Abstract

A therapy development with recombinant protein is a new and potential innovation to destroy cancer stem cells (CSCs). Various ways of killing CSCs include the provision of polyvalent anti-protein antibodies that code pluripotency. Therefore, it takes a mixture of Oct-4, c-Myc, Sox2, and Klf4 protein antigens that can stimulate the formation of polyvalent antibodies. This study aimed to construct Sox2 recombinant by identifying the target genes by reverse transcriptase PCR and then arranging their designs to be inserted into the cloning vector pET101/D-TOPO®. The target gene was developed by finding the complete sequences of Sox2 nucleotides on the NCBI GenBank. The growth on LB-ampicillin agar plates was amplified by PCR to obtain colonies with pET101/D-TOPO® vectors and inserts of the pluripotent gene of CSCs, then the PCR results were observed through electrophoresis. A total of fifteen colonies have DNA bands with a base pair of about 300 bp in length. The recombinant clones produced Sox2 genes with a base length of 330 bp.

Abstract

Indonesia adalah negara yang memiliki warga negara mayoritas adalah umat muslim. Sebagai umat Islam, kita diwajibkan mengkonsumsi makanan/minuman yang halal. Seiring dengan hal tersebut Pemerintah telah mengeluarkan Undang-Undang Nomor 33 Tahun 2014 tentang Jaminan Produk Halal, maka semua pelaku industri baik di bidang makanan/minuman olahan, produk farmasi dan lain-lain wajib memiliki sertifikat halal. Namun hingga saat ini belum banyak indutri olahan yang mengajukan sertifikat halal terutama pada industri UMKM (usaha mikro, kecil dan menengah). Fakultas Farmasi dan sains (FFS) UHAMKA yang telah memiliki Pusat Kajian Halal UHAMKA (PKHU), berkewajiban untuk mensosialisasi dan mengedukasi kepada seluruh masyarakat mengenai pentingnya “halal” suatu produk. Target pada kegiatan workshop ini adalah pelaku usaha di bidang pengolahan makanan/minuman UMKM di wilayah Duren Sawit Jakarta Timur. Metode yang digunakan adalah dengan melakukan pelatihan/workshop tentang tatacara pengajuan sertifikat halal dan dokumen-dokumen yang harus dipersiapkan. Workshop diawali dengan sosialisasi pentingnya sertifikat halal kemudian dilanjutkan dengan pretest. Pre-test untuk melihat sejauh mana pemahaman terkait sertifikasi halal. Workshop diakhiri dengan post-test, untuk melihat peningkatan pemahaman setelah diberikan sosislaisasi dan pelatihan terkait sertifikasi halal. Hasil yang didapat adanya peningkatan pemahaman peserta (p<0,05) dibandingkan antara sebelum dengan sesudah pelatihan. Kesimpulan dari kegiatan ini adalah workshop yang diberikan bermanfaat untuk para pelaku usaha UMKM dalam meningkatkan pemahaman terkait tatacara sertifikasi halal.

Abstract

Vannamei shrimps (Litopenaeus vannamei), also known as white leg shrimps are widely cultivated and consumed by many people. However, most consumers have removed the shrimp’s head which integrates with its digestive tract. The digestive tract of white leg shrimp contains digestive enzymes, including amylase. This study aimed to determine the protein content and amylase activity from Vannamei shrimps’ digestive tract using the size exclusion chromatography method. The protein isolation of size exclusion chromatography was prepared in three steps, namely; centrifugation, precipitation, and dialysis. The protein from the dialysis step was purified by using gel filtration chromatography. Each obtained fraction was determined the protein content and amylase activity by using spectrophotometer UV-Vis method.
