Abstract

Background: Stichopus hermanni contains protein, glycosaminoglycans and EPA-DHA, which accelerate oral wound healing. Ethanol is solvent with universal nature and frequently used in the extraction. This study aims to know the characterization and potency of Stichopus hermanni in oral wound healing. Methods: Stichopus hermanni was dried by freeze-dry method and extracted using ethanol solvent. Characterization was examined the levels of protein, glycosaminoglycans and EPA-DHA. In vivo, male wistar rats made traumatic ulcer at the lower labial mucosa by using thermal burn. Thirty-two rats are divided into four groups (n=8), negative control group (K), positive control group (AH) was given hyaluronic acid gel 0.2%, and treatment groups were given Stichopus hermanni 60%-80% (SH60-SH80) gel. The ulcer formed was given gel for four days. The rat’s labial mucosa was measured. The difference between the first and fourth day is tabulated. Data were analyzed by Anova and Tukey-HSD tests. Results: The result of Spectrophotometry show the contents of protein (19.39), glycosaminoglycans (2.79–3.98). The result of in vivo showed a significant difference in ANOVA test (K=1.07; AH=1.27; SH60=1.89; SH80=1.66). In the Tukey-HSD test, the SH60 group shows a significant difference between K and AH groups. Conclusion: Stichopus hermanni ethanol extract gel contain protein and Glycosaminoglycans and potency for oral wound healing.

Abstract

Background: Tooth loss can be treated by using dentures. Heat cured acrylic resins are the the most common denture base materials but it has porosity that causing leftover food, plaque, bacteria, and Candida specially Candida albicans to adhere. Hydroxyapatite is chemical compound that can reduce residual monomer which causing porosity inside heat cured acrylic resin base denture. Purpose: to determine the effectivity of adding hydroxyapatite powder for reducing porosity of heat cured resins acrylic based denture. Methods: Sample were acrylic resin plate (20x10x4mm) divided into 4 groups, control group was acrylic resin without hydroxyapatite, and the treatment group was added with hydroxyapatite with concentration 2%, 5%, and 10%. After the polymerization are finished the sample was soaked on aquadest for 1 day and weighed using electronic weighing scale. The data tested with One Way ANOVA and followed by LSD test. Results: The average of porosity level on Control, treatment 1, treatment 2, treatment 3 group were 8,53%;7,75%,4,74%,5,55%. There are significant different in the porosity level between the control group and all treatment groups (p<0.05). Conclusion: Hydroxyapatite with concentration 2%, 5%, and 10% showing effectivity on reducing porosity level on heat cured acrylic resins base plate

Abstract

Background: The traumatic ulcer is the most common oral soft tissue lesion that cause physiologic dissorder (pain) in patients, so it need a medicine to improving healing processs.Stichopus hermanii contains glycosaminoglycans, omega-3 and other agents that accelerate wound healing. Their influence againts signaling cell for proliferation and angiogenic for healing procce. Endoglin plays in a number of cellu- lar processes and the elevated expression during wound healing. Purpose: Knowing the effectivity of ethanol extract of Stichopus hermanii towards the expression on traumatic ulcer maturation phase in oral mucosa. Materials and Methods: The research subjects were forty eight wistar rats divided into 6 groups. Traumatic ulcer was made in the center-rat’s-labial-mucosa. The X1 group was smeared with sterilized aquades as the control group. The X2 was smeared with 0,2% hialuronic acid, X3 with 60% ethanol extract, and X4 with 80% ethanol extract. They were smeared once a day during 4 days. On the sixth day, the rat’s labial mucosa was biopsied to make histological preparation in order to study the amount of fibroblast. Results: Kruskal-Wallis test showed not significant difference (p=0,000) after applying Stichopus hermanii extract for expression of endoglin in Conclusions: ethanol extractStichpus hermanii not effective for expression of endoglin on traumatic ulcer maturation phase in healing procces.

Abstract

Background: Bone healing on defect requiring the cells to form new bone. Fibroblast growth factor -2 belong to one of signaling control that play part in differentiation of fibroblast to osteoblast cell. Hydroxyapatite application on bone healing will aid bone regeneration as structure. Application of Sardinella longiceps oil that contain EPA and DHA as immunodulator could accelerate and optimize inflammatory phase in bone healing. Purpose: The aimed of this study to determine the effectivity of the combination of Sardinella longiceps oil and application of hydroxyapatite on the expression of fibroblast growth factor -2 (FGF-2) in bone healing process. Materials and Methods: The experimental units (male wistar rat, n=20) were devided into two groups, control (K) and treathment (P) groups. Treathment groups were given Sardinella longiceps oil per oral seven days before the surgery to make the defect, and continued for seven days later. Control groups were not given Sardinella longiceps at all. All of the wistar rat were have two defects in their right femur, which is use hydroxyapatite (K+ and P2) and no use hydroxyapatite (K- and P1). Defect was made as big as half diameter of round bur in femur. Rats were sacrificed seven days after surgery, and femur were sliced transversally and performed immunohistochemistry staining with Anti-bFGF Polyclonal antibody (Bioss®). Examination were done under light microscope. Data were analyzed using one-way Anova for FGF-2 expression. Result: Sardinella longiceps oil increased the mean of FGF-2 expression on P2 group : 15,29±5,251, compare to K- : 5,57±2,070 , K+ : 12,14±3,976 and P1 : 8,14±1,676 (P<0.05). Conclusion: The combination of Sardinella longiceps oil and hydroxyapatite application effective the expression of fibroblast growth factor-2 in the process of bone healing.

Abstract

Background: In dentistry, complication post-extraction, disease periodontal, prosthetic surgery, pathological conditions, and trauma can cause alveolar bone loss and even lead to tooth loss. The use of hydroxyapatite derived from blood cockle shells is osteokondiction. Omega-3 in sardine oil will stimulate the healing process of damaged bone with decreasing in the number of the osteoclasts. Purpose: The aimed of this study to determine the Effectivity of Grafting Combination Blood Cockle Shell and Sardine Fish Oil to Decrease in The Numberof Osteoclasts in The Process of Bone Repair Methods: Animals used in research, as many as24 male Wistar rats, which then divided into 4 groups, namely the control group and the treatment group. The control group subdivided into two groups, normal healing and human application's paste. In the treatment group, also divided into two groups, group with blood cockle shell application and combination of blood cockle shell and sardine oil. Osteoclasts viewed using HPA on the 7th day. Results: Decrease in The Number of Osteoclasts on K- group : 2.67±1,033, P1 : 2.33±0.09, K+ : 1.5±0.09, P2 : 0.83±0.54. Because the data not homogeneous then used test Kruskal-Wallis test and Mann-Whitney. P1 with P2 have a difference significantly compared 0,048 , P2 with K- 0,026 (p<0,05). Conclusions: Combined administration of blood cockle shell (Anadara granosa) and sardine oil (Sardinella longiceps) significantly influence the amount of osteoclasts in the process of bone repair on the 7th day.
