Abstract

Jambolan is a rare local fruit requiring an effective propagation technique to mantain its sustainability as vegetative propagation by cuttings. This study aims to determine the effect of plant growth regulator types and its soaking duration on jambolan cuttings growth. The study was conducted in Experimental Garden I and Plant Physiology Laboratory, Agriculture Faculty of Syiah Kuala University started from July to November 2021. The study used a 4 x 3 factorial completely randomized design. The factors studied were PGR types and soaking duration. The best jambolan cuttings growth caused by PGR was found in 40 gL-1 NAA treatment. The best jambolan cuttings growth caused by soaking duration was found in 10 hours soaking duration treatment. The best jambolan cuttings shoots growth was found in combination of 100% coconut water and 10 hours soaking duration treatment. Abstract

Tujuan pada penelitian ini untuk mengetahui pengaruh konsentrasi BAP dan IAA dan  interaksi kedua faktor tersebut terhadap multiplikasi tunas pisang barangan merah secara in vitro. Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan Tanaman Fakultas Pertanian Universitas Syiah Kuala, Darussalam, Banda Aceh. Penelitian ini berlangsung dari bulan April sampai Agustus 2021. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) pola Faktorial 4 x 4, yang terdiri dari 2 faktor. Faktor pertama adalah konsentrasi BAP dan faktor kedua adalah konsentrasi IAA. Faktor pertama ialah konsentrasi BAP, yang terdiri dari 4 taraf: kontrol, 2, 4, dan 6 mg L-1. Faktor kedua ialah konsentrasi IAA, yang terdiri dari 4 taraf: kontrol, 0,5,  1, dan 1,5 mg L-1. Pengamatan meliputi persentase tumbuh tunas, jumlah tunas, dan tinggi tunas. Hasil penelitian menyatakan bahwa perlakuan BAP berpengaruh terhadap parameter persentase tumbuh tunas, jumlah tunas dan tinggi tunas. Persentase tumbuh tunas terbaik (100%) dijumpai pada konsentrasi 2 mg L-1 BAP dan 4 mg L-1 BAP. Jumlah tunas terbanyak (1,7 tunas) pada konsentrasi 2 mg L-1 BAP dan 4 mg L-1 BAP. Tinggi tunas tertinggi (3,2 cm) dijumpai pada konsentrasi 4 mg L-1 BAP. Perlakuan IAA berpengaruh terhadap parameter persentase tumbuh tunas, jumlah tunas dan tinggi tunas. Semua parameter tersebut memiliki rerata terbaik yaitu pada perlakuan kontrol. Persentase tumbuh tunas terbaik yaitu dengan rerata 58,3 %. Jumlah tunas terbanyak yaitu dengan rerata sebanyak 1 tunas. Tinggi tunas tertinggi yaitu dengan rerata 1,6 cm. Shoot Multiplication of Banana (Musa acuminata Colla.) cv. Barangan Merah in Various Concentrations of Benzyl Amino Purine (BAP) and Indole Acetic Acid (IAA) under In Vitro Condition This study aimed to determine the effect of concentrations of BAP and IAA with the interaction between two factors on the multiplication of barangan merah banana shoots in vitro. The research was conducted at the Plant Tissue Culture Laboratory, Faculty of Agriculture, Syiah Kuala University, Darussalam, Banda Aceh. This research from April to August 2021. This study used a completely randomized design (CRD) with a 4 x 4 factorial pattern, which consisted of 2 factors. The first factor is the concentration of BAP and the second factor is the concentration of IAA. The first factor was BAP concentration which consisted of 4 levels: control, 2, 4, and 6 mg L-1. The second factor was the concentration of IAA which consisted of 4 levels: control, 0.5, 1, and 1.5 mg L-1. The observed parameters including shoot growth rate, number of shoots, and shoot height. The results showed that BAP treatment had an effect on the parameters of shoot growth rate, number of shoots, and shoot height. The best shoot growth rate (100%) was found at a concentration of 2 mg L-1 BAP and 4 mg L-1  BAP. The highest number of shoots (1.7 shoots) at a concentration of 2 mg L-1 BAP and 4 mg L-1  BAP. The highest shoot height (3.2 cm) was found at a concentration of 4 mg L-1 BAP. The IAA treatment affected the parameters of shoot growth rate, number of shoots, and shoot height. All of these parameters have the best average, namely the control treatment. The best shoot growth rate is with an average of 58.3%. The highest number of shoots is with an average of 1 shoot. The highest shoot height was with an average of 1.6 cm.

