Abstract

Diabetes mellitus is a major source of morbidity in developed countries. The morbidity and mortality areincreased in patients with diabetes mellitus compared with the general population. Relationship between highsugar or fat consumption with the increasing of glucose or blood cholesterol concentration has been wellunderstood, but the mechanism of diabetes mellitus as a result of streptozotocin injection and its relation withglucose and cholesterol concentration still need to be investigated. So far, many research mention thatstreptozotocin causes only the damage of a cell of pancreas which that directly relates to metabolism of bloodsugar. The purpose of the research is to investigate the mechanism of pancreas damage in its relation with lipidperoxidation. Sixteen male Wistar rats, 200 grams of bodyweight were used as animal models. Rats weredivided into two groups of eight animals each. Group I was not injected with streptozotocin, while Group II isinjected intraperitoneally with streptozotocin 50 mg/kg BW.Three days after the injection, blood samples weretaken intracardially for Malondialdehida (MDA), glucose and total cholesterol analysis. Abstract

Enterectomy is an operative method that has very dangerous risk. The purpose of this research was done to see the physiological view after surgery, especially in the total leucocyte, C-reactive protein (CRP) andfibrinogen concentrations. Nine female, healthy local dogs, 10 kgs of body weight were used as experimental study. The dogs were adapted for a week into individual cages and fed commercial dog-food and water adlibitum. In the day of 7, all dogs were fasted 12 hours for anaesthesia preparation. The dogs were then divided into 3 Groups of 3 each. Group I was used as control, it had normal enterectomy surgery. Group II was animals that had enterectomy surgery with one hole leakage. Group III was animals that had enterectomy surgery with two special hole leakages. Enterectomy itself was done on to jejunum with 5 cm of gut should be cut away. Anastomosis method was done using end to end method and interrupted suturing method with chromic cat gut 0/3. The result of the research showed that there is similarly increasing level of leucocyte and CRP in the second day after surgery and decreased gradually until the sixth day, but different type of changes in fibrinogen level. The increasing level of CRP has advantages, such as it can be used as eidker marker of inflammation or infection, or worse condition after enterectomy.Keywords : enterectomy, leucocyte, C-reactive protein, fibrinogen, inflammation

Abstract

Osteogenic activity  in  normal fracture  healing will start  early  at  around  1 week after  the  repair ,  but not necessarily occur in fracture healing with the aid of grafts. Demineralized freeze dried bovine bone xenograft (DFDBBX) is a bone graft material made from cows that are    widely produced recently to assist the process of bone  grafting  in  humans,  but  has  not  been  used  on  animals.  It  does  not  know  how  it  works  in  helping  to  repair  the bone.  T wenty male SD rats  weighing 150 g, were divided into  2 groups of 10 rats  each. Group I  was the  control group, the  rats  were fractured  and reconnected  immediately  using intramedullar  pin fixation,  Group II  was a group  of  rats  that  were    fractured  and  reconnected  immediately  using  pin  fixation  intramedular  plus  graft material of freeze dried bovine bone xenograft. Fracture of the left femur was made in diaphysis part of bone. Evaluation  of  joints  performed  regularly  at  1  week  ,  2  weeks,  4  weeks,  6  weeks  and  8  weeks  post  surgery respectively by selecting at random, each group of 2 rats. Rats at each observation were collected two  head / group,      rats    were  then  killed,  the  bone  parts  to  be  joined  was  taken  and  stored  in  10%  of  formalin  for histopathologic  examination.  Some  pieces  of  the  bones  were  used  for  the  analysis  of  bone  morphogenetic  protein 2 (BMP-2 ) by immunohistochemical method. The results were then analyzed descriptively to see the changes directly on the X-rays and tissue. Overall results of this study showed that the graft material of DFDBBX were osteocondusive  rather  than osteoinductive  properties.  Nevertheless,  the  biomaterial  is  useful  for  orthopedic purposes  primarily  of  handling  cases  with    partial  fracture  of  bone  tissue  which  is  lost.  Due  to  the  nature osteocondusive  of  DFDBBX  will  make  cells  to  grow  and  to  unify  the  separate  bone  fragments.Keywords  :  fracture,  osteogenic  activity ,  demineralized  freeze  dried  bovine  bone  xenograft,  bone morphogenetic  protein-2,  immunohistochemical  analysis

