Abstract

Tujuan penelitian ini adalah untuk mengetahui efek lama pembekuan semen terhadap kadar MDA spermatozoa dan persentase viabilitas spermatozoa. Sampel yang digunakan berupa  semen beku sapi Limousin dengan lama pembekuan 2 bulan dan 3 bulan yang dikoleksi dari BBIB Singosari. Semen beku yang telah dithawing diencerkan dengan Phosphate Buffer Saline (PBS) kemudian disentrifugasi untuk memisahkan spermatozoa dan seminal plasma. Kadar MDA spermatozoa diukur dengan menggunakan uji TBA dan spektrofotometer. Pengamatan viabilitas spermatozoa dilakukan dengan menggunakan pewarnaan eosin-negrosin. Data berupa kadar MDA spermatozoa dan persentase viabilitas dianalisis dengan uji-t berpasangan dan korelasi pearson menggunakan spss 16.0. Hasil analisis menunjukkan bahwa kadar MDA spermatozoa pada lama pembekuan 2 bulan (0,0777±0,008 nM) dan 3 bulan (0,0919±0,016 nM) tidak berbeda nyata (p<0,05). Terdapat korelasi negatif antara persentase viabilitas dan kadar MDA pada spermatozoa. Kata kunci: MDA, semen beku, spermatozoa

Abstract

The aim of this research was to determine the polymorphisms of BMP-15  and GDF-9 gene of  Kacang goat. DNA was isolated from blood samples of five female goats with salting out method. Quantitative and qualitative  analysis of DNA was measured using spectrophotometer and agarose gel electrophoresis. To get DNA fragment GDF-9 gene was amplified using Forward 5'-AGTTTGTACTGAGCAGGTCT -3 ', Reverse 5'-ACTCCGCTTCGTATGTCAG-3', while to get  DNA fragmen BMP-15 gene was amplified using Forward primer 5’-CAAGGAGGGGACCCCTAAAT-3’, reverse primer 5’- ACCAGAGGCTCAAGAGGAGC-3 for GDF-9. The results of amplification was a specific DNA band with the length of 1400 bp for GDF-9 gene  and 400 bp for BMP-15 gene. Restriction with HaeIII enzyme of GDF-9 PCR product was resulted a haplotype with fragment size 1100 bp, 300 bp, and 100 bp. While, restriction with AluI enzymes of BMP-15 PCR product was resulted a haplotype which consist of two fragments with sizes 180 bp and 120 bp. It was concluded that there are no polymorphism of GDF-9 and BMP-15 gene of Goat Beans.

Abstract

Tujuan penelitian ini adalah untuk mengetahui profil protein spermatozoa sebelum dan setelah pembekuan. Protein spermatozoa diisolasi dari 10 sampel sapi Friesian holstein dengan pemisahan fraksi atas dan bawah hasil sentrifugasi. Profil protein kemudian dianalisis menggunakan SDS PAGE dengan konsentrasi separating gel 12,5%. Data yang diperoleh dianalisis secara deskriptif dengan membandingkan profil protein spermatozoa sebelum dan setelah pembekuan. Hasil analisis menunjukkan bahwa pada fraksi atas semen segar ditemukan 12 pita protein spermatozoa dengan berat molekul 144, 128, 99, 90, 83, 59, 31, 20, 14, 12, 10 dan 9 kDa, dan pada semen beku ditemukan 8 pita protein spermatozoa dengan berat molekul 144, 128, 99, 90, 83, 59, 31, 14 dan 12 kDa. Sementara itu pada fraksi bawah semen segar ditemukan 16 pita protein dengan berat molekul 128, 116, 103, 90, 75, 69, 48, 40, 31, 26, 20, 14, 10, 9, 7 dan 6 kDa, dan 11 pita protein dengan berat molekul 128, 116, 103, 90, 75, 69, 48, 40, 31, 20 dan 14 kDa pada semen beku. Dari penelitian yang telah dilakukan dapat disimpulka bahwa terdapat perbedaan profil protein spermatozoa sebelum dan setelah pembekuan yang dilihat dari variasi berat molekul protein pada masing – masing perlakuan. kata kunci        : pembekuan, protein, SDS PAGE, spermatozoa.

