Abstract

The aim of this study was determine the effect of temperature shock on hatching capability of the catfish eggs and its larval survivals.The study was conducted at the Board of Freshwater Aquaculture (BBAT ) in Tatelu  Dimembe District, North Minahasa Regency, North Sulawesi Province. Twelve tanks were set in this experiment. Two hundred (200) eggs were put in each tank with controlled temperature of 25 C, 27 C , 29 C , and 32 ºC. Each temperature was replicated 3 times. The experimental design used in data analysis was completely randomized design (CRD).  The results showed that the treatment temperature showed significant effect on  hatching of  eggs, while the difference in temperature had no effect on the survival rate of catfish larvae.   Keywords:  Temperature shock, eggs, hatching capability, survival rate,  larvae, Clarias gariepinus.

Abstract

This research intended to evaluate the area suitability of  Scylla spp at Likupang II Village, North Minahasa. The research was done descriptively by observing water quality, area availability. SWOT analysis was used to determine the development strategy.   Data were collected by interview, observation and questionnaire.  Research result showed that the potential of Likupang II Village was suitable for Scylla spp culture.  SWOT analysis showed internal factors affected the development of Scylla spp culture.  The strength were location, water quality, government support; the weakness were seed availability, opportunity, government policy and treating were season and safety.   Keywords : Scylla spp, SWOT analysis, , Likupang II, area suitability

Abstract

The research intends to determine the optimal concentration of honey addition in sperm dilution on sperm motility, fertilization and eggs hatchability of Nile Tolapia (Oreochromis niloticus). Experimental design used was a Complete Randomized Design (CRD) with three times repetition of four treatments. The concentration of honey addition used for treatment are A (0 ml of honey in 100 ml of physiological NaCl), B (0.60 ml of honey in 99.40 ml of physiological NaCl), C (0.65 ml of honey in 99.35 ml of physiological NaCl), D (0.70 ml of honey in 99.30 ml of physiological NaCl). Parent used consisted of five female parent and five male parent with 250 – 500 mg weight. Sperm dilution containers used was 12 plastic cups and the container fertilization and hatching eggs container used was 12 brass plates.  Spermatozoa motility observation performed immediately after dilution process. Fertilization observation was performed after 12 hours of fertilization process, and Egg Hatchability observation was performed after 60 hours of fertilization.  The observation results showed that the addition of honey in sperm dilution contributed significant effect on spermatozoa motility, fertilization and eggs hatchability of Nile Tolapia (Oreochromis niloticus). This observation showed that the addition of 0.70 ml of honey in a sperm dilution was the best treatment seen from the average percentage of spermatozoa motility (73,33 %), fertilization (80,00%) and eggs hatchability (77,33 %.) Abstract

This study aimed to 1) determine the best combination between ketapang and banana peels for the propagation of life feed mosquito larvae; 2) determine the culture media that can produce mosquito larvae optimally. The containers used in this study were 24 buckets with a size of 5 litres. Some of the materials used in the study were dried ketapang leaves, goroho banana peels, kepok banana peels and water. This study used a factorial Completely Randomized Design (CRD) consisting of two factors and three replications. The first factor was the difference in the number of ketapang leaves. The second factor was the difference in banana peels types. The first factor included the difference in the number of ketapang leaves, consisting of: Treatment 0 = without ketapang leaves, Treatment 1 = 50 grams of ketapang leaves, Treatment 2 = 75 grams of ketapang leaves, Treatment 3 = 100 grams of ketapang leaves. The second factor included differences in banana peels, which consist of: Treatment 1 = one kepok banana peel, Treatment 2 = one goroho banana peel. The results were different types of banana peels affected the numbers of mosquito larvae, where the best was kepok banana peels; the difference in weight of ketapang leaves did not have a significant effect on the number of mosquito larvae. The container used in this study was a bucket with a volume of 5 L. The banana peels used were goroho banana peels and ripe yellow kapok banana peels. Ketapang leaves were preapared with a weight 0f 0, 50, 75, and 100 grams, respectively. Shoe banana peels and  goroho banana, one skin eachand ketapang leaves 0, 50, 75, and 100 grams, respectively, put together wit the jalah used to wrap between ketapang leavesand spatu banana peels and goroho banana, then put into a container that has been filled wit 3 L of water.

Abstract

This study aimed to analyze the survival rate of freshwater crayfish Cherax quadricarinatus, using different natural feeds, namely Alona sp, Daphnia sp. and Artemia sp. Observing the growth of juvenile crayfish C. quadricarinatus. The study was conducted for two months (April - May 2019), at the Aquaculture Technology Laboratory. In this study, there were three treatments, namely different types of natural food. The treatments given were: Treat A Alona sp .; Treatment B Daphnia sp; and Treatment C Artemia sp. Feeding 25 individuals during the maintenance period is carried out twice a day, namely in the morning at 08.00 - 09.00 and 17.00 - 18.00 WIB. This study used a completely randomized design showing that the average value of SR (survival) in each treatment such as treatment A (Alona sp) was 60%, then treatment B (Daphnia sp) was 66.67% and finally treatment C ( Artemia sp) of 66.67%. Observation results for 21 days of mainten ance found an increase in length between 2 - 4 mm. The analysis of variance (ANOVA) showed that the differences in the types of natural food did not have a significant effect (P> 0.05) on the survival and growth of juvenile crayfish kept for 21 days