Abstract

Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan Tanaman Fakultas Pertanian Universitas Syiah Kuala, Darussalam, Banda Aceh.Penelitian berlangsung dari bulan April sampai Juli 2021.  Penelitian ini terdiri dari 2 faktor, faktor pertama adalah konsentrasi BAP yang terdiri dari 5 taraf yaitu kontrol,  0,25 mg L-1 , 0,5 mg L-1, 0,75 mg L-1, dan 1 mg L-1. Faktor kedua adalah konsentrasi NAA yang terdiri dari 4 taraf yaitu kontrol, 0,25 mg L-1 , 0,5 mg L-1, dan 0,75 mg L-1, sehingga terdapat 20 kombinasi perlakuan. Hasil penelitian menunjukkankonsentrasi BAP dan NAA berpengaruh pada persentase pembentukan kalus terbaik (100%) dijumpai pada konsentrasi 0,25 mg L-1  NAA  dan 0,75 mg L-1  NAA.  Persentase jumlah akar dengan konsentrasi 0,75 mg L-1 NAA menghasilkan jumlah akar yang  lebih banyak yaitu sebesar 7,8 akar, dibandingkan dengan perlakuan lainnya. Induction of Patchouli Callus (Pogostemon cablin Benth,) using Benzyl Amino Purine and Naphtalene Acetic Acid under In Vitro Condition The purpose of this study was to determine the effect of BAP and NAA concentrations and the interaction between these two factors on the growth of explant shoots. patchouli shoots of the Tapak Tuan variety in vitro. This research was conducted at the Plant Tissue Culture Laboratory, Faculty of Agriculture, Syiah Kuala University, Darussalam, Banda Aceh. The study took place from April to July 2021. This study consisted of 2 factors, the first factor was the concentration of BAP which consisted of 5 levels, namely control, 0.25 mg L -1, 0.5 mg L -1, 0.75 mg L - 1, and 1 mg L -1. The second factor wasthe concentration of NAA which consisted of 4 levels, namely control, 0.25 mg L -1, 0.5 mg L -1, and 0.75 mgL-1, so there were 20 treatment combinations. The results showed the combination of BAP and NAA had an effect on the proportion of callus formation, the best percentage of callus formation (100%) was found atoncentrations of 0.25 mg L -1 NAA and 0.75 mg L -1 NAA. The percentage of the number of roots with a concentration of 0.75 mg L-1 NAA resulted in a larger number of roots, which was 7,8 roots, compared to other treatments.