Abstract

This research aims to evaluate the safety of dexamethasone use as an anti-inflammatory and analgesic on the handling of femur fracture of rats by bone graft method. The sample used was 32 male Wistar rats aged 2 months were divided into two groups, 16 rats for each. Group I was fractured rats and fixed with intramedullary pin without bone graft, whereas Group II was fractured and fixed with intramedullary pin and demineralized freeze-dried bovine bone xenograft (DFDBBX). Group I and II were then divided into two groups for each, Group IA, IB, IIA, and IIB, eight rats for each. Group IA and IIA were not injected with dexamethasone, whereas Group IB and IIB were injected with dexamethasone 0.5 mg/kg BW/ day intramuscularly. At the end of the first week, blood was collected from eight rats (two rats for each) of Group IA, IIA, IB, and IIB for total leucocytes and blood Calcium analysis. Urine was also collected for urine Calcium analysis. Rats were then killed to collect the femur for histopathologic analysis using HE staining and immunohistochemistry to analyze the expression of TGF?-1 using Streptavidin-biotin method. Subsequent sampling was conducted at the end of 2nd, 4th, and 8th weeks of treatment, respectively. The total data of leucocytes, blood, and urine Calcium were statistically analyzed using a 2x2x4 factorial method, while the histopathologic and immunohistochemistry specimens were analyzed descriptively under the microscope based on the alteration of the bones. The statistical analysis showed that dexamethasone increased the concentration of urine Calcium (p<0,05), the concentration of blood Calcium insignificantly (p>0,05) affected by dexamethasone but cause homeostatic calcium disorder and the total leucocytes significantly (p<0,05) affected by grafted materials. Histopathologic analysis of the femur showed that there was an increased infiltration of inflammatory cells in the fracture area from the group untreated with dexamethasone. Whereas the immunohistochemistry analysis showed positive results from all groups. Based on the whole analysis, it can be concluded that although dexamethasone depressed inflammation process, it can also disturb the fracture healing process.

Abstract

Leptin is a hormone produced by adipocytes and play an important role in wound healing. The objectiveof this research was to study the role of leptin in wound healing in mice experimentally given high fat dietsand its correlation to zinc. Thirty two male Sprague Dawley rats at three months of age were used in thisstudy. Rats were randomly allotted into four groups (A,B,C and D) of 8. Rats in group A and B were fednormal diet, while rats in group C and D were fed high fat diet. After two months of treatment, skin incisionsurgery was performed at the back side of the rat. Incision wound was closed with single interruptedsuture. The wound of rats in group A and C were treated with vaseline, while in the group B and D weretreated with zinc 10%. One day after surgery, blood sample were collected frpm four rats from each groupand analysed for leptin (Rat leptin ELISA Int, Genway Biotech Inc). Wounded skin from all animals werealso taken for histopathological examination (Haematoxylin and Eosin). Three days after the surgery, thesame analysis were done for the remaining rats. Leptin level was analyzed statistically using ANOVA forfactorial experiment, while histopathologic analysis was done descriptively. The results showed that theleptin level was significantly affected by time (P<0.05), as leptin level in blood at three days after surgerywas significantly lower than the level at one day after surgery. Meanwhile, histopathological examinationshowed that the percentage of epidermal closure in animals in group A,B,C,and D were 75%, 100%, 25%and 75%, respectivelly. Therefore, it was concluded that topical application of zinc might have significanteffect on the wound healing of the rats fed normal diets as well as these that given a high fat diet.

Abstract

Penelitian ini bertujuan mengetahui peran zink pada kesembuhan luka dalam kondisi berat badan berlebih. Dalam penelitian digunakan 20 ekor tikus Sprague Dawley jantan berumur tiga bulan (150-200 g). Tikus dibagi secara acak menjadi empat kelompok (A, B, C, dan D), masing-masing kelompok terdiri atas lima ekor. Kelompok A dan B diberi pakan normal, sedangkan kelompok C dan D diberi pakan lemak tinggi. Setelah dua bulan perlakuan pakan, seluruh tikus dioperasi untuk membuat luka irisan pada kulit, kemudian luka ditutup dengan jahitan. Luka jahitan pada kelompok A dan C diberi aplikasi topikal unguenta vaseline album (tanpa zink), sedangkan luka jahitan pada kelompok B dan D diberi aplikasi topikal unguenta zink 10% dengan bahan dasar vaselin album. Lima hari setelah operasi, seluruh tikus diambil darahnya untuk dianalisis terhadap jumlah leukosit total kemudian diikuti pengambilan jaringan kulit untuk pemeriksaan histopatologis dan penghitungan jumlah leukosit di jaringan. Hasil penelitian menunjukkan adanya interaksi yang signifikan antara pakan dan aplikasi topikal terhadap jumlah leukosittotal dalam darah (P0,05). Jumlah leukosit di jaringan mengalami penurunan setelah aplikasi topikal zink, meskipun hasilnya tidak signifikan (P0,05). Analisis histopatologis menunjukkan epidermis sudah tertutup pada seluruh tikus perlakuan, namun demikian sebagian tikus masih menunjukkan dermis yang terbuka, yaitu kelompok A sebanyak 60%, kelompok B sebanyak 60%, kelompok C sebanyak 60%, dan kelompok D sebanyak 40%. Dapat disimpulkan bahwa proses kesembuhan luka setelah pemberian topikal zink pada tikus yang diberi pakan lemak tinggi menunjukkan hasil yang baik, yaitu ditandai dengan penutupan lapisan epidermis dan dermis, serta menurunnya jumlah leukosit di jaringan.
