Abstract

Tumbuhan Pala (Myristica fragrans Houtt) termasuk dalam famili Myristicaceae. Tumbuhan ini secara etnobotani terbukti mempunyai aktivitas antibakteri. Penelitian tentang isolasi senyawa flavonoida (total flavonoida) uji aktivitas antibakteri daun Pala terhadap Staphilococcus aureus dan Escherrichia coli telah dilakukan. Ekstraksi 3 Kg (3000 g) daun Pala dengan metanol diperoleh ekstrak metanol 417,60 g (13,92%). Ekstrak metanol di larutkan ke dalam etil asetat diperoleh ekstrak etil asetat 104,1 g (5,96%). Hasil partisi ekstrak etil asetat dengan n-heksana diperoleh total flavonoid 32,76 g (31,47%). Hasil hirolisis total flavonoid dengan HCl 6% diperoleh 1,8 g total flavonoid (bebas glikon). Hasil uji aktivitas antibakteri ekstrak metanol dan ekstrak etil menunjukkan bahwa ekstrak etil asetat daun pala mempunyai aktivitas antibakteri terhadap Staphilococcus aureus dan Escherrichia coli, sedangkan ekstrak metanol tidak aktif.

Abstract

Blumea balsamifera L leaves were isolated and tested for antioxidant activity using the 1,1-diphenyl-2-picrylhydrazyl(DPPH) method. B. balsamifera leaves were macerated as much as 1300 gram using a methanol solvent thenpartitioned with petroleum ether, n-hexane, and extracted with ethyl acetate and acetone to obtain petroleum ether, nhexane, ethyl acetate, acetone and methanol extracts by weight and% yield respectively, namely 6.34 g (5.43%), 4.97 g(4.25%), 7.34 g (6.28%), 14.14 g (12.11%) dan 5.66 (4.85%). Phytochemical tests of petroleum ether and n-hexaneextracts showed the presence of steroid compounds, while ethyl acetate, acetone, and methanol extracts showed thepresence of alkaloids, saponins, flavonoids, terpenoids, and phenolic compounds. The antioxidant activity test ofpetroleum ether, n-hexane, ethyl acetate, acetone, and methanol extracts obtained IC50 values respectively 177.47 ppm,192.66 ppm, 4.28 ppm, 7.33 ppm and 8.61 ppm. Acetone extract of B. balsamifera (BBA) is separated by chemicalcomponents using chromatography of gravity column so that the combined fraction of acetone B. balsamifera extract(BBA1-BBA11) is obtained. Antioxidant activity test of BBA1, BBA2, BBA3, BBA4, BBA5, BBA6, BBA7, BBA8, BBA9,BBA10 and BBA 11 fractions IC50 values obtained were 147.17 ppm, 81.36 ppm, 10.31 ppm, 3.84 ppm, 1.18 ppm, 3.37ppm, 6.76 ppm, 9.32 ppm, 19.26 ppm, 16.82 ppm, and 30.71 ppm. Abstract

Abstract. It has tested the antioxidant activity of n-hexane extract respective roots, bark, fruit and seed crops of nutmeg against DPPH. Each extracted by maceration method with n-hexane. The extraction of each plant nutmeg with n-hexane extract obtained yield of n-hexane respectively 8,35%; 81,5%; 11,89% and 55,63%. The result of the antioxidant activity of n-hexane extract of each plant nutmeg against DPPH with a concentration of 25 ppm, 50 ppm and 100 ppm is obtained IC50, respectively, are 0,216 ppm, 63,755 ppm, 43,998 ppm and 11,599 ppm and positive control vitamin C ( IC50 = 3,657 ppm). Vitamin C is a standard compound is more often used than butyl for very high antioxidant activity. The antioxidant activity increases from high to low at the roots, seeds, fruits and bark of plants nutmeg in a row. Based on data GC-MS chemical components of each successive extracts of roots, bark, fruit and seeds are 41, 41, 44, 43 components of secondary metabolites. The main components of each part of the plant based on data from GC-MS are a 5-Octadecanoic acid, linalool, eugenol, myristicin, methoxyeugenol. Abstract

Abstract.The antioxidant activity from methanol extract of nutmeg leaves (Myristica fragrans Houtt) was performed using 2,2-diphenyl-1-picrylhydrazil (DPPH). Antioxidant assay using DPPH of the methanol extract at a concentration of 25 ppm, 50 ppm and 100 ppm, showed high antioxidant activity with IC50 = 36.31 ppm and Vitamin C showed antioxidant activity with IC50 = 3.657 ppm. The methanol extract of nutmeg leaves were partitioned with chloroform to obtain chloroform extract of the leaves with IC50 = 28.30 ppm. isolation of the active compounds from the chloroform  extract of the nutmeg leaf using column chromatography yielded 6 mix fractions MFMD 1, MFMD 2, MFMD 3, MFMD 4, MFMD 5 and MFMD 6. The test results of antioxidant activity in very strong category is fractions MFMD 4, MFMD 3, MFMD 2, MFMD 6 and MFMD 5 with IC50 of each are 26.590 ppm, 27.239 ppm, 29.639 ppm, 39.766 ppm and 55.436 ppm, while antioxidant activity in a weak catagory is fraction MFMD 1 with IC50 is 126.270 ppm. This shows that the fraction of the leaf has a better inhibitory activity, because some of the active compound contained in the fraction can synergize in inhibiting free radicals, so that the IC50 value of the fraction is stronger than the extract. Keywords: Antioxidant, chromathography, Nutmeg (Myristica fragransHoutt), methanol extract, leaves