Abstract

Abstrak. Pisang raja adalah salah satu jenis pisang unggul dan diminati masyarakat karena dapat dimakan baik segar maupun dalam bentuk olahan dengan rasa dan aroma yang khas. Produksi pisang raja mengalami penurunan pada beberapa tahun terakhir. Solusi untuk mendapatkan persediaan bibit dalam jumlah banyak yaitu dengan cara kultur jaringan. Penelitian ini bertujuan untuk memperoleh konsentrasi NAA dan Kinetin yang tepat serta interaksi antara keduanya terhadap pertumbuhan tunas pisang raja secara kultur jaringan. Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan Fakultas Pertanian Universitas Syiah Kuala pada bulan April hingga Agustus 2018. Penelitian ini menggunakan Rancangan Acak Lengkap Faktorial 3x3 dengan 2 faktor perlakuan. Faktor pertama yaitu konsentrasi NAA (3 taraf yaitu 1, 2 dan 3 ppm). Faktor kedua yaitu konsentrasi Kinetin (3 taraf yaitu 2,5, 5 dan 7,5 ppm) yang diulang sebanyak 3 kali. Hasil penelitian menunjukkan persentase hidup eksplan tertinggi yaitu pada kombinasi NAA 1 ppm dan kinetin 5 ppm. Persentase eksplan hidup terendah yaitu pada kombinasi NAA 1 ppm dan kinetin 2,5 ppm, NAA 2 ppm dan kinetin 2,5 ppm, NAA 3 ppm dan kinetin 2,5 ppm dan NAA 3 ppm dan kinetin 5 ppm.Effectiveness of NAA and Kinetin Concentration on Shoot Growth of Raja Banana (Musa paradisiaca. L) by In VitroAbstract. Banana plantain is one of the superior and popular species of bananas because it can be eaten both fresh and in processed form with a distinctive taste and aroma. The production of plantain has experienced a decline in the last few years. Solution to get a large number of seedlings by tissue culture. This study aimed to obtain the concentration of NAA and Kinetin is right and the interaction between them on the basis of plantain shoot growth tissue culture . This research was carried out in the Tissue Culture Laboratory of Agriculture Faculty, Syiah Kuala University in April to August 2018. This study used  a Completely Randomized 3x3 Factorial Design with 2 treatment factors. The first factor is NAA concentration ( 3 levels, namely 1, 2 and 3 ppm ) . The second factor is the Kinetin concentration ( 3 levels, 2.5 , 5 and 7.5 ppm ) repeated 3 times . The results showed the highest percentage of explant life was in the combination of NAA 1 ppm and kinetin 5 ppm. The percentage of the lowest live explant is 1 ppm NAA combination and 2.5 ppm kinetin , 2 ppm NAA and 2.5 ppm kinetin , 3 ppm NAA and 2.5 ppm kinetin and 3 ppm NA A and 5 ppm kinetin.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi giberelin dan dosis pupuk kalium, serta interaksi keduanya terhadap pertumbuhan dan hasil tanaman mentimun varietas Mercy F1 secara partenokarpi. Penelitian ini dilaksanakan di lahan kebun Gampong Ilie-Ulee Kareng-Banda Aceh, Laboratorium Fisiologi Tumbuhan dan Laboratorium Hortikultura Fakultas Pertanian Universitas Syiah Kuala. Rancangan yang digunakan adalah Rancangan Acak Kelompok pola faktorial 4 x 3 dengan 3 ulangan. Faktor yang diteliti yaitu konsentrasi giberelin (Kontrol, 100, 200 dan 300 ppm) dan dosis pupuk kalium (Kontrol, 100 dan 200 kg ha-1). Hasil penelitian menunjukkan bahwa konsentrasi giberelin berpengaruh sangat nyata terhadap jumlah biji per buah dan bobot buah per tanaman. Hasil mentimun terbaik dijumpai pada konsentrasi giberelin 300 ppm dalam menurunkan jumlah biji per buah. Hasil penelitian juga menunjukkan bahwa dosis pupuk kalium berpengaruh sangat nyata terhadap bobot buah per tanaman dan bobot buah per buah, berpengaruh nyata terhadap jumlah buah per tanaman dan panjang buah per buah. Hasil mentimun terbaik dijumpai pada dosis 200 kg ha-1 pupuk kalium dalam meningkatkan bobot buah per buah dan panjang buah per buah. Interaksi antara konsentrasi giberelin dan dosis pupuk kalium berpengaruh nyata terhadap jumlah buah per tanaman dan jumlah biji per buah. Hasil mentimun terbaik dijumpai pada kombinasi konsentrasi giberelin 300 ppm dengan dosis pupuk kalium 100 kg ha-1.(Parthenocarp Fruit Initiation of Cucumber (Cucumis sativus L.) Mercy F1 Variety due to the Concentration of Gibberellins and the Dose of Potassium Fertilizer)Abstract. This study aims to determine the effect of gibberellins concentration and potassium fertilizer dosage, as well as their interaction on the growth and yield of Mercy F1 cucumber plant by parthenocarpy. This research was carried out in the gardens of Gampong Ilie-Ulee Kareng-Banda Aceh, Plant Physiology Laboratory and Horticultural Laboratory, Faculty of Agriculture, Syiah Kuala University. The design used was a Randomized Factorial Block Design 3 x 4 with 3 replications. The factors observed were the concentration of gibberellins (Control, 100, 200 and 300 ppm) and the dose of potassium fertilizer (Control, 100 and 200 kg ha-1). The results showed that the concentration of gibberellins had a very significant effect on the number of seeds by fruit and fruit weight by plant. The best cucumber yields were found at a concentration of 300 ppm gibberellins in reducing the number of seeds by fruit. The results also showed that the dose of potassium fertilizer had a very significant effect on fruit weight by plant and fruit weight by fruit, significantly affected the number of fruits by plant and fruit length by fruit. The best cucumber yield was found at a dose of 200 kg ha-1 of potassium fertilizer in increasing fruit weight by fruit and fruit length per fruit. The interaction between the concentration of gibberellins and the dose of potassium fertilizer significantly affected the number of fruits by plant and the number of seeds by fruit. The best cucumber yields were found in a combination of 300 ppm gibberellin concentration with a dose of 100 kg ha-1 potassium fertilizer.
