Abstract

Freeze-drying technique is an alternative method on sperm preservation. However, this process can results in DNA damage. Adding antioxidants before freezing can reduce the detrimental effects of ROS on spermatozoa. Alpha lipoic acid (ALA) is one of the potent antioxidant that can be used to protect the sperm DNA. This study was aimed to determine the effect of ALA supplementation before freeze-drying on the DNA fragmentation index (DFI) of human spermatozoa. Nine semen samples were collected and evaluated for DFI (O1) then divided into 3 groups; freeze-drying without ALA (O2), freeze-drying with ALA 1,25 mg (O3) and freeze-drying with ALA 2,5 mg (O4).All three groups were stored at 4oC for 1 week. Sperm DFI were evaluated before and after freeze-drying. The result of this study showed a significant increase in sperm DFI in all three groups after freeze-drying (p<0,05) when compared to DFI before freeze-drying (14,67 ±3,295). However there was no significant difference between O2 (26,90 ±9,180) and O4 (29,29 ±5,524) group where the supplementation of 2,5 mg ALA did not have significant effect on protecting the DNA of freeze-dried human spermatozoa. The highest DFI was in the O3 group (44,64 ±11,717). Therefore, ALA supplementation before freeze-drying does not have a significant effect on protecting the DNA of freeze-dried human spermatozoa.

Abstract

Congenital deafness will impact the quality of life of affected individuals if they do not obtain early detection and intervention. Socialization of congenital deafness is needed, thereby the community can engage in early detection and intervention efforts. Socializing about the early detection and intervention of congenital deafness. Educational activities are carried out with counseling using the lecture method, discussion, and pretest and posttest. The average pretest result was 62.03 (± 11.51) and the posttest average was 88.39 (± 13.41). Based on the comparison between the pretest and posttest results, we found a significant difference (p < 0.0001) statistically. Public knowledge of congenital deafness needs to be improved in order to increase community participation in terms of  early detection and intervention.

Abstract

Background: The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a virus with a high virulence which cause SARS-CoV-2, a disease with potentially dangerous implications for human health and pandemic. The involvement of other organs in the spread of this virus is still being debated. Considering the presence of (Angiotensin Converting Enzyme-2 (ACE-2) and Transmembrane Serine Protease 2 (TMPRSS2) in the reproductive organs including male reproduction,the male reproductive system possiblity for spreading SARS-CoV-2 should be studied. Reviews: Five studies were reveal the presence of SARS-CoV2 in semen. The reported results are inconsistent. Some of these studies also used unclear methods and procedures, which led to bias in the final results. Ongoing studies are needed to confirm the definite findings before specific recommendations can be made for further management. Summary: There is no definite interpretation of whether SARS-CoV-2 spreads through semen, but protection is still needed when it comes into contact with the semen.

Abstract

Background: LenshookeTM Semen Quality Analyzer (SQA) X1 Pro is an automated semen analysis. The accuracy of LenshookeTM SQA X1 Pro has never been analyzed with World Health Organization (WHO) standard method. Aim: This study aims to examine whether the LenshookeTM SQA X1 Pro method provides reliable results according to the WHO standard method. Methods: This study was a laboratory analytic observational study using 60 patients in Andrology clinic of Dr. Soetomo Hospital. The concentration, progressive motility (PR), total motile sperm count (TMSC), and morphology results of the LenshookeTM SQA X1 Pro and standard method were analyzed statistically using correlation, Bland Altman, and diagnostic test. Results: Significant correlation between two methods were found in all parameters (concentration: r = 0,970; PR: r = 0,781; TMSC: r = 0,952; morphology: r = 0,568). The mean difference for concentration, PR, TMSC, and morphology between the two examination methods were 1,165 million/ml, 7,05%, 7,584 million/ejaculate, and 2,25%. However, it found that the correlation and agreement were weaker in sample with low number of spermatozoa per high power field. The results revealed a sensitivity of 100%, 81%, and 59% for oligozoospermia, astenozoospermia, and teratozoospermia, respectively. The specificities were shown to be 100%, 74%, and 100% for oligozoospermia, astenozoospermia, and teratozoospermia, respectively. Conclusion: The LenshookeTM SQA X1 Pro gives a reliable result for determining oligozoospermia and asthenozoospermia, but in the situation that the clinicians need the accurate data, standard method should be used.

Abstract

Background: Technologies are replacing manpower many fields including medical field. Several devices have been marketed for replacing/reducing manpower in the medical field including male infertility. Here we reviewed several technologies that developed in male infertility. Review: Computer assisted sperm analysis (CASA), Automatic assessment of biochemical marker of seminal plasma, B-mode ultrasound, and automatic sperm cryopreservation can be applied routinely. Several updates i.e. automatic histopathology assessment, ultrasound strain elastography, magnetic resonance imaging (MRI) stronger than 3.0 T, Artificial intelligence for predicting the presence of sperm in azoospermia cases, automatic sperm selection, and automatic intracytoplasmic sperm injection (ICSI) need more studies before their application. Summary: Prudent choice based on valid studies is needed in order to give a comprehensive management to patient with male infertility without using useless technology.

Abstract

The cryopreserve technique using liquid nitrogen is used to preserve human sperm. Nevertheless, this technique has several disadvantages, such as bacterial/viral contamination and its expensive cost. Freeze-drying process offers more simple and cheaper of sperm storage technique. This study aimed to investigate the effect of storage temperature and period of freeze-dried human sperm on DNA integrity. Human sperm samples of 15 donors were prepared with simple washing techniques. Post-washing samples (T0) were divided into 4 groups with the same volume. The four group of samples carried on the freeze-drying process and stored based on different temperature and period: 4°C for 1 week (T1), room temperature (24-25ºC) for 1 week (T2), 4°C for 3 months (T3), and room temperature for 3 months (T4). Sperm DNA integrity was analyzed before and after the storage period of each group.
