Abstract

Several studies have shown that pure compounds from west sumatera medicinal plants have beneficial therapeutic effects so that they are potential candidates for active pharmaceutical ingredients (API). Andalas Sitawa Fitolab has been able to produce 10 pure isolates. The development of a new drug candidate requires an in silico study to predict physicochemical properties, potential target, and toxic properties. The purpose of this study was to initially screen the structure of candidates to predict the potential the compound as an API by using big data and machine learning. The chemical structure were analyzed using software and servers. The Software used was Marvin Sketch, QSAR Toolbox, Swiss Potential Target and ChemBioDraw. Results showed that log P of compounds revealed in a range of -0.54 to 4.64, Polar Surface Area (PSA) in range of 20.23 to 315.21. Asiaticoside did not meet Lipinski's rules. Compounds with high potential hazard were catechin, curcumin, andrographolide, asiaticoside deoxyelephantopin, ethylmethoxycinnamate, alpha-mangostin and piperine. The compounds such as curcumin, alpha mangostin, plumbagin, and piperine were predicted to have spesific target proteins. This study concluded that asiaticoside compounds have a high potential hazard, if it was developed as an API.Keywords: analysis of physical-chemical properties, in silico, pure isolate, toxicology

Abstract

SARS-CoV-2 has become a pandemic in the world. The virus binds to the Angiotensin-Converting Enzyme 2 (ACE2) receptor, which is found in epithelial cells such as in the lungs, to generate the pathology of COVID-19. It is essential to analyze the characteristics of ACE2 in understanding the development of the disease and study potential new drugs. The analysis was carried out using computer simulations to speed up protein analysis that utilized Artificial Intelligence technology, databases, and big data. Homology modeling is a method to exhibit homologous of protein families, hence the model and arrangement of protein sequences modeled are established. This research aims to determine the possibility of mutations in ACE2 by performing the mutation prediction. The result shows reliable homologous modeling with the score of GA341, MPQS, Z-DOPE, and TSVMod NO35 were 1; 1.28252; -0.47; and 0.793, respectively. Moreover, Gene Ontology (GO) analysis describes that ACE2 has a molecular transport function in cells while there are no mutations found occurred in ACE2 analyzed using SIFT and PROVEAN.

Abstract

The number of consumer demands for Mitra's taro chips has not been fulfilled due to limited production, which is still traditional using manual equipment and because of the lack of knowledge about business planning and the lack of business development strategies through the application of the marketing mix (4P) using information technology and computers in response to the Covid-19 pandemic. 19. Therefore, Partners need process innovation in the form of modern equipment for product packaging. Paradigm changes to be able to adapt in maintaining their business by using garlic as an antioxidant as a natural preservative for taro chips, as well as knowledge of marketing applications using internet technology to meet consumer demand and reach a broader market. Besides that, it also pays attention to aspects of business legality, such as having a business license to ensure product quality and maintain customer trust. This activity aims to help partners develop their business in the snack food sector, namely taro chips that continue to grow in Tungkar Village, Situjuah District, Limo Nagari through equipment assistance and 4P marketing mix strategies these micro-businesses can survive and increase their results. Partners' theoretical benefits from this activity will be to gain knowledge about business strengthening through innovation (paradigms and processes). Practically, Partners will get business development assistance through more modern equipment for packaging taro chips that are durable and long-lasting. Partners can implement marketing strategies from the 4P aspect using internet technology to reach consumers more broadly.

Abstract

One of the plants that can be used as traditional medicine is mahogany (Sweitenia mahagoni). The presence of flavonoid compounds in mahogany seeds is suspected as a compound that serves to inhibit bacterial growth. One of the antibacterial effects is to treat diarrhea diseases caused by Shigella dysenteriae. Diarrhea is infectious intestinal disease a public health problem in developing countries. The aim of this research is to know and measure the inhibition zone of ethanol extract of mahogany seed to the growth of Shigella dysenteriae bacteria with concentration 25%, 50%, 75%, 100%. This research is a descriptive study in vitro using disc method. The result of the research has found that the average inhibition zone diameter at 25%, 50%, 75%, 100% concentration is 6mm, 8mm, 8.33mm, 10.33mm. Negative controls do not provide inhibition and positive control yields an average diameter of 21.33 mm. Based on the above results, it can be concluded mahogany seed extract provides resistance to Shigella dysenteriae.

