Abstract

Cardiovascular disease can occur due to disruption of the hemostatic system by forming blood clots in blood vessels, or commonly known as deep vein thrombosis. The curative action that has been carried out in the form of heparin anticoagulant administration had many side effects such as bleeding, hematuria, thrombocytopenia, and hypersensitivity; therefore, alternative anticoagulant ingredients such as onion bulbs have emerged. Dayak onion bulb (Eleutherine bulbosa) is known to contain eleutherinol compound, which has anticoagulant activity. This study aims to determine the potential anticoagulant activity of Dayak onion bulbs extract (Eleutherine bulbosa) on human blood samples. This anticoagulant activity test was carried out by observing the length of time for blood clotting in the extract treatment group with several concentrations (0, 10, 12.5, 16, 25, and 50%) by Clotting Time (Lee-White) method. The results showed a significant delay in blood coagulation time in the group treated with Dayak onion bulbs extract as increasing the concentration. About 50% Dayak onion onion bulb extract was showed a twofold increase in blood coagulation time thanthat of no extract administration. So that, Dayak bulb onion has the opportunity to be an alternative natural anticoagulant. Keywords: anticoagulants; Dayak onion bulbs (Eleutherine bulbosa); clotting time

Abstract

Tuberkulosis merupakan suatu penyakit kronik menular yang disebabkan oleh Mycobacterium tuberculosis yang menyebabkan jumlah kematian global mencapai 1.3 juta pada tahun 2020 dan ditambah parah setelah adanya pandemi COVID-19 yang mengakibatkan terbatasnya layanan tuberkulosis, kasus di Indonesia selalu mengalami peningkatan tiap tahunnya, tahun 2019 ditemukan sebanyak 543.874 kasus meningkat dari 420.994 kasus di tahun 2017. Metode pemeriksaan yang banyak digunakan di negara endemik tuberkulosis adalah Mikroskopis, namun terdapat beberapa kekurangan dalam penggunaanya dibandingkan dengan metode Tes Cepat Molekuler. Penelitian ini bersifat observasional dengan pendekatan cross sectional yang bertujuan untuk mengetahui perbedaan hasil pemeriksaan Mycobacterium tuberculosis pada pasien suspek tuberkulosis dengan menggunakan Tes Cepat Molekuler dan Mikroskopis di RSUD Karangasem. Penelitian ini dilaksanakan dari bulan Desember 2021 sampai April 2022 menggunakan sampel sputum yang diperoleh dari RSUD Karangasem. Penelitian menunjukan adanya perbedaan hasil sensitivitas dan spesifisitas pada dua metode. Sensitivitas metode Tes Cepat Molekuler lebih tinggi daripada metode Mikroskopis (100% dan 75%) sedangkan nilai spesifisitas metode Mikroskopis lebih tinggi daripada metode Tes Cepat Molekuler (100% dan 95%).

Abstract

Quality control is an activity that can improve the quality of the laboratory which is carried out byconducting an examination using a control serum. Homemade lyophilized serum is an alternative control serumthat is relatively inexpensive and easy to obtain than commercial control serum. This study aimed to determinethe stability of reconstituted homemade lyophilized control serum stored for 8 weeks at a temperature of (-2)ºCto(-4)ºCand(-20)ºConcreatinineandBUNlevels. This research was conducted at the Clinical Laboratory of Poltekkes Kemenkes Surabaya in March-May 2022 with the type of experimental research and a one group pretest-posttest design. The reconstitutedhomemade lyophilized serum was stored at (-2)ºC to (-4)ºC and (-20)ºC, the serum was checked once a week for8weeks. The results of homemade lyophilized serum creatinine and BUN levels after reconstitution stored at (-2)ºC to (-4)ºC and (-20)ºC were not out of the control limit ±2SD so they could be declared stable with a CVcreatinine value 4-6% and BUN 7-8%. Based on the regression test analysis, it is known that storage time has aneffect of 51.3% on serumcreatinine levels stored at (-2)ºC to (-4)ºC and has an effect of 79.3% on serumcreatinine levels stored at (-20)ºC. Storage time has an effect of 76% on serum BUN levels stored at (-2)ºC to (-4)ºCand75.1%effectonserumcreatininelevels storedat(-20)ºC.

