Abstract

Perlakuan hormon pertumbuhan pada persilangan antar spesies Glycine max (L.) Merr. kultivar Orba (betina) dengan G. tomentella Hayati PI 441003 (jantan) nyata dapat meningkatkan keberhasilan membentuk polong hybrid. Perlakuan NAA 25 mg/l + IAA 5 mg/l memberikan hasil paling baik karena dapat meningkatkan persen keberhasilan membentuk polong mulai umur 8 hari setelah penyerbukan (hsp) sebesar 49.0%, menunda umur polong gugur terlama (54.1 hsp), meningkatkan ukuran polong (panjang 32.0 mm dan lebar 7.3 mm), panjang biji (2.6 mm) dan jumlah biji per polong (2.0), juga dapat meningkatkan bobot biji (0.917 g). Embrio dari polong hibrid ini diharapkan menjadi lebih mudah diselamatkan secara in vitro sehingga dapat meningkatkan keberhasilan membentuk tanaman.

Abstract

Ubijalar merupakan pangan pokok utama bagi suku-suku yang hidup di wilayah yang sumber pangan alternatifnya sangat sedikit seperti suku Dani di kawasan Lembah Baliem yang hampir tidak memiliki tanaman tradisional lain seperti sagu. Ubijalar tidak hanya di budidayakan di dataran tinggi (daerah pegunungan), akan tetapi juga di dataran rendah yang umumnya ditanam oleh masyarakat asli kabupaten Jayawijaya yang menetap di dataran rendah seperti di Kabupaten dan Kota Jayapura. Eksplorasi plasma nutfah ubijalar di dataran rendah Papua dilaksanakan tahun 2008 bekerjasama dengan Dinas Pertanian dan Ketahanan Pangan Propinsi Papua. Dari eksplorasi tersebut telah dikumpulkan sebanyak 40 aksesi ubijalar yang berasal dari beberapa lokasi dataran rendah di kabupaten dan kota Jayapura pada ketinggian 300-500 meter diatas permukaan laut. Evaluasi potensi hasil dan karakter agronomis dari varietas lokal yang di koleksi ini dilaksanakan di tiga lokasi dataran rendah di Tumpang, kabupaten Malang, Jawa Timur pada tahun 2009, Kecamatan Waibu, Kabupaten Jayapura propinsi Papua tahun 2010 dan Kecamatan Wajak, Kabupaten Malang, Jawa Timur pada tahun 2011. Penelitian disusun menurut rancangan acak kelompok dengan tiga ulangan dan 40 perlakuan (varietas lokal) dan ukuran petak 2m x 5m. Panjang gulud 5 meter dengan jarak tanam antar gulud 100 cm dan dalam gulud 25 cm. Hasil penelitian menunjukkan bahwa varietas dan lokasi berpengaruh nyata terhadap hasil dan komponen hasil. Hasil umbi tertinggi diperoleh di daerah Tumpang, kabupaten Malang, Jawa Timur, diikuti oleh lokasi Wajak, kabupaten Malang Jawa Timur dan Waibu, kabupaten Jayapura, Papua dengan rataan hasil masing-masingnya 27,6 ; 25,7 dan 22,6 t/ha. Sedangkan klon yang memberikan rataan hasil yang tinggi dan konsisten di 3 lokasi adalah Ornanining Kakurap 1, Batatas 2 dan Yoka 5 yang memberikan hasil masing-masingnya 34,1; 31,8 dan 29,9 t/ha. Kultivar lokal Sota 2 memiliki bentuk umbi yang bagus serta bentuk dan ukuran umbi yang seragam dengan rata-rata hasil 25,3 t/ha dan bahan kering umbi 32%. Varietas lokal yang memberikan hasil yang tinggi dan kualitas umbi yang baik dignakan untuk tetua persilangan untuk memperbaiki potensi hasil di dataran rendah.

Abstract

This research aimed to investigate the incompatibility levels of controlled cross-pollination in sweetpotato, based on normal seedling percentage and their correlation with seed vigour. The controlled cross-pollination in sweetpotato faces issues due to its cross-incompatibility and self-incompatibility characteristics. Currently, the incompatibility level in sweetpotato is investigated based on the fruit set percentage. However, this criterion lacks the ability in accurately predicting the number of new clones. Therefore, it is essential to study new parameters to create a better investigation of incompatibility in sweetpotato. The materials used in this research consisted of eight sweetpotato clones as female and four sweetpotato clones as male parents. Cross-pollinations were done reciprocally. The experiment was conducted at Indonesian Legume and Tuber Crops Research Institute from April to December 2014. The result showed that totally 5,188 times crossing produced about 25% fruit sets and 10% normal seedlings. The use of normal seedlings percentage as a new parameter in evaluating cross-pollination has apparently seemed to be more effective than the fruit sets percentage method because numbers of new clones could be known accurately. This research revealed that the normal seedling could be used as a new parameter in determining the incompatibility level in sweetpotato controlled cross-pollination.

Abstract

Two AI-tolerant genotypes (KB 44001 and Genjah Jepang) and two AI-sensitive genotypes of soybean (Kedelai Lumut and KB 46015) were subjected to Al concentration (1.50 mM Al with pH 4.0) in hydroculture for 3 weeks; the growth and their physiological responses were identified by measuring the changes of pH media, NO3ˉ /NH4+ uptake, ion absorption and organic acid content in the tissues. The result revealed that Al-tolerant genotypes soybean show a much better growth than the sensitive genotypes. Moreover, the tolerant genotypes showed the different responses to Al stress; they changed the pH media more significantly, accumulated less Al in the roots and accumulated more oxalate acid than the sensitive genotypes. Both the tolerant and sensitive genotypes had similar pattern n in N03ˉ /NH4+ uptake and other cation uptake. The results suggested that tolerance of soybean genotypes to Al toxicity is associated with their ability in the exclusion AI.

Abstract

Latex yield of rubber tree (Hevea brasiliensis Muell. Arg.) is a complex trait influenced by external and internal factors. In this trial, F1 progenies of two populations (crossing PB 260 as high latex yielder x PN as low latex yielder) and their parents were used to study the characteristic of yield components, namely physiology, anatomy and growth characters which influence latex yield. Based on statistic analysis, it showed that latex yield of two populations had high variation among their F1 progenies. The other variables such as girth, bark thickness, number of latex vessel rings, plugging index, sucrose content and inorganic phosphate content also showed high variation. Based on path analysis and stepwise regression, it showed that the number of latex vessel rings and plugging index had bigger direct effect on determining latex yield of A population (PB 260 x PN 7111), while girth and plugging index variables had bigger direct effect on latex yield of C population (PB 260 x PN 7).
