Abstract

Paraquat (1-1’-dimetil-4-4’-bipiridinium diklorida) merupakan herbisida non-selektif, kontak langsung dengan spektrum lebar, dan banyak digunakan untuk mengendalikan pertumbuhan gulma. Paraquat apabila terdistribusi dalam lingkungan perairan, akan mempengaruhi populasi fitoplankton serta mengganggu produktivitas lingkungan perairan tersebut. Penelitian ini bertujuan untuk mengetahui toksisitas paraquat terhadap Chlorella sp., suatu alga yang penting dalam sistem rantai makanan dalam lingkungan perairan, karena efek herbisida terhadap alga dapat mempengaruhi level trofik diatasnya. Sebanyak 2,0 mL paraquat dengan konsentrasi 2226,0 mg/L; 1187,2 mg/L; 593,6 mg/L; 296,8 mg/L; 148,4 mg/L 74,2 mg/L; 37,1 mg/L dan 18,6 mg/L, ditambahkan ke dalam 250,0 mL kultur Chlorella sp. dengan kepadatan 150 x 104 sel/mL dalam media air laut. Kepadatan sel alga dalam media tersebut ditentukan pada selang waktu jam 0, 24, 30, 36, 42, 48, 54, 60, 66, dan 72, menggunakan Haemocytometer Neubauer Improved di bawah mikroskop dengan perbesaran 400 kali.Hasil penelitian ini menunjukkan bahwa toksisitas paraquat berupa penghambatan laju pertumbuhan populasi Chlorella sp. Hambatan laju pertumbuhan meningkat dengan peningkatan konsentrasi paraquat. Pada konsentrasi yang sangat tinggi (> 471,11 μg/L), peningkatan konsenrasi paraquat praktis tidak mengakibatkan peningkatan hambatan laju pertumbuhan populasi Chlorella sp.Toksisitas paraquat terhadap Chlorella sp. secara kuantitatif dinyatakan dengan harga EC50, yaitu sebesar 125,8 μg/L.

Abstract

A homology model of human 3β-HSD type 2 has been developed from homology modeling techniques using Phyre2 server and refi ned by ModRefi ner. The PROCHECK, QMEAN and ProSA-web online tools were carried out to evaluate the stereochemical quality of the model. The Ramachandran plot resulted from PROCHECK showed that 84.5% residues are in the most favored region, 13.7% are in the additional allowed region, 1.5% are in the generously allowed region and 0.3% are in the disallowed region. The QMEAN (Z-score) are 0.509 (-3.006) and Z-score of ProSA-web is -7.10. The negative values of protein fold energies also found in almost all sequences. Furthermore, molecular docking was also applied to validate the model using MOE. The hydrogen bonding interactions with Tyr154, Ser124, and Ser218 are found in all docked substrates as well as known inhibitors (trilostane and epostane). A dataset of azasteroid inhibitors were also docked into the substrate active site of human 3β-HSD2. These docked structures were utilized to construct corresponding docking-based QSAR equation by employing genetic algorithm (GA) statistical analysis. The contructed best QSAR equation has a robust predictive power according to its statistical parameters, hence may be applied to supersede the default scoring function provided by docking software. These results indicate that the human 3β-HSD2 model was successfully evaluated as a good model.
