonous substance of pesticides will affect all of the organisms, either target and non-target organisms. Fish can play the role of an indicator of genotoxic presence in aquatic environments. Polysaccharide extracts from sargassum have promising anti-genotoxic potential. This study aimed to analyze the anti-genotoxic activity of brown seaweed (Sargassum polycystum) methanol extract against erythrocyte and micronuclei of tilapia exposed by methomyl-base pesticide. Brown seaweed (Sargassum sp.) purchased from farmers in Sumenep Regency, Madura, East Java, then macerated using methanol 1: 3 (w / v) for 3x24 hours at room temperature. The phytochemical screening was including flavonoids, alkaloids, triterpenes, saponins, and tannins. Tilapia fish (TL ± 9-12cm) purchased from the Technical Application Unit of Freshwater Fish (UPT Perikanan Air Tawar), Sumberpasir, Malang, East Java. The result of this study showed that exposure of methomyl-based pesticides in the concentration of 4.015 ppm indicates the formation of micronuclei of 318.33 ‰. The increased concentration of extract treatment is directly proportional to the decrease of micronuclei. It means that sargassum extract can reduce the genotoxic effect on exposed tilapia by methomyl-based pesticides. The best concentration of Sargassum sp. extract that can reduce genotoxic was D (200 ppm). Keywords: Antigenotoxic, Extract, Methomyl, Pesticides, Sargassum sp., Tilapia.

Abstract

This research investigates the phytochemical composition, antioxidant activity, and antibacterial efficacy of Ipomoea pes-caprae extracts collected from Pantai Talang Siring Pamekasan, Madura. Phytochemical analysis revealed the presence of alkaloids, flavonoids, phenolic compounds, terpenoids, tannins, and saponins in the extracts, indicating their rich bioactive profile. Antioxidant activity assessed via the DPPH assay demonstrated concentration-dependent scavenging of free radicals, with methanol and ethanol extracts exhibiting superior activity compared to the aqueous extract. Antibacterial assays against Escherichia coli and Staphylococcus aureus revealed significant inhibitory effects, with methanol and ethanol extracts displaying greater potency than the aqueous extract. These findings underscore the potential of Ipomoea pes-caprae as a source of natural antioxidants and antimicrobial agents, warranting further investigation into its pharmacological applications and therapeutic potential.

Abstract

The study aimed to determine the effect of using a mixture of kappa-carrageenan/Chitosan and Iota carrageenan/Chitosan as encapsulating materials for Lactobacillus acidophilus and Bifidobacterium bifidum on viability, encapsulation yield, moisture content and aw of microcapsules. The research method is an experimental laboratory design, ANOVA, LSD using the Excel program. The results showed that using chitosan with a DD of 83.5% in the i-carrageenan/Chitosan mixture gave the highest viability of Bifidobacterium bifidum. The combination of i-carrageenan/chitosan coating ingredients with a concentration ratio of 0.5%:1.5% produced probiotic viability (L aci + B bifi) of 6.56 log CFU/g, while probiotic viability (B bifi) was the highest with a viability 6.70 log CFU/g. The highest probiotic encapsulation yield (L aci+B bifi) was 72.85%. The water content of the microcapsules (L aci+B bifi) was 8.96%, and the aw value was 0.63. It is recommended to carry out further testing using a processing factor for its viability.

Abstract

Budidaya ikan air tawar saat ini semakin maju ditunjang meningkatnya sarana prasarana memadai berupa peralatan perikanan, makanan obat-obatan, teknik budidaya yang disempurnakan maupun penaggulangan dan pencegahan hama dan penyakit ikan. Mitra Kegiatan Hibah Pengandian Masyarakat ini adalah Kelompok Tani di Desa Wonoasri, Kecamatan Grogol, Kabupaten Kediri, Jawa Timur. Desa Wonoasri adalah suatu desa yang secara geografis terletak di Kecamatan Grogol Kabupaten Kediri. Desa tersebut memiliki jumlah penduduk 1.692 jiwa dengan luas wilayah 1,03 km2 sedangkan kepadatan penduduk mencapai 1.479 jiwa/km2. Berdasarkan hasil survey, terdapat permasalahan mengenai (1) Beberapa kolam terdapat ikan lele yang mengalami stress dan kematian, bahkan hampir setiap hari selalu ada yang mati, salah satunya saat pergantian air dimulai. (2) Banyak gejala penyakit yang muncul, seperti luka, kulit mengelupas, jamur. (3) Tidak adanya ketentuan pengurasan kolam air ikan. (4) Terdapat ikan lele yang berwarna merah kecoklatan. (5) Ukuran tubuh ikan lele yang tidak sama per kolam, bahkan yang seharusnya bobot ikan besar dan waktunya siap untuk dipanen, tetapi ikan lele kebanyakan cenderung berukuran kecil dibandingkan kolam yang lainnya. Solusi untuk mengatasi berbagai masalah di Desa Wonoasri meliputi memberikan edukasi tentang pemanfaatan lahan, penyuluhan mengenai bibit ikan air tawar yang cocok, pelatihan budidaya ikan seperti lele, gurami, mas, nila, dan mujair, serta pelatihan dan pendampingan dalam mencegah dan menanggulangi Hama Penyakit Ikan (HPI). Dengan penerapan solusi ini, diharapkan masyarakat dapat memaksimalkan potensi lahan, mengelola kolam ikan dengan efektif, mengembangkan usaha budidaya ikan secara mandiri, dan mengurangi kerugian akibat HPI. Metode yang digunakan adalah: ceramah, demonstrasi, diskusi, tanya jawab, dan mitra pendamping. Hasil yang dicapai adalah (1) mitra memiliki pengetahuan tentang pengelolaan budidaya ikan air tawar, (2) mitra memiliki keterampilan mengolah sumber daya alam yang ada di sekitar, (3) pemanfaatan lahan kosong.

