Abstract

Biodiversity of macrozoobenthos in Pasir Island, Brebes, is under-explored. On the other hand, the baseline information regarding macrozoobenthos is essentially required for better future environmental management. This research aims to analyze macrozoobenthos community structure in the Pasir Island that has not been documented. Surveys were carried out in the rainy season. In this study, a total of 10 sampling locations were classified into two different groups that represent “control” and “disturbed” station. The main structural parameters of the macrozoobenthos identified at each station were specific richness (S), abundance (number of individuals m-2, N) and the Pielou’s index (J). Additionally, the pairwise comparison between groups was conducted by using analysis of similarity (ANOSIM) and the similarity percentage (SIMPER) to obtain the description of community structure. A total of 1054 specimens were counted from the “control” and “disturbed” stations. These specimens belonged to five animal classes, namely, Bivalvia, Gastropoda, Malacostraca, Cephalopoda and Echinoidea. ANOSIM and SIMPER analysis demonstrated that a significant difference was found between “control“ and  “disturbed“ stations with the percentage of dissimilarity value of 88.82%. In addition, environmental variables such pH, salinity and type of substrates contributed significantly to this difference. This result provides insight regarding macrozoobenthos diversity and ecological information that may contribute to further conservation management in the Pasir Island, Indonesia.

Abstract

Analisis diversitas diatom yang berasosiasi dengan makroalga dapat dimanfaatkan sebagai bahan referensi dalam kegiatan pengelolaan dan konservasi wilayah pesisir. Data yang dihasilkan juga berguna untuk membaca kualitas lingkungan dan bioprospeksi. Penelitian ini bertujuan untuk melihat diversitas komunitas diatom yang berasosiasi dengan Padina sp di Pulau Pramuka dan Semak Daun. Kedua pulau memiliki karakteristik yang berbeda, Pulau Pramuka yang berpenghuni dan terdapat berbagai macam aktivitas masyarakat, sedangkan Pulau Semak Daun tidak berpenghuni. Penelitian ini dilaksanakan pada September 2019 – Mei 2020 di Laboratorium Mikrobiologi dan Bioteknologi Molekuler, Fakultas Perikanan dan Ilmu Kelautan, Universitas Padjadjaran. Proses Next Generation Sequencing (NGS) dilakukan oleh Novogene Co., Ltd di Singapura menggunakan primer Rbcl 646F – 998R dengan target amplicon 331 bp. Hasil penelitian didapatkan kelimpahan tertinggi pada ordo Bacillariales di semua stasiun (41 – 49%), disusul dengan ordo Fragilariales dan Naviculales.Nilai OTUs tertinggi terdapat pada stasiun 4 (508), disusul dengan stasiun 3 (500), 1 (481), dan 2 (471). Indeks Shannon dan Simpson berada di kisaran >5 dan 0 < D < 0.5, yang berarti di semua stasiun memiliki keanekaragaman yang tinggi dan memiliki dominansi yang rendah.

Abstract

Marennine is a blue-green pigment produced by diatoms of the Haslea genus, one of which is Haslea ostrearia. This marennine pigment is water-soluble and confirmed to contain polyphenols and glycosides. There are two forms of marennine pigment: the intracellular form of marennine (IMn) and the extracellular form of marennine (EMn). Marennine pigments exhibit various biological activities such as antibacterial, antiviral, antioxidant, allelopathic, and inhibiting the growth of other diatoms. With this biological activity, marennine can be used in various fields, one of which is aquaculture.This research aims to determine the level of toxicity and analyze the effect of exposure to the extracellular marennine produced by H. ostrearia on the juvenile blood clam Tegillarca granosa. By using the toxicity test method, Blue water which is supernatant of H. ostrearia (BW) was tested on juvenile of commercially important bivalve species, the blood clams Tegillarca granosa, with three treatments, namely control (0 mg/L), treatment A (0.25 mg/L) and treatment B (0.5 mg/L of BW). The observation parameters of this study were LC50-72 h with one-way ANOVA analysis. The results showed that the BW produced by H. ostrearia increased the survival of juvenile blood clams T. granosa up to 27.7% in treatment B (0.5 mg/L) compared to control within 72 h. The one-way ANOVA analysis revealed that the control and treatment B (0.5 mg/L) were significantly different. This study shows that there is potential to develop the use of marennine in shellfish aquaculture.