Abstract

Kulit buah G. Mangostana kaya akan senyawa antioksidan yang dapat menghambat radikal bebas. Terdapat hubungan antara pemanasan dan aktivitas antioksidan suatu bahan alam, dikarenakan pengaruhnya terhadap komposisi senyawa bioaktif antioksidan. Tujuan penelitian ini adalah untuk melihat pengaruh suhu pengeringan terhadap aktivitas antioksidan kulit buah G. Mangostana. Pengeringan dilakukan pada suhu 30, 60 dan 900C selama 72 jam. Pengukuran aktivitas antioksidan dilakukan dengan metode DPPH dan hasil pengamatan dianalisis menggunakan ANOVA dua arah. Hasil penelitian menunjukkan bahwa suhu pengeringan berpengaruh nyata terhadap aktivitas antioksidan kulit buah G. Mangostana (P < 0,05). Kulit buah G. Mangostana perlakuan terbaik adalah pengeringan pada suhu 300C. Dengan nilai IC50 20,349 µg/mL.

Abstract

Vitamin C is a water-soluble antioxidant that affect the body immune system. Vitamin C deficiency is related to the high risk of infection disease manifestation. Consuming vitamin C of 100-200 mg/day is effective to optimize physiological functions of tissues and cells. Fruits are a natural source of vitamin C that can be consume to achieve vitamin C requirement intake, such as Ananas comocus. However, the vitamin C content in Ananas comocus can be varied depending on some factors, such as their varieties, growth environment, and preparation. This research aims to determine vitamin C content in some fresh Ananas comocus fruits from three plantations in Teluk Meranti area, Pelalawan using Spectrophotometry UV-Vis method, that is a suitable method for vitamin C content determination. The result shows that the calibration curve of vitamin C giving a good linearity, with r value of 0,9943. Vitamin C content in the three plantation was varied. The content from the plantation 1, 2, and 3 was 51,1889; 48,3320; dan 144,200 mg/100 g. Keywords: fresh fruit, spectrophotometer, vitamin C content determination, Teluk Meranti, Ananas comocus

Abstract

ABSTRACT Drying process is an important factor which is affect natural medicine products quality. The process influence secondary metabolite composition of natural medicines, such as polyphenol compounds, which manage their pharmacological activity. Polyphenol compound is an essential parameter of antioxidant effect of natural products which is degraded by high temperature. Thus, drying process at high temperature can damage total polyphenol content of natural medicine which decrease its antioxidant activity. This research aims to determine antioxidant activity of Pometia pinnata with variance of drying process. The samples was dried at 30, 60 and 90°C. Antioxidant activity then was measured using DPPH method. The result shows that the highest antioxidant activity is given by sample dried at 60C (IC50=49.3608), followed by 30 and 90C. Finally, it can be concluded that the drying process influence antioxidant activity of P. Pinnata leaf.   Keyword: drying, P. Pinnata, antioxidant, polyphenol, secondary metabolite, DPPH. ABSTRAK Proses pengeringan merupakan salah satu faktor yang mempengaruhi kualitas obat bahan alam. Proses pengeringan ini mempengaruhi komposisi kandungan metabolit sekunder dalam bahan, seperti senyawa polifenol, yang akhirnya mempengaruhi efek farmakologisnya. Senyawa polifenol merupakan salah satu parameter tinggi rendahnya aktivitas antioksidan suatu bahan alam yang sangat mudah rusak akibat pemanasan. Sehingga, pengeringan bahan pada suhu tinggi mampu merusak komposisi polifenol total suatu bahan alam yang akhirnya menurunkan aktivitas antioksidannya. Oleh karena itu, pemilihan suhu pengeringan pada proses pembuatan simplisia sangat penting untuk mempertahankan mutu bahan tersebut. Penelitian ini bertujuan untuk mengetahui pengaruh suhu pengeringan terhadap aktivitas antioksidan daun matoa (Pometia pinnata). Pengeringan dilakukan pada suhu 30, 60 dan 90°C. Aktivitas antioksidan diuji dengan metode DPPH menggunakan microplate reader. Hasil menunjukkan bahwa aktivitas antioksidan paling tinggi diberikan oleh bahan pada pengeringan 60°C (IC50=49,3608), dan diikuti oleh pengeringan suhu 30°C (IC50=64,8404) dan 90°C (IC50=68,2175). Selanjutnya, tidak ada perbedaan nyata nilai IC50 pada pengeringan suhu 30 dan 90°C. Hasil ini menunjukkan bahwa suhu peningkatan dan penurunan suhu dapat mempengaruhi aktivitas antioksidan daun P. pinnata yang kemungkinan disebabkan oleh perubahan komposisi polifenol totalnya.   Kata kunci: proses pengeringan, P. Abstract

