Abstract

Research about the hydrolysis of l-menthyl acetate catalyzed by lipase enzyme from Candida antarctica recombined Aspergillus oryzae has been observed by in vitro and in silico. The reaction by in vitro method performed with variations time and solvent. The result obtained that lipase enzyme from Candida antarctica recombined Aspergillus oryzae were able to catalyze the hydrolysis reaction of l-menthyl acetate to produce l-menthol. The best result is observed at 16 hours with ethanol solvent, which give % conversion about 13,76%. The research followed by in silico using the docking method,  observed that mechanism between amino acid residues to macromolecules as well as the influence of solvents on such interactions. The residues that involved in the reaction are Glu, Thr, Asp, Gln, Ser, His. The ethanol solvent shows a good interaction with macromolecules by forming hydrogen bonds.

Abstract

Structure-odor relationship (SOR) has previously studied by semantic numerically in different Fragrance. We hypothesise that in silico method such as molecular dynamics, together with docking of the interaction between human olfactory receptor (OR1G1) and ligands, can offer extremely valuable tools of modelling SOR. Hence, the present study was carried out to express the SOR of citronellal oil fraction compare with reference smelling of floral, musk, green, wood, and fruit by employing docking and multiple discriminant analysis (MDA). Our study reveals that the number dissociation constant (Kd), bond distance, HOMO-LUMO (AE), dipole moment, kind of amino acids, Log P, surface area and hydropathy as the variable SOR from in silico anaysis. Our result has shown ligands and OR1G1 interacted with Van Der Waals and electrostatic model. MDA analysis shown molecule reference floral and fraction of lemongrass oil  have similar correlation based on variable SOR with linier regression of all variable SOR to Kd value for every reference odor is R2 = 1.

Abstract

Microwave-assisted synthesis of zinc oxide-silver (ZnO-Ag) nanocomposite using clove (Syzygium aromaticum L.) oil has been investigated. Variation of solvents and irradiation time of microwave was studied. The product characterization was carried out using XRD, FT-IR, and SEM-EDS. The XRD analysis indicates ZnO-Ag nanocomposite has a polycrystalline structure. The ZnO peak was detected at 2θ = 31.99; 34.58 and 68.05, whereas the Ag peak was detected at 2θ = 38.36 and 44.49. The increasing of irradiation time reduces the crystallite size, and generally has size range between 9 and 12 nm. SEM-EDS confirmed the existence of the ZnO-Ag nanocomposite with percentage of Zn (46.89%), O (29.72%) and Ag (23.39%). Moreover, antibacterial evaluation on Staphylococcus aureus give the inhibition zone in 13.3 mm. This result slightly gives better activity than the reference.

Abstract

Kandungan senyawa aktif dalam tanaman dapat di analisis dengan metode uji fitokimia. Salah satu tanaman herbal yang banyak dikonsumsi adalah secang (Caesalpinia sappan). Secang biasa di simpan dalam bentuk sediaan teh herbal, diseduh dengan air panas dan diminum untuk mengurangi radang tenggorakan. Dalam rangka mengenalkan kegiatan riset sederhana di sekolah dilakukan pelatihan uji fitokimia metabolit sekunder tanaman lokal (dalam hal ini secang) kepada guru-guru kimia yang tergabung dalam Perkumpulan Pendidik Sains Kimia Indonesia (PPSKI). Metode yang dilakukan dengan pemberian materi secara daring menggunakan media zoom meeting, tutorial melalui video, diskusi, tanya jawab, dan pemberian tugas. Dalam pelatihan ini diberikan juga soal-soal pretes dan postes untuk mengukur kemampuan peserta sebelum dan sesudah pelatihan. Terdapat peningkatan pengetahuan peserta sebanyak 20% setelah pelatihan. Pelatihan uji fitokimia ini memberikan wawasn baru kepada para guru kimia untuk melakukan riset sederhana dengan bahan yang tersedia.

Abstract

Star anise (Illicium verum) is widely used as a medicinal herb and spice. The extracts, essential oil and oleoresin, can be produced by hydro-distillation and steam distillation, while solvent and supercritical fluid extraction can be used to extract oleoresin. In this work, the star anise is distilled to obtain the oil, subsequently, its residue is extracted by solvent extraction to get the oleoresin. Whole and grounded star anise fruit is distilled by hydro-distillation for 8, 12, 16, 20, and 24 hours, thus the highest yield is obtained at 20 hours from grounded fruit, and the highest trans-anethole content of 68.50% is obtained at 8 hours from grounded fruit. Residue from the whole fruit-distillation process is extracted by Soxhlet extraction with three types of solvent (ethanol, diethyl ether, and n-hexane) for 6 hours. The highest yield was obtained from ethanol extraction with 1.16% yield and the highest content of trans-anethol was obtained from n-hexane fraction. One sample with the highest content of anethole from whole and grounded fruit-distilled oil and oleoresin are picked, and later they are tested to measure their antioxidant capacity by Ferric Reducing Antioxidant Power Assay (FRAP). Abstract

Breast cancer is among the diseases with the highest mortality rate in women in the world. The triple negative cancer subtype with aggressiveness and metastatic ability causes the sufferer to be difficult to treat. Targeted treatment efforts of natural ingredients such as limonene and its derivatives are more profitable due to their easy excretion process. Screening the activity of compounds through docking specifically provides convenience in the synthesis process in the laboratory. Limonene compounds and their derivatives will be interpreted against the enzymes MMP-2 (PDB ID: 3AYU), MMP-9 (PDB ID: 4H1Q) and Cyclin A2 (PDB ID: 2V22) involved in the cellular function of triple-negative breast cancer using PyRx 9.0 software. The docking results showed that the limonelyl salicylate compound provided the best binding affinity value against the enzymes MMP-2, MMP-9, and Cyclin A2 with successive values of -7.7, -8.8, and -6.7 kcal/mol.