Abstract

Pregnancy can cause immune system changes. It is characterized by a decrease in the activity of immunocompetent cells. The use of antibiotics was intended to combat pathogenic microorganisms, but antibiotics have negative effects on pregnant women. The use of antibiotics can be replaced with extracts of Elephantopus scaber and Sauropus androgynus because both plants have chemical compounds that act as immunomodulators. This study was aimed to determine the activity of lymphocytes B220+, TER119+, and GR-1+ on bone marrow pregnant mice given the combination of extracts of Elephantopus scaber and Sauropus androgynus after infected with Salmonella typhimurium. This research uses seven treatment groups name: (K-) 0.05% NaCMC without bacterial infection; (K+) 0.05% NaCMC infected by bacteria; (P1) E. scaber 200 mg.kg-1BW infected by bacteria; (P2) E. scaber 150 mg.kg-1BW and S. androgynus 37.5 mg.kg-1BW infected by bacteria; (P3) E. scaber 100 mg.kg-1BW and S. androgynus 75 mg.kg-1BW infected by bacteria; (P4) E. scaber 50 mg.kg-1BW and S. androgynus 112.5 mg.kg-1BW infected by bacteria; and (P5) S. androgynus 150 mg.kg-1BW infected bacteria. The initial dose of E. scaber and S. androgynus was 50 mg.kg-1. Each treatment has three repetitions, surgery performed on day 12th and 18th. Lymphocyte cells isolated from bone marrow, the obtained results were analyzed by flowcytometry and statistical analysis using SPSS 16.0 one-way ANOVA, Tukey test and path. Based on the results from ANOVA tables, the formulations on mice that can restore their normal conditions with B220+ cells is E. scaber 150 mg.kg-1BW and S. androgynus 37.5 mg.kg-1BW, TER119+ cells is S. androgynus 150 mg.kg-1BW, while the GR-1+ cells affected by the surgery. Those three dose formulations can be used to obtain the optimum value which can increase the number of lymphocytes and not harmful to the developing fetus. Abstract

The aim of this research is to know the effect of diluent Tris-egg yolk medium added with Morinda citrifolia L. extract towards the sperms abnormality after storage at -80°C for 24 hours. The extracts added in diluent Tris-egg yolk medium were 0% (P0), 10% (P1), 20% (P2), and 30% (P3). The result showed that the percentage of sperms abnormality in P0, P1, P2, and P3 were respectively 8.67%, 6.67%, 5.67%, and 10%. P2 was the lowest percentage, while P3 was the highest percentage of sperms abnormality. In addition, this research found twelve kinds of sperms abnormality divided into categories: major abnormality included the broke tail, absent head, detached head, dag defect, tapered head, proximal droplet, round head and abaxial; and minor abnormality included the coiled tail, shoe hook tail, bent tail, and distal droplet. It has been concluded that the allotment of 20% Morinda citrifolia L. extracts in Tris-egg yolk medium able to maintain the sperm morphology at -80°C storage for 24 hours. Abstract

This study aims to determine the effect of monosodium glutamate on LH and testosterone levels in serum of adult male rats (Rattus norvegicus). A total of 12 male Wistar-strained rats divided into three groups with 4 rats per group. Control group was given distilled water, while M1 was given MSG at a dose of 4mg g-1 BW for 15 days and M2 was given MSG at a dose of 4 mg g-1BW for 45 days. All substances were given orally. At the end of the study, rats were sacrificed with a dislocation to draw blood from the heart. Then, the blood was incubated at room temperature to obtain serum. Serum was used to analyze LH and testosterone levels using the ELISA kit. Data were analyzed using one way ANOVA (p ≤ 0.05) with SPPS 16.0 for windows. The results showed that serum LH levels after MSG treatments for 45 days was significantly decreased compared with the control. The serum testosterone levels M2 was significantly difference with the control group (p ≤ 0.01). This study predicts that monosodium glutamate have a direct and indirect effect on male reproductive organs. Keywords: Luteinizing hormone, Monosodium glutamate, testosterone.

Abstract

Hyperglycemia is a medical condition when the blood glucose levels increase exceeds the reasonable limit. Hyperglycemia is a typical sign of diabetes mellitus (DM). Indonesia is the sixth-ranked country in the world after China, India, United States, Brazil, and Mexico. Treatment with herbs is currently being developed. Pare (Momordica charantia) and Belimbing wuluh (Averrhoa bilimbi) are plants that found around us. Some studies state that each of these plants can be anti-diabetic. The hyperglycemia can cause an immune system disorder characterized by pancreatic β cell death involving IL-1β, kappa B (NF)-κB nuclear factor, and Fas. The ability of NF-kB activation will affect the number of cytokines expressed by T cells, namely TNF-α, and IFN-γ. The purpose of this study is to determine the effect of NF-kB activation on blood glucose levels in hyperglycemia mice. The results showed that the positive control treatment showed an increase in the number of NF-kB activations in CD4 and CD8 cells. EPBW (combination of Averrhoa bilimbi extract and Momordica charantia) administration results at a dose of 10 mg.kg-1 BW showed a reduction in the amount of activated NF-kB as a substitute for the reduction. In addition, that dose can reduce blood sugar levels in mice hyperglycemia model.

