Abstract

Aktivitas kelompok wanita tani Desa Dawuhan Kulon Kecamatan Kedungbanteng Kabupaten Banyumas, selama ini adalah bercocok tanam padi, tanaman sayuran dan tanaman hias. Mulai tahun 2020 desa tersebut sedang menggalakkan program budidaya ikan, Kepala Desa menghendaki agar kelompok wanita tani terlibat dalam kegiatan itu. Tujuan pelatihan dan pendampingan ini adalah memberikan pengetahuan dan keterampilan kepada kelompok tersebut dalam pembenihan ikan Nila dan Nilem. Metode yang digunakan partisipatif yang direalisasikan dalam bentuk pelatihan, dengan tahapan  kegiatan: 1) persiapan, 2) pelaksanaan kegiatan (knowledge transfer, praktik, pembuatan demplot), 3) monitoring dan evaluasi, 4) pendampingan.  Hasil yang diperoleh adalah anggota kelompok wanita tani berhasil memproduksi benih ikan Nila dengan cara pemijahan alami dan memproduksi benih ikan Nilem melalui pijah induksi. Keberhasilan dalam memijahkan tersebut mengindikasikan bahwa anggota kelompok wanita tani di Desa Dawuhan Kulon  telah bertambah pengetahuan dan keterampilannya dalam melakukan pembenihan ikan air tawar.

Abstract

Macrobenthos is one of the biotic water constituent and can be used as bioindicator in determining water quality conditions. Water quality can be seen in terms of community structure of macrobenthos. The present study aimed to determine the physical and chemical water qualities in Penjalin Reservoir and to know the community structure of macrobenthos contained in the waters of Penjalin Reservoir. Community structure includes Density (organism/m2), Relative Density, Dominance Index (C) and Diversity Index (H ') as well as to determine the relationship between the water quality (physical and chemical) and the macrobenthos density. The physical and chemical qualities of Penjalin Reservoir water was good based on class III water quality standard according to PP No. 82 Ttahun 2001. Based on observation, 11 macrobenthos were obtained. Diversity Index (H') based on the criteria of Lee  et al. (1978) showed moderate values (2.07 to 1.66) suggesting a low contamination level. The relationship of macrobenthos density and its chemical and physical quality of water pH, temperature, light penetration, depth, DO, BOD was weak, but had strong relationship with TSS. The results showed a linear regression equation Y = 898.76 X - 236.946 (Y = density and X = TSS) which could be interpreted that the more value of macrobenthos density the higher value of TSS contained in the water.

Abstract

Geosesarma shows intraspecific carapace color variation, which might lead to species misidentification. The problem can be solved using DNA barcoding. There is one research about Geoserarma from the southern slopes of Mount Slamet, but samples were only collected from the Banjaran River for morphological identification. Here, we collected samples from wider areas covering south slope and applied molecular identification. This research aims to assess Geosesarma diversity in south-slope Mount Slamet Central Java, Indonesia based on the cytochrome c oxidase 1 gene barcoding. Surveys were carried out at six sites. Taxonomic identification was done using the barcoding technique. Four morphotypes were obtained during the research. Three morphotypes with the square carapace were identified as Geosesarma, while the remaining one morphotype was included in Parathelphusa. The three Geosesarma morphotypes were barcoded as Geosersarma dennerle because their genetic identity was more than 97% of the G. dennerle sequence in Boldsystems. In contrast, the Parathelphusa morphotype was barcoded as P. convexa with a genetic identity of 97.50%. It can be concluded that the Geosesarma crab on the south-slope Mount Slamet only consists of one species but has carapace and claw color variations. The data are essential for Geosesarma market development and conservation in the region.

Abstract

River Mengaji originates from a water source on the southern forest slopes of Mount Slamet. Gastropods are important biotic components in river ecosystems and can be used as bioindicators due to their sensitivity to water quality changes. Gastropod abundance is closely related to the physical and chemical parameters of water quality. This study aimed to determine the abundance of gastropods, water quality, and the relationship between gastropod abundance and water quality in River Mengaji, Banyumas Regency. Species gastropod identified samples collected. The relationship between gastropod abundance and water quality was analyzed using Spearman's Rank Correlation. The result showed that the gastropods found in Sungai Mengaji consist of five species, namely Melanoides tuberculata, Sulcosprira testudinaria, Radix rubiginosa, Tarebia granifera, and Filopaludina javanica. The highest abundance of gastropods was observed at Station IV, followed by Stations II, III, and I. Water quality conditions were generally good enough, except for temperature and BOD at Station IV. The high abundance of F. javanica at Station IV is related to the high temperature and BOD conditions in Sungai Mengaji, Banyumas Regency.

Abstract

This study addresses taxonomic uncertainty surrounding color variants of Procambarus clarkii in aquaculture and conservation contexts. We investigated whether commercially significant color morphs represent distinct subspecies or phenotypic variations of a single species. Using the cytochrome c oxidase subunit I (COI) gene as a molecular marker, we analyzed four color morphs (blue, white, orange, and brown) from aquaculture facilities in Banyumas Regency. DNA extraction, PCR amplification, and sequencing were performed on 20 specimens. Results showed high sequence homology (98.7-99.8%) across all variants, confirming they belong to a single species. Genetic distance analysis revealed minimal divergence (0.2-1.3%), insufficient for subspecies classification. Phylogenetic reconstruction demonstrated specimens clustered by genetic similarity rather than color or geographic origin, indicating coloration results from genetic mutations rather than environmental adaptations. This COI-based approach provides a molecular framework for taxonomic classification of P. clarkii varieties, with implications for breeding programs, variety certification, and management of this economically important yet potentially invasive species.

Abstract

Udang genus Penaeus termasuk udang laut dari familia Penaeidae. Ciri yang membedakan udang Penaeus dengan spesies udang Penaeidae lainnya adalah bagian atas dan bawah rostrum terdapat gigi. Salah satu komoditas udang Penaeid yang bernilai ekonomis penting adalah udang putih. Udang putih yang dikenal di pasaran terdiri dari 2 spesies yaitu P. indikus dan P. merguensis, yang secara morfologi sangat mirip. Teknik Truss morfometrics diharapkan dapat memudahkan pengenalan ke 2 spesies tersebut. Tujuan dari penelitian ini adalah untuk mendapatkan rasio jarak truss dengan panjang standar yang dapat digunakan untuk membedakan spesies udang P. indikus dan P. merguensis. Metode yang digunakan dalam penelitian ini adalah survei di Pelabuhan Perikanan Samudera Cilacap, dan pengambilan sampel secara Purposive Random Sampling. Sampel diambil tiga kali dengan interval sepuluh hari. Variabel yang diamati adalah karakter morfologi dan morfometrik. Karakter morfometrik terdiri dari 23 jarak trus. Data morfometrik dianalisis “compmenggunakan SPSS “independent-Samples T Test”. Hasil penelitian diperoleh 11 karakter morfologi yang dapat digunakan sebagai pembeda P. indicus dan P. merguiensis namun yang paling mudah digunakan sebagai penentu spesies adalah rasio panjang carapace dgn panjang standar yang memunjukan P. indicus lebuh besar dibanding P. merguiensis.
