Abstract

Pollution that occurred in Pasuruan area’s watershed Wangi–Beujeng river, District of Beji, Indonesia has been initiated in 2007 and continues to this day. The activity was caused by many factors such as the industrial (I), household wastes (II) and agriculture wastes (III) as well as the erosion process.The aims of this study to analyzed the heavy metal pollution that occurs in the Wangi river flow and the effect of gill histology and antioxidant activity on gambusia fish. The research method of observation at river flow and sampling for laboratory test.Three sites were assigned for chemical sampling and tissue histologycal in this study. Fish (Gambusia affinis, local name: Gatul) and water were used as indikatorfrom each site to determine of cadmium (Cd), lead (Pb) and mercury (Hg) concentration using Atomic Absorption Spectrophotometry (AAS). The study was indicated that the streams previously used by residents for daily activities. Results showed level of cadmium (Cd), lead (Pb) and mercury (Hg) in Gambusia affinis exceeded the permissible standard (0.01 ppm Cd;0.03 ppm Pb;0.001 ppm Hg)respectively. Based on histologycal, the tissue showed of damage ofchloride cell (CC) which was used in ion homeostasis process and heavy metal route in grill fish. While protease activities, CAT, MDA and peroxidasse was increased in each sampling area, with significant different (>0.95) between the three sampling sitesKeywords: Chloride cell, Heavy metal, Toxicity, Wangi river

Abstract

Desa Tambak Kalisogo merupakan salah satu desa dampingan PT. PGN,Tbk melalui program Corporate Social Responsibility (CSR). Hingga saat ini TPA di desa itu memiliki kapasitas sampah lebih dari 92%. Maggot merupakan salah satu serangga yang dapat digunakan pada teknik biokonversi dan tidak berpotensi menyebarkan penyakit. Metode yang digunakan yakni persiapan kegiatan, pelaksanaan kegiatan, sosialisasi, pelatihan pemilahan sampah dan penanganan sampah organik untuk media budidaya maggot, pelatihan dan praktik budidaya maggot, pelatihan kewirausahaan, serta monitoring dan evaluasi kegiatan. Hasil evaluasi menunjukan bahwa masyarakat Desa Tambak Kalisogo melalui BUMDes ingin serius melaksanakan kegiatan budidaya maggot dengan fasilitasi pembangunannya dari pendanaan CSR PT. PGN, Tbk. Kegiatan dari persiapan, pelatihan, hingga fasilitasi pembangunan paket budidaya maggot saat ini sudah terimplementasikan dan sudah menghasilkan maggot mulai dari stadia telur, stadia larva, hingga stadia lalat.

Abstract

Modern strategies incorporate preventive measures such as extracellular products, as soluble proteins with antigenic properties, as well as generating a broad immune response. The aim of research is to determine and analyze the effect of the extracellular product of E. tarda on enhancing the immunity of Nile tilapia (O. niloticus). The research design with treatment durations of d0, 3rd, 6th, 9th day, and control (A, B, C, D, K). ECP vaccination was injected intraperitoneally at a dose of 0.1 ml/fish. The parameters analyzed are the expression of IL-1, IL-6, and IFN-, phagocytic activity, leukocyte differential, total white blood cells, and relative percent survival. The results of phagocytosis activity were highest in treatment C at 84.18% 1.81. The highest leukocyte differential in treatment C was lymphocytes at 56% 2.00, monocytes at 28% 2.00, and 16% 0.00, with a total leukocyte count of 29 x 10^3 cells 2.64, and RPS values ranging from 83.33% to 100% over 9 days of post-vaccination maintenance. Then, post-vaccination, there was an increase in the expression levels of the cytokine genes IL-1 and IL-6, but there was no increase in the expression levels of the IFN- gene compared to the control in Nile tilapia.