Abstract

This study aims to evaluate the nutritional value and heavy metal lead (Pb) content in suckermouth catfish (Pterygoplichthys sp.) egg meal as a candidate feed additive in fish feed. The fish eggs were obtained from fishermen's catches in public waters in East Java, then cleaned, dried, and processed into meal for analysis. The proximate parameters tested included protein, fat, crude fiber, and ash content using the AOAC standard method, while Pb content was analyzed using Atomic Absorption Spectrophotometry (AAS). The results showed that suckermouth catfish egg meal had the following nutritional composition: protein 39.25±0.02%, fat 9.19±0.01%, crude fiber 2.36±0.03%, and ash 4.95±0.02%. This nutritional profile indicates that suckermouth catfish eggs are a high source of animal protein and have a low fiber content, thus having the potential to improve feed quality through the addition of essential amino acids and energy. Heavy metal Pb content was not detected within the sensitivity limits of the instrument, thus this product meets the safety aspects of feed ingredients related to heavy metal contamination. Overall, the results of this study indicate that suckermouth catfish egg meal has competitive nutritional value compared to several conventional feed ingredients and is safe from Pb contamination. Thus, this ingredient has the potential to be developed as an alternative feed additive in fish feed formulations.

Abstract

Pesticides are recognized as serious pollutants in the aquatic environment with the potential to cause genotoxic on the aquatic organism, especially fish. The micronucleus (MN) assay has been used to evaluate genotoxicity of many compounds in polluted ecosystems such pesticides. The aim of this study to determine genotoxic effect of methomyl-based pesticide on tilapia (Oreochromis niloticus). Fish were exposed to six different concentrations base on range finding test (0 ppm, 3.2 ppm, 4.2 ppm, 6.5 ppm, 8.7 ppm and 10 ppm) of methomyl-based pesticide. The micronucleus were collected from peripheral blood erythrocyte of fish after 96 h exposure. Peripheral blood samples smears were stained with Giemsa, MN frequencies were counted and statistically analyzed using one-way ANOVA. The result of this study showed after 96 hours exposed to methomyl-based pesticide, at concentration 0 ppm causes 0% mortality, at concentration to 3.2 ppm causes 30% mortality, at concentration 4.2 ppm causes 60% mortality, at concentration 6.5 ppm causes 70% mortality, at concentration 8.7 ppm causes 80% mortality, at concentration 10 ppm causes 100% mortality of fish test. Lethal Concentration 50 (LC50 - 96 hours) of methomy-base pesticide towards tilapia (O. niloticus) is 4.015 ppm. Through micronuclei assay during 96 hour exposure of methomyl-based pesticide, the result shows that frequencies of micronuclei in erythrocyte of fish test at concentration at 0ppm is 12‰, 18‰ and 16‰; at concentration at 3.2ppm is 33‰, 26‰ and 29‰; at concentration at 4.2ppm is 41‰, 38‰ and 46‰; at concentration at 6.5ppm is 68‰, 81‰ and 82‰; at concentration 8.7ppm is 133‰, 130‰ and 137‰; at concentration 10ppm is 163‰, 166‰ and 156‰. It revealed that methomyl-based pesticide exposure induced after 96 h significantly (P<0.05) increased genotoxic potentials simultaneous with increased concentration.Keywords: Genotoxic, Methomyl, Micronucleus Assay, Pesticide, Tilapia.

Abstract

Intensification of tilapia (Oreochromis niloticus) farming around agricultural land increases the risk of contamination by the toxic and genotoxic pesticide methomyl, which can trigger oxidative stress, and immune system disorders. The use of natural materials with antioxidant potential, such as kremah (Alternanthera sp.) leaf extract, is an alternative to mitigate these effects. This study was conducted to evaluate the effect of immersion in kremah leaf extract on the leukocyte differential of tilapia exposed to methomyl. A completely randomized design was applied, consisting of four treatments with three replications, namely control, and doses of 20, 30, and 40 ppm. The fish were exposed to 3.2 ppm methomyl for 96 hours and then immersed in the extract for 14 days. The observed parameters comprised leukocyte differential, clinical signs, and water quality, all of which were analyzed using one-way ANOVA followed by an LSD test (p < 0.05). The findings indicated that methomyl exposure led to an increase in monocytes while reducing lymphocyte levels. Immersed in 40 ppm extract for 14 days provided the best results by decreasing monocytes (4.00%), as well as increasing lymphocytes (82.00%) and hemoglobin levels to near normal. It was concluded that kremah leaf extract effectively mitigates oxidative stress and genetic damage caused by methomyl, with an optimal dose of 40 ppm for 14 days.
