Abstract

Fertility is the success of spermatozoa in fertilizing an egg cell, while hatchability is a number that shows an egg's high or low ability to hatch. This research aims to determine the fertility and hatchability of KUB chicken eggs that have been stored cold by adding carrot juice to the basic diluent Ringer Lactate Egg Yolk. There were 60 KUB chickens divided into two treatment groups, each comprising 30 KUB hens. The group of eggs resulting from artificial insemination used fresh semen with Ringer Lactate diluent (P0), and eggs resulting from artificial insemination used liquid semen, which had been stored for 2 days with carrot juice supplemented with RLKT diluent (P1). The parameters observed were fertility and egg hatchability. Data were analyzed using descriptive analysis. The results showed that spermatozoa fertility at P0 was 87.11% while P1 was 56.09%. While the average percentage of hatchability shows a PQ result of 75.69 while P2 is 93.91%, it can be concluded that the fertility of KUB chickens using fresh semen is better than using liquid semen, but the hatchability of KUB chicken eggs using liquid semen is better than that of fresh semen. Further research is needed on the success of using liquid semen in AI applications in KUB chickens.

Abstract

ABSTRAK Penelitian ini bertujuan untuk mengetahui apakah Aeromonas hydrophila dapat dideteksi dengan menggunakan teknik imunohistokimia. Imunohistokimia adalah suatu proses untuk mendeteksi antigen (protein, karbohidrat, dll.) Terhadap sel jaringan dengan prinsip reaksi antibodi yang berikatan dengan antigen di dalam jaringan. Parameter penelitian ini adalah interaksi antigen dan antibodi yang divisualisasikan dengan munculnya warna coklat pada ginjal yang diinfeksi Aeromonas hydrophila. Mencit (Mus musculus) umur tiga bulan seberat 30 gram diinfeksi Aeromonas hydrophila dengan dosis 106 CFU / ml. Setelah seminggu, ginjal disiapkan untuk preparat histopatologi dengan pewarnaan imunohistokimia. Antibodi primer yang digunakan adalah antibodi kelinci (New zealand white) dengan dua perlakuan (P1 dan P2). P1 diinfeksi dengan menggunakan protein spesifik Aeromonas hydrophila 30 kDa dan P2 diinfeksi dengan menggunakan protein utuh Aeromonas hydrophila. Hasilnya ditunjukkan dengan membandingkan P1 dan P2. Hasil penelitian menunjukkan bahwa terdapat perbedaan warna coklat yang lebih banyak melalui P2 dibandingkan dengan P1. Kata kunci—Aeromonas hydrophila, Mencit (Mus musculus), Antigen, Teknik Imunohistokimia. ABSTRACT This research was aimed to determine whether the Aeromonas hydrophila can be detected by using immunohistochemistry technique. Immunohistochemistry was a process to detect antigens (proteins, carbohydrates, etc.) towards the cells of the tissues with antibodies reaction principle that binded to antigen in the tissues. The parameter of this research was an interaction between antigens and antibodies which were visualized by the appearance of brown color in the kidneys which were infected by Aeromonas hydrophila. The three-month old mice (Mus musculus) with 30 grams of weight were infected by Aeromonas hydrophila with 106 CFU/ml of doses. After a week, the kidneys were prepared for histopathology preparation by using immunohistochemistry staining. The primary antibodies used were rabbits’ antibodies (New zealand white) with two treatments (P1 and P2). P1 was infected by using spesific proteins of Aeromonas hydrophila 30 kDa and P2 was infected by using the whole proteins of Aeromonas hydrophila. The results were showed by comparing P1 and P2. The results showed that there was a difference in the brown color which was more through in P2 compared to P1. Abstract

Sektor peternakan berpotensi dalam pemenuhan kebutuhan protein masyarakat, khususnya ternak sapi potong sebagai penghasil utama daging. Akan tetapi, rendahnya produksi hijauan pakan ternak seringkali menjadi permasalahan terutama saat musim kemarau. Maka, pengembangan usaha dengan penggunaan teknologi dalam pakan ternak, sangat penting untuk mendukung aspek efisiensi usaha. Upaya perkembangbiakkan sapi potong salah satunya dilakukan oleh Gapoktan Dharma Tani di Kecamatan Srengat, Blitar. Penelitian ini bertujuan untuk menganalisis faktor yang berpengaruh dalam tingkat adopsi pengawetan pakan sapi potong. Penelitian ini merupakan penelitian kuantitatif dengan jenis penelitian deskriptif, pengujian instrument menggunakan expert judgement. Teknik pengumpulan data dikakukan dengan survei dan wawancara. Pengambilan sampel penelitian ini menggunakan teknik purposive sampling, dengan sampel sejumlah 30 anggota GAPOKTAN Dharma Tani. Penelitian ini dilakukan di Kecamatan Srengat Kabupaten Blitar. Penelitian dimulai dengan mengetahui karakteristik dan kondisi adopsi peternak, yang kemudian digunakan sebagai sasaran penyuluhan untuk mengetahui tingkat adopsi. Hasil penelitian diketahui bahwa peternak berada pada tingkat “Sangat Kurang Adopsi” sebelum dilakukan penyuluhan. Rancangan penyuluhan dibuat sebagai skema kegiatan dalam pengawetan pakan dari limbah tongkol jagung dan indigofera zollingeriana. Tingkat adopsi setelah dilakukan penyuluhan berada pada Cukup Adopsi. Indikator adopsi dengan peningkatan tertinggi yaitu Tahap Mencoba. Pentingnya teknologi pengawetan dinilai efektif agar peternak dapat menyimpan pakan lebih lama, sehingga produktivitas peternakan lebih optimal.

Abstract

This study aimed to determine the quality of chicken semen from Kampung Unggulan Balitbangtan (KUB) in Ringer lactate-egg yolk diluent with carrot extract supplementation during cold storage. A total of 6 KUB males were selected for the experiment and each of the was subjected to 5 collections of semen. The experiment was performed using a Randomized Block Design (RBD) with 4 treatments. These treatments included P0 (100% Ringer lacate-egg yolk without carrot extract), P1 (80% Ringer lacate-egg yolk + 20% carrot extract), P2 (60% Ringer lacate-egg yolk + 40% carrot extract), P3 (Ringer lactate yolk 40% + 60% carrot extract). The research variables consisted of motility, viability, abnormalities of spermatozoa, and plasma membrane integrity. Data were analyzed using Analysis of Variance (ANOVA) followed by Duncan's test. It was discovered that the percentage of motility and viability up to 48 hours of storage showed significant differences (p<0.05) between treatments. The best results were in group P2, with motility and viability being 40.33 ± 6.60% and 60.86 ± 7.84%, respectively. Abnormalities of spermatozoa up to 24 hours of storage showed significant differences (p<0.05) between treatments, and not significantly different (p>0.05) at 48 hours of storage. Furthermore, plasma membrane integrity failed to show a significant difference between groups at 1 hour of storage and was significantly different (p<0.05) at 24 and 48 hours of storage. The best or the highest result is in the P2 group, with plasma membrane integrity being 71.89 ± 2.40%. It was concluded that the use of 40% carrot extract supplementation in Ringer lacate-egg yolk diluent tends to maintain the quality of KUB chicken semen in cold storage for up to 48 hours and was suitable for the application of artificial insemination