Abstract

Hyperglycemia is a condition of excessive blood glucose in blood plasma caused by damage to the pancreatic beta-cell structure that causes impaired insulin secretion. This study aimed to investigate the combination of Bitter Melon and Star Fruit ethanol extract (SBME) on pro-inflammatory cytokines production in hyperglycemic albino mice models. This study was treated in 25 female albino mice weighing 25-30 and 9 weeks old. STZ was given in a dose of 145 mg.kg-1 BW intraperitoneally. Hyperglycemic mice were given orally with SBME ethanol extract at doses 10, 40, and 160 mg.kg-1 BW for two weeks. After treatment, pro-inflammatory molecules were analyzed by flow cytometry from the splenic cell. This study showed that SBME treatment can reduce the production of pro-inflammatory cytokines especially IL-1b, but increases IL-6 in hyperglycemic mice. After treatment with SBME at a dose of 40, the IL-1b pro-inflammation molecule decreased significantly (p>0,05). It reached a normal physiological level, but the dose has not been able to reduce the IL-6 pro-inflammation molecule significantly. The conclusion of this study was Bitter Melon and Star Fruit ethanol extract (SBME) with a treatment of 40 mg.kg-1 BW can suppress IL-1b pro-inflammatory cytokines in hyperglycemic mice models, but has not been able to reduce the expression of IL-6 pro-inflammatory cytokines. Suggesting this medicinal herb might be a useful strategy for future therapeutic interventions in degenerative diseases or diseases involving cell activation, but a study of doses is needed.Keywords: Averrhoa bilimbi L., Hyperglycemia, Inflammation, Momordica charantia L.

Abstract

The aim of this research is to know the level of MDA, motility, and viability spermatozoa of C. carpio L. in cold storage time at 4°C. This study used treatment extenders that were NaCl 0.9% as control, diluent of egg yolk with concentrate 0%, 5%, 10%, 15%, and storage in 0 h, 24 h, 48 h, 72 h, and 96 h at 4°C. The data were analyzed using ANOVA (P<0.05). The results showed that the quality (motility and viability) of spermatozoa C. carpio L. punten strain in cold storage could be maintained at 96 h. The optimum storage for motility was found in diluent of egg yolk concentrate 5% at 48 h. MDA levels of semen from C. carpio L. Punten strains after being stored at cold temperatures increased at 0 h to 24 h, then decreased after 48 h of storage and increase at 72 h - 96 h stored. Egg yolk concentration and storage time had a significant effect on MDA levels. There is a negative correlation between MDA levels and motility. There was no correlation between MDA levels and the viability of spermatozoa C. carpio L. punten strain. Keywords: C. carpio L, Egg yolk, Malondialdehyde, Motiliy, Viability

Abstract

This study was conducted to investigate the effect of MSG treatment for short and long term on the semen quality of adult male rats. Twelve male adult Wistar rats with 200-300g of body weight (BW) and 3-4 month of age were used in this study. The animals were divided randomly into 3 groups. M0 was used as a control, M1 and M2 were given with MSG 4mg/gBW for 15 and 45 days respectively. The experimental animals were sacrificed on the days 16th (to M1 group) and 46th (to M0 and M2 groups). The epididymis was isolated and semen quality (motility, viability, concentration, and abnormality of sperm) was evaluated. The results showed motility and concentration of M1 and M2 were not significantly decreased compared to M0. MSG treatment also significantly reduced viability and increased abnormality of sperm. Analysis of sperm abnormality character shows that the use of long-term MSG caused a formation of the primary abnormality (round and double head sperm) and increased the secondary abnormality (bent neck, curve tail, coiled tail, headless, and tailless) compared to control. Conclusion, semen quality decreases with consumed MSG for the long term. For this reason, reconsidering the use of MSG as an enhancer for the teste of food is very important.Keywords: epididymis, Monosodium L-Glutamate, semen quality.
