Abstract

This study aims to see the genetic diversity of the musk mango (Mangifera casturi) plant in South Kalimantan based on DNA barcode techniques using sequences from the rbcL gene and the matK gene. The plant samples used in this study were the leaves of the musk mango (Mangifera casturi) taken from eight regencies and cities in South Kalimantan Province, namely from Amuntai, Astambul, Barito Kuala, Peat, Kandanga, Babirik, Tabalong, and Tanah Laut. Isolation and DNA analysis of the musk mango plant was carried out in the research laboratory of the Faculty of Biotechnology, Atma Jaya Catholic University of Indonesia, Jakarta. The analysis of the genetic diversity of the musk mango began with the stage of amplifying genomic DNA (gDNA) using rbcL and matK primers. The results of the DNA isolation samples showed a high concentration level of 17-91 ng/µL with a purity of 1.72 -1.92 ng/µL. When PCR was carried out using rbcL and matK primers, from 8 areas in South Kalimantan, 5 samples (Babirik, Tabalong, Kandangan, Amuntai, Batola, Peat, and Tanah Laut) were amplified with rbcL primers and 6 samples (Tanah Laut, Babirik, Tabalong , Amuntai, Peat, and Kandangan) amplified with matK primer. The relationship between the musk mango in South Kalimantan can be seen from the phylogenetic tree.

Abstract

Abstrak Inhibitor tirosinase digunakan dalam kosmetik sebagai pencegah terjadinya hiperpigmentasi. Penelitian ini bertujuan untuk uji aktivitas inhibisi terhadap enzim tirosinase dari ekstrak etanol daun pepaya (Carica papaya L.) secara in vitro. Pemilihan inhibitor dari bahan alam memiliki tingkan keamanan yang lebih aman daripada sintesis. Ekstrak etanol daun pepaya diperkirakan memiliki aktivitas inhibitor tirosinase karena mengandung senyawa flavonoid. Daun pepaya diekstraksi secara maserasi dengan etanol 70%, hingga diperoleh ekstrak kental. Pengujian dilakukan dengan L-DOPA sebagai substrat, ekstrak etanol daun pepaya dengan konsentrasi 20000, 10000, 5000, 2500, 1250, 625 dan 312.5 µg/ml dengan kontrol positif asam kojat lalu diukur serapannya dengan menggunakan microplate reader pada panjang gelombang 490 nm. Hasil penelitian menunjukkan bahwa ekstrak etanol daun pepaya (Carica papaya L.) memiliki aktivitas inhibitor tirosinase dengan nilai IC50 sebesar 12158,8751 µg/ml dan potensi relatif sebesar 6,879 x 10-3 kali asam koja Abstract Tyrosinase inhibitors used in cosmetics as a deterrent occurrence of hyperpigmentation. This study aimed to test the activity of the enzyme tyrosinase inhibition of the ethanol extract of Carica papaya (Carica papaya L.) in vitro. Election inhibitors from natural materials have security safer than synthetic. The ethanol extract of papaya leaves is estimated to have activity of tyrosinase inhibitor compounds that contain flavonoids. Papaya leaves extracted by maceration in 70% ethanol, to obtain a thick extract. Testing is done with L-DOPA as a substrate, the ethanol extract of papaya leaf with a concentration of 20000, 10000, 5000, 2500, 1250, 625 and 312.5 ug / ml with the positive control kojic acid and absorbance was measured using a microplate reader at a wavelength of 490 nm. The results showed that the ethanol extract of papaya leaves leaves of papaya (Carica papaya L.) have activity of tyrosinase inhibitor with IC50 value of 12158.8751 ug / ml and the relative potency of 6.879 x 10-3 times kojic acid.

Abstract

ARI is still a major health problem in Indonesia. The purpose of health counseling in RW 01 Kalibaru, Cilincing North Jakarta is to increase public awareness about the dangers of ARI and the importance of Clean and Healthy Living Behavior (PHBS). The methods used include interactive lectures, questions and answers, and demonstrations of making hand sanitizers. The activity was attended by 36 participants, the majority of whom were female (97.1%) with an age range of 21-65 years. To measure participants' understanding, an evaluation was conducted after the material was given. The results showed that 97.1% of participants had good knowledge about ARI and PHBS after the counseling was given. Participants were able to correctly answer questions related to the definition of ARI, symptoms, prevention, and principles of PHBS. In conclusion, this counseling is effective in increasing community knowledge about ARI and PHBS. It is expected that this better knowledge can encourage behavioral changes in the community.

Abstract

Padina australis is a species of brown macroalgae with potential as a natural source of bioactive compounds, including antifungal agents. This study aimed to identify the functional groups present in the methanolic extract of Padina australis using Fourier Transform Infrared (FTIR) spectroscopy and to evaluate its antifungal activity against Malassezia globosa through the disk diffusion method. FTIR analysis revealed the presence of hydroxyl (3433.41 cm⁻¹), aliphatic C–H (2916.47 cm⁻¹), carbonyl (1735.99 and 1643.41 cm⁻¹), amine (1573.97 and 1242.20 cm⁻¹), C–O, and carboxylate (COO⁻) groups, indicating key secondary metabolites such as flavonoids, phenols, esters, amines, and polysaccharides. Antifungal testing showed that the extract at a concentration of 100 ppm produced the largest inhibition zone (16.58 ± 4.71 mm), while the minimum inhibitory concentration (MIC) was determined at 12.5 ppm with an inhibition zone diameter ≥12 mm. These findings suggest that Padina australis holds promising potential as a natural source for developing antifungal therapies against Malassezia globosa.
