Abstract

In order to screen natural antioxidant, the research about antioxidant of some Euphobiaceae herbs, have been conducted. The air-dried herbs  of Euphorbia heterophylla L, Phyllanthus acidus (L.) Skeels, and Phyllanthus buxifolius Muell Arg were extracted with metanol. The obtained extract was concentrated and then suspended to produce n-hexane, ethyl acetat and aqueous fractions. Free radical scavenger activity against DPPH (1,1-diphenyl-2-pycrylhydrazyl) measured by spectrophotometric method  and the IC50 value was determined. The compounds of  active fraction had been identified by TLC method. All of the herbs showed activity as DPPH scavenger. Among these herbs, Euphorbia heterophylla L. and Phyllanthus buxifolius Muell, Arg. exhibited a strong free radical scavenging of ethyl acetat fraction with IC50 value 5,88 µg/ml and 4,64 µg/ml. The result of TLC by mobile phase n-buthanol-acetic acid-water (4:1:5) and acetic acid 15% showed flavonoid compound.Keywords: Euphorbiaceae herbs, antioxidant, DPPH, flavonoid

Abstract

Kemuning (Murraya paniculata (L.) Jack) is one of Indonesia medicine plants. Kemuning leaves can be used to decrease cholesterol. The experiment was aimed to make tablets of kemuning leaves extract with gelatin as binding agent that fulfilled the requirements of tablet’s physical quality. Extract of kemuning leaves was obtained from kemuning leaves powder by maceration method using ethanol 70%. The obtained extract was soaked for several days and then evaporated till thick extract was obtained. Kemuning leaves extract tablets were made in 3 formulation with difference concentrations of binding agent, i.e. formula I contained 1% of gelatin mucilage 5%, formula II contained 2% and formula III contained 3%. The method used to make tablets of kemuning leaves extract was wet granulation method. The obtained granules were physically tested in cluding flowing time, the angle of repose, and loss on drying. The obtained tablets were physically tested including weight homogeneity, hardness, friability and disintegration time. The obtained data was statistically analyzed using SPSS 11.0 for windows program i.e. one way anova added with one factor at 95% convidence. The conclusion of the experiment was that kemuning leaves extract could be made tablets that fulfilled therequirement of tablet’s physical quality. The result of the experiment showed that the three formulations produced tablets with weight homogeneity, hardness, friability and disintegration time that fulfilled the requirement of good physical quality of tablet.

Abstract

Antioxidant is very important to keep human’s health because of it can scavenge free radical. Flavonoid is usual natural antioxidant got on plants. Euphorbia heterophylla L. which is used by people as traditional herbal medicine and it has traditional name kate mas is known contain of flavonoid, tannin and saponin. The aim of the experiment was to find out the antioxidant activity of kate mas (Euphorbia heterophylla L.) herb and the potency of methanolic extract, hexane, etil acetat and water fractions, and also of kate mas herb on DPPH (2,2-difenil-1-pikrilhidrazil) radical with IC50 parameter. Kate mas herb were extracted by maceration method using methanol. The obtained extract was partitioned with water and hexane, and then with etil acetat. The antioxidant activity of the obtained fraction was tested against DPPH radical. The test was conducted in 4 series of concentrations by adding 4.0 ml test solutions with 1.0 ml DPPH 0.45 mM. The radicals scavenging activity was measured with spectrophotometer at 515 nm wavelength and determined the IC50 value. The experiment used rutin as positive control. The result of the experiment showed that kate mas herb had antioxidant activity with IC50 of methanolic extract, water fraction, n-hexane fraction, and etil acetat fraction: 44,43 ppm; 130,63 ppm; 212,81 ppm; and 5,88 ppm respectively. The etil acetat fraction has the strongest activity compairing to the other with the IC50 is almost near often rutin’s IC50 is 5,11 ppm as the positif control.

Abstract

The aim of this experiment was to find out the effect of Bandotan (Ageratum conyzoides, L) herbs ethanol extract that given orally on decreasing bleeding and blood coagulation time in white male rats (Rattus norvegicus) wistar strain. Bandotan herbs were extracted by soxhlet using 70% ethanol as solvent. The animals test were divided in five groups, each group consist of 5 mice. Group I was given dose 69,58 mg, group II was given dose 104,37 mg, and group III was given dose 139,16 mg/200 g BW. Group IV as positive control was given tranexamic acid 4,5 mg/200 g BW. Group V as negative control was given CMC 1%. The animals test were given sub cutaneous heparin injection for 5 days to induce bleeding and inhibit blood coagulation, and then were given test preparation for 6 days and the time of bleeding ceasing and blood coagulation were recorded once every 2 days during test preparation was given. \The statistic results of the study showed that ethanol extract of goat weed herbs given orally could decrease bleeding time and blood coagulation in white male rats. According to the result of two ways anova statistical test, ethanol extract of goat weed herbs with dose of 104,37 mg/200 g BW is effectivest in decreased bleeding and blood coagulation time with ability to stop bleeding time 85,45% and blood coagulation time 70%.

Abstract

Seligi (Phyllanthus buxifolius (BI.) M.A.) leaves contain flavonoid, polifenol, and saponin that have antioxidant activity and thought to potentially as antihiperlipidemic. This research was aimed to find out the effect of seligi leaves ethanol extract to increase HDL level and decrease LDL level in white rats. The subject of the research was 30 male white rats in 150-200 gram body weight and 2-3 months age. Rats were divided into 6 groups randomly then were given BR II and water everyday. Group I, II, and III were normal control group, the positive control (simvastatin), and negative control respectively. The treatment group was given seligi leaves extract in doses of 75; 150; and 300 mg/kg bw/day. The tested animal were given cow fat and yolk for two weeks up to hypercholesterolemia state, tested preparation was given on day-14 until the day-28. The HDL and LDL levels were measured on the day-0, 14, and 28 by CHOD-PAP method. The result of researchshowed that seligi leaves ethanol extract orally administration could increase the HDL and decrease the LDL levels of blood serum. Seligi leaf ethanol extract with dose 300 mg/kg bw/day had equal effectiveness to simvastatin.