Musa acuminata × balbisiana fruit peel is a waste that can be utilized to produce a biodegradable, non-toxic, and stable natural cellulose. Cellulose characteristics depend on their isolation process, such as delignification and hydrolysis process, and their natural sources. This research aims to determine the effect of different delignification and hydrolysis process on characteristic of isolated cellulose from Musa acuminata × balbisiana fruit peel. The isolation was conducted by different delignification process, which used sodium hydroxide with heating at 80° C for 20 and 30 minutes, and hydrolysis process using chloric acid and sulfuric acid. The isolated cellulose then was characterized using spectrophotometer FT-IR. The result showed that there were a decrease in absorption intensity, wave number shifts, and some missing peaks of the isolated cellulose by the delignification and hydrolysis process compared to the standard cellulose used.

Abstract

Bakteri, virus dan mycoplasma dapat menyebabkan pneumonia. Klebsiella pneumoniae adalah penyebab umum infeksi resisten antimikroba pada pasien rawat inap. Miroba ini secara alami resisten terhadap penisilin. Pnemoniae dapat disebabkan oleh bakteri, virus dan mycoplasma. Bakteri Klebsiella pneumoniae mampu menginfeksi dan memperparah pasien yang terkena penyakit COVID-19. Bakteri ini telah resisten terhadap antibiotik. Dilakukan metode molecular docking untuk memprediksi interaksi antara klebsiella pneumoniae dan senyawa bahan alam yang terdapat pada database andalas sitawa yaitu Alpha Mangostin, Andrographolide, Asiaticoside, Catechin. Curcumin, Deoxyelephantopin, Ethyl methoxycinnamate, Hydroxychavicol, Piperine dan Plumbagin. Perangkat lunak yang digunakan dalam simulasi yaitu Autodock Vina. Optimasi geometri dilakukan dengan MMFF94 untuk senyawa bahan alam. Dari hasil diperoleh senyawa yang memiliki energi bebas gibbs yang terbaik adalah Asiaticoside dengan energi -10.22 (kcal/mol). Asiaticoside memiliki interaksi pada asam amino yaitu Ikatan Hidrogen pada asam amino Asn79, Arg151 selanjutnya Ikatan Van der walls pada Ile152, Met156, dan Ser156. Ikatan Pi-Alkyl pada asam amino Tyr125, His195 dan ikatan Alkyl pada asam amino Arg155. Simulasi dinamika molekul senyawa Asitiacoside dengan protein fluktuasi terbesar pada residu asam amino 245-248 yaitu Leusine, Tyrosine, Phenylalanine, dan Glutamine dengan rentang fluktuasi 3,2169-3,2525 nm

Abstract

The trend of independent drug use behavior (self-medication) among Indonesian people, especially in West Sumatra Province, continues to increase. Without knowledge about the correct use of drugs, people will be at risk to drug use problems. Therefore, education on drug use needs to be implemented to increase public knowledge regarding drug use. By increasing their knowledge, the public will have rational drug use behavior. The aim of this community outreach is to increase public knowledge regarding the use of over-the-counter drugs and limited over-the-counter drugs in treating health problems. The method of activity carried out is education through face-to-face home visits. This activity was carried out in Nagari Sikucur, V Koto Kampung Dalam District, Padang Pariaman Regency. There were 30 community members or family representatives who were the targets of this drug use education and home visit. The implementation of this activity involved five teams consisting of pharmacy lecturers with pharmacists’ qualification, Master of Pharmacy students who had registered as pharmacists, and Bachelor of Pharmacy students. The activity started with an introduction, continued with a pre-test, providing education, post-test, and handing over self-medication drug packages to the community. The results of this home visit-based education show that the community's knowledge score regarding drugs is higher than before the education. It can be concluded that the objectives of this activity were achieved well. To achieve better activity results, preliminary studies, collaboration with stakeholders, time allocation, and adequate technical preparation of activities are required.
