Abstract

The aim of this study was to evaluate the effect of superovulation treatment using combination of CIDR, FSH and hCG in beef cattle as donor embryos using MOET programme. All animals had been palpated to evaluated the ovary status and normal cows were used as donor and synchronized with CIDR (Eazy BreedTM). At day 10 of oestrus cycle, cows were divided into two groups, first group: cows were injected intramuscularly with FSH (Antrin) 40 IU per cow with decreasing doses (for 4 days) and second group cows were treated the same way but at day 5 after FSH injection, they were injected intramuscularly with 1,500 IU hCG (Chorulon). Embryo collection was done at day 7 after Artificial Insemination (AI). Average number of corpora lutea (CL) in animals that superovulated with CIDR, FSH and hCG was significantly higher (P < 0.05) compared to animals treated with CIDR and FSH only (5.52). Average number of embryo collection and number of transferable embryos were also higher in group treated with hCG (6.00 vs 5.44) compared with those treated without hCG (4.33 vs 3.17). The conclusion is hCG superovulation injection with CIDR and FSH can increase the respon of superovulation. Key words : superovulation, CIDR, FSH, hCG, beef cattle

Abstract

The occurrence of genetic changes in Bali and Brangus cattle crossing and it's relationship to quantitative traits (e.g. growth and birth weight) were identified using microsatellite markers. Sixteen microsatellite loci were amplified using polymerase chain reaction (PCR) technique, the amplified products were then separated on vertical discontinuous polyacrylamide gel electrophoresis (PAGE) and silver-staining method was applied to detect the fragment. The evaluation to the stability and allele inheritance pattern were checked using Chi-Square segregation analysis. The results of this research showed that allele numbers average detected in Brangus and Bali cattle and their progeny were 2.31, 2.56, and 2.75 respectively. At most microsatellite loci, the allele distributions incline to concentrate and to form bimodal trend. Of 16 micro satellite loci tested, 5 microsatelitte markers namely INRA 037, HEL 9, CSSM 66, INRA 035 and ETH 225 indicated a significant response to average daily gain. Abstract

A total of 134 Pesisir cattle were genotyped for growth hormone (GH) gene by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The genotype and allele frequencies of the GH MspI and GH AluI Pesisir cattle were determined. The GH MspI gene frequencies for the C and T allele were 0.209 and 0.791 respectively, while GH AluI gene frequencies for the L and V allele were 0.992 and 0.008 respectively. The chi-square analysis indicated that this population is not in Hardy-Weinberg Equilibrium status. Expected heterozygosis value (He) for GH MspI and GH AluI were 0.3306±0.0266 and 0.0149±0.0073 respectively. The PCR-RFLP GH MspI marker has higher genetic variability compare to PCR-RFLP AluI marker. This finding showed that GH MspI T allele was favorable as a GH marker for Bos indicus breeds. Abstract

The aim of this study was to know the viability of embryo after fertilized in vitro with frozen separated sperm (sexing sperm). Eleven frozen embryos were transferred to six Bali cow and five FH cow recipients. Two Bali cow recipients were positively pregnant but only one born as male calf which was appropriate with sex of straw being inseminated (Y sperm). One FH cow recipient was also pregnant but aborted in 2.5 months of pregnancy. It was concluded that embryo resulted from in vitro fertilization (IVF) with sexing sperm could grow and develop to be a calf which was appropriate with the sex of straw inseminated.