Abstract

This study aimed to examine the best concentration of nutmeg oil for anesthesia of freshwater crayfish (Cherax quadricarinatus) seeds and analyze the length of time to faint, the length of time to faint and the length of time to wake up for the seeds of freshwater crayfish (C. quadricarinatus) . This research was carried out at the Aquaculture Technology Laboratory, Faculty of Fisheries and Marine Sciences, Sam Ratulangi University, Manado. The time of the research was carried out from April 18, 2021 - May 18, 2021. This study used a completely randomized design (CRD) consisting of 5 treatments and 3 replications, thus there were 15 experimental units. Treatment A with a dose of nutmeg oil (0.7 mL), treatment B (1 ml), treatment C (1.3 mL), treatment D (1.6 mL) and treatment E (1.9 mL). Parameters observed during behavioral research, length of time to faint, length of time to faint and length of time to wake up. The results of the study using the ANOVA statistical analysis test and the BNT follow-up test (Least Significant Difference) showed that different doses of nutmeg oil had a very significant effect on the length of time to faint, the length of time to faint and the length of time to wake up. The length of time to pass out the fastest, the longest length of time unconscious and conscious longest duration was obtained in treatment E (1.9 mL).

Abstract

The purpose of this study were (1) to determine the morphometric characteristic of pearl oyster Pinctada margaritifera in Arakan, Mantehage Bango, and Talengen waters of North Sulawesi Province, (2) to determine the ideal location for the feasibility of pearl oyster culture. Determination of the sampling location is done by purposive sampling. The shell morphometric dimensions measured were shell length, shell height, hinge length, shell thickness and shell weight. The measured water quality were temperature, salinity, dissolved oxygen, pH, and depth. The morphometric characteristic of P. margaritifera oyster in Arakan waters have shell length (PC) ranging from 44,25 – 169,77 mm with an average of 86,50 mm, Mantehage Bango waters  have PC ranged from 38,73 – 106,09 mm with an average of 80,31 mm, and Talengen waters have PC ranged from 25,89 mm to 105,34 mm with an average of 68,97 mm. Arakan waters are dominated by medium size P. margaritifera (53,78%), it is suspected that Arakan waters have pearl oyster stocks in nature for aquaculture activities. Evaluation of the suitability of waters for pearl oyster culture P. margaritifera in Arakan waters scored 77,7% categorized as suitable (S2), the location of Mantehage Bango waters scored 80% categorized as suitable (S2) and the location of Talengen waters 68,8% categorized as conditional (S3).

Abstract

The research was carried out to investigate the best amount of large freshwater snail (Filopaludina javanica) meat as feed for the growth and survivals (SR) of freshwater lobster C. quadricarinatus.  The size of freshwater lobster used in this study was 1.70 – 23.90 gr.  Lobsters were kept in aquariums measuring 30 x 30 x 40 cm for 30 days. The experimental design used was a completely randomized design (CRD) with three treatments and three replications that is A (4%), B (7%) and C (10%) of feed. Data analysis was used linier regression. The results showed the (R2) marks close to 1 means that the time of experiment effects the growth of lobster in all treatments, 4% feed (R2 =0,8919), 7% (R2=0,9942) and 10%(R2=0,9484, where the highest coefficient regression was treatment B (0,5818) followed by treatment A (0,4625) and C (0,42), respectively.  The highest survival rate was found in treatment B (91.7%), treatment A (75%) and C (66.7%), respectively.  Results of analysis showed that the size and the length of time of experiment effects the time of growth and survival.

Abstract

Disease control using medicinal plants is an alternative to antibiotics and chemicals in aquaculture. One type of plant with the potential for health management is Polyscias fruticosa or sea kedondong which contains flavonoids, saponins, methanol, and tannins that are efficacious as antihistamines, antioxidants, antivirals, antibacterials, anti-inflammatories, and anti-cancer. The aim of the study was to examine the effect of kedondong laut leaf extract on increasing the phagocytic activity of tilapia (Oreochromis niloticus), and to determine the appropriate concentration of kedondong laut leaf extract to increase the phagocytic activity of tilapia (O. niloticus) seeds. The leaves of kedondong laut are cleaned, washed, and dried for 14 days at room temperature and then ground using a blender to obtain an extract of coarse flour. The flour was extracted using the maceration method with 70% alcohol solvent with a ratio of coarse flour and the solvent 1:5. After 24 hours of soaking, the extract was filtered using Whatman paper No. 42, and the dregs were soaked again with the same volume of solvent for the same soaking time and filtered again. This process is done 3 times until the extract looks clear. The extract was then concentrated using a rotary evaporator to obtain a concentrated extract of about 30% of the initial total extract. The concentrated extract is then sprayed evenly using a sprayer into the feed according to the set treatment dose. The research design used was Completely Randomized with 5 treatments, each with 3 replications. The treatments used were A: 0%, B: 1%, C: 2%, D: 3%, and E: 4% extract/kg feed. Treatment feed was given for 4 weeks at a dose of 5%/body weight/day, and the frequency of feeding was 2 times a day at 08:00 and 16:00. The results showed that the administration of sea kedongong leaf extract had a very significant effect on the phagocytic activity of tilapia (p<0.01). The best results were achieved in fish treated with B (1% extract/kg) for 28 days. Keywords: medicinal plants, maceration, immune modulators, phagocytosis activity, disease control
