Abstract

Plant biochemical resistance towards virus infection can be evaluated through measurement of salicylic acid accumulation and peroxidase concentration. A research was conducted to study biochemical resistance of chillipepper to infection of Pepper yellow leaf curl Begomovirus (PYLCV) the causal agent of pepper yellow leaf curl disease. Virus inoculation was done using insect vector, Bemisia tabaci. Salicylic acid accumulation and peroxidase concentration on 6 chillipepper genotypes (35C2, IPB C15, IPB C26, IPB C10, IPB C14, dan IPB C12) was measured 120 h after inoculation. The results showed that salicylic acid accumulation and peroxidase concentration were higher on plants after virus infection. Susceptible genotypes (35C2, IPB C15, IPB C26) tend to have higher salicylic acid accumulation and peroxidase concentration than those of resistant genotype (IPB C12). Positive correlation was observed between biochemical resistance (salicylic acid accumulation) and structural resistance (length of palisade meshophyll cell).Key words: Begomovirus, Pepper yellow leaf curl virus, peroxide enzyme, resistance mechanism, salicylic acid

Abstract

Inoculation of mild virus strain prior to severe virus strain to protect plant against viral disease is the principle of cross protection. Five mild strains of Chili veinal mottle virus (ChiVMV), i.e. -KAR, -SPR, -SKT, -CSR, and -PGL were used as cross protection agent to protect chili pepper plants against severe strain infection of ChiVMV-CKB. The mild strains were inoculated mechanically prior inoculation of severe strain and the efficiency of cross protection was evaluated by observing symptom development and measuring crop yield. Inoculation of mild strains 7 days prior inoculation of severe stain was not able to protect the plant from infection of severe strain ChiVMV-CKB. Protective effect was observed when mild strains were inoculated at 14, 21, and 28 days prior inoculation of severe strain. Symptom development was suppressed or delayed, and crop yield was not significantly different with healthy plants. It was suggested that to obtain the best protection against severe strain, the mild strain should be applied as early as possible before the occurrence of severe strain infection.

Abstract

Planting pepper hybrid varieties offers an alternative strategy to meet needs of chili pepper in Indonesia. The aim of the experiment waere to evaluate and select nine pepper hybrids thus obtained peppers hybrid potential to be released as hybrid varieties of chili pepper. The experimental design used was Randomized Complete Block Design (RCBD) with one factor and three replications. Plant mateterials were nine pepper hybrid and five commercial hybrid of chili peppers. IPB CH3, IPB CH50 and IPB CH28 pepper hybrid  produced higher selection index value than comercial hybrid of chili pepper. The IPB IPB CH3, IPB CH50 and IPB CH28 pepper hybrid were potential to be released as hybrid varieties of chili pepper.
