Abstract

Background: World Health Organization (WHO) estimates that the incidence of leptospirosis was ≥ 500.000 cases annually with worldwide distribution. The Case Fatality Rate (CFR) of leptospirosis in Indonesia in 2004-2012 reached > 5%. Leptospirosis in Banyumas discovered since 2010 and until May 2014 found two suspected leptospirosis in Beji Village District of Kedung Banteng and Kedung Pring village District of Kemranjen. The purpose of this study was to identify the reservoir of leptospirosis. Method: The study was cross-sectional. The Location conducted in Kedung Pring village, Kemranjen sub district and Beji village, Kedung Banteng sub district in Banyumas. Result: The species that found in the location were Rattus tanezumi and Suncus murinus, but in Beji village also founded the Mus musculus. The trap success in each location was 15% for Beji Village kedung Banteng sub district and 14.14% for Kedung Pring village, Kemranjen sub district. Concusion: All of species that found in the study are known potentially to transmit leptospirosis.                                                                                                                                       Keywords: Leptospirosis, Banyumas, Rats

Abstract

Leptospirosis in Sukoharjo was discovered in 2014. Examination with RDT (Rapid Diagnostic Test) leptotek supported by clinical symptoms showed 6 positive cases of Leptospira, and until March 2015 one person was found to be Leptospira positive. The aim of this study was to identify rats as the main reservoir of leptospirosis, calculate the catching rate of rats and to detect the presence of pathogenic Leptospira in rats. This study was a cross-sectional survey, and was conducted in Pabelan village Kartasura Sukoharjo on May 2015. Polymerase Chain Reaction (PCR) assay was conducted in Bacteriology Laboratory Balai Litbang P2B2 Banjarnegara to detect leptospira in the kidney of the rats. Data were analyzed descriptively. Results of rats and shrew catching obtained Rattus tanezumi, Rattus norvegicus and Suncus murinus. The species most commonly found was balanced between R. tanezumi and S.murinus (46%). The trap success rate inside and outside the house are 1.50% and 5%, respectively. Result of laboratory test showed from 13 rats kidneys, two kidneys were found to be Leptospira positive and was from R.tanezumi and R.norvegicus.

Abstract

Curcumin (1,7-bis(3-methoxyphenyl 4'hidroksi) -1.6 heptadien, 3,5-dione) is the yellow pigment of Curcuma longa. Curcumin has various biological activities such as antioxidant, antibacterial, antifungal, antiprotozoal and antivirus. Various benefits of curcumin can not be separated from the weakness which is not stable to pH and light. Pentagamavunon-0 (PGV-0) were made by changing the β diketone group on cluster analog of curcumin into monoketon while eliminating active methylene group so it is more stable to pH and light. PGV-0 also has potential as a stronger antioxidant and antiinflammatory agent than other curcumin analogues. The objective of this research was to determine the safe concentrations of curcumin and PGV-0 on vero cells due to the increased use of the two compounds through the cytotoxic test. This research includes experimental research. Cytotoxic test performed with microculture tetrazolium technique (MTT) method. Use of MTT to evaluate the cytotoxic is based on changes of tetrazolium salt into formazan crystals by mitochondrial enzyme succinate dehydrogenase with the help of cellular NADH. The results showed that the safe concentrations of curcumin and PGV-0 on vero cells respectively are 6.25 and 1.5625 ppm. Based on the cytotoxic test the secure concentration of curcumin was higher than PGV-0.