Abstract

ABSTRACTCigarettes contain various harmful substances that can endanger health. One of them is nicotine which causeslipid metabolism disorders, including a decrease in HDL cholesterol levels and an increase in total cholesterollevels. Smoking habits in people with comorbidities such as diabetes mellitus can exacerbate existingdyslipidemia conditions. If not managed properly, it can cause complications that lead to increased mortality inpeople with diabetes mellitus. This study aims to determine the differences in HDL cholesterol and totalcholesterol levels in active smokers and passive smokers patients with diabetes mellitus at PuskesmasTanjunganom, Nganjuk Regency. This research uses observational study with a cross sectional approach whichwas conducted in January-May 2021 on 30 patients with diabetes mellitus at Puskesmas Tanjunganom, NganjukRegency, consisting of 15 active smokers and 15 passive smokers. Checking total cholesterol levels was carriedout at Puskesmas Tanjunganom, Nganjuk Regency. While the examination of HDL cholesterol levels wascarried out at UPT Labkesda, Nganjuk Regency. Based on the independent sample t-test statistical test, it wasfound that the p-value of difference HDL cholesterol levels in active smokers and passive smokers with diabetesmellitus was 0,000 and the difference total cholesterol levels in active smokers and passive smokers withdiabetes mellitus was 0.012. So it can be concluded that statistically there are differences in HDL cholesteroland total cholesterol levels in active smokers and passive smokers who suffer from diabetes mellitus atPuskesmas Tanjunganom, Nganjuk Regency.Keywords: Diabetes Mellitus; Cholesterol Levels; Active Smoker; Passive SmokerABSTRAK Rokok mengandung berbagai zat berbahaya yang dapat menggangu kesehatan. Salah satunya nikotin yangmengakibatkan gangguan metabolisme lipid antara lain penurunan kadar kolesterol HDL dan peningkatan kadarkolesterol total. Kebiasaan merokok pada orang dengan penyakit penyerta seperti diabetes melitus dapatmemperburuk kondisi dislipidemia yang sudah ada. Apabila tidak dikelola dengan baik dapat mengakibatkankomplikasi yang menyebabkan peningkatan mortalitas penderita diabetes melitus. Penelitian ini bertujuan untukmengetahui perbedaan kadar kolesterol HDL dan kolesterol total pada perokok aktif dengan perokok pasif yangmenderita diabetes melitus di Puskesmas Tanjunganom Kabupaten Nganjuk. Penelitian ini merupakan penelitanobservasional dengan pendekatan cross sectional yang dilakukan pada bulan Januari-Mei 2021 terhadap 30pasien penderita diabetes melitus di Puskesmas Tanjunganom Kabupaten Nganjuk yang terdiri atas 15 orangperokok aktif dan 15 orang perokok pasif. Pemeriksaan kadar kolesterol total dilakukan di LaboratoriumPuskesmas Tanjunganom Kabupaten Nganjuk. Sedangkan pemeriksaan kadar kolesterol HDL dilakukan di UPTLabkesda Kabupaten Nganjuk. Berdasarkan uji statistik independent sampel t-test diperoleh p-value perbedaankadar kolesterol HDL pada perokok aktif dengan perokok pasif penderita diabetes melitus sebesar 0,000 sertaperbedaan kadar kolesterol total pada perokok aktif dengan perokok pasif penderita diabetes melitus sebesar0,012. Sehingga dapat disimpulkan bahwa secara statistik terdapat perbedaan kadar kolesterol HDL dankolesterol total pada perokok aktif dengan perokok pasif yang menderita diabetes melitus di PuskesmasTanjunganom Kabupaten Nganjuk.Kata Kunci : diabetes melitus; kadar kolesterol; perokok aktif; perokok pasif
