Abstract

The rhizome of Alpinia zerumbet (Pers.) B.L. Burtt & R.M. Sm) contains phenolic compounds, has antihyaluronidase activity which can reduce wrinkles on the skin. This study aims to determine the physical characteristics and antihyaluronidase activity of the nanostructured lipid carrier (NLC) of the Alpinia zerumbet rhizome extract in-vitro. Alpinia zerumbet rhizome powder (4/18) was extracted by kinetic maceration method using 70% ethanol solvent, concentrated with a rotary vacuum evaporator at 100 mm Hg at a temperature of ± 50oC. The resulting extract was prepared into NLC nanoparticles using polyethyleneglycol 8-beeswax, isopropyl myristate and acrylyl glucoside with the high speed homogenization-ultrasonication method. The resulting NLC extracts were characterized by nanoparticle size and morphology, polydispersity index and zeta potential and antihyaluronidase activity test was determined using the Sigma Aldrich and Tu & Tawata methods with slight modifications. The results of the NLC characterization of Alpinia zerumbet rhizome extract were 171.3 nm; 0.37; -26.8 mV; and spherical shape. The results of the antihyaluronidase activity of Alpinia zerumbet rhizome extract loaded NLC was 552.75 µg/mL. It was concluded that the Alpinia zerumbet rhizome extract loaded NLC can be formulated into topical preparations which have the potential to be antihyaluronidase with the ability to reduce wrinkles on the skin.

Abstract

Gold sea cucumber extract (Stichopus hermanni) is rich in collagen and unsatturated fatty acid also known omega 3 such as docosahexaenoic acid and eicosapentaenoic acid that highly effective as an antioxidant. Oligomeric proanthocyanidin is the main content in grape seed oil and the most powerful antioxidant agent that can protect collagen. The aim of research was to determine the effect of gold sea cucumber extract concetration on antioxidant activity of grape seed oil nanoemulsion. The three formulas with results test of antioxidant activity with DPPH method using 15% Brij CS12® and 30% propylene glycol has done which the oil phase was dispersed in the aqueous phase.Variation that have been done is to use the antioxidant activity (IC50) of gold sea cucumber extract for 2, 4, and 6 times. The resulting preparations was conformed of organoleptic, nanoemulsion type, clarity, globule size, viscosity and rheology, pH and freez thaw test. Antioxidant activity of gold sea cucumber extract and grape seed oil were (IC50= 53.79 and 17.41 μg/mL respectively). The nanoemulsion was analysed using two-way ANOVA method. Antioxidant activity at storage for 0; 2 and 4-weeks of formula I, II, III were (IC50= 49.15; 49.45, 51.15 µg/mL), (IC50= 44.12, 47.48, 48.14 µg/mL), and (IC50= 42.14, 43.45, 45.33 µg/mL) respectively. Can be concluded that the higher concentration of gold sea cucumber extract, the antioxidant activity of nanoemulsion is higher, but it decreased during the storage.

Abstract

Liquorice root extract (Glycyrrhiza glabra L.) contains glabridin, an isoflavone substance that has the ability to inhibit the oxidation of L-tyrosin and L-DOPA in the formation of melanin, therefore it can be used as skin whitening. The aim of the study was to determine the potential of the best skin whitening cream using positive control of kojic acid. Formulated 1 blank formula, 3 cream formulas using various concentrations of ethanol extract 70% liquorice roots of (0.01, 0.11, 1.01)% based on the value of tyrosinase inhibition activity in vitro from extracts of (50, 75, 100)%. Spectrophotometric method is used to measure the absorption of dopacrome by a microplate reader which was incubated at 37°C at λ 490 nm for 20 minutes. Oil in water cream was prepared by mixing extract with cream base at a temperature of 70-75°C at a speed of 400 rpm for 25 minutes. Stability test was carried out for 4 weeks at room temperature storage and a temperature of 40°C, evaluated for physical quality. The results of determination of IC50 of kojic acid was 20.88 µg / mL; IC50 of extract variation were (126.75; 1130.90; and 10092.41) µg / mL respectively. Cream has milky white-yellowish color; smell of flowers; soft texture; homogeneous; there is no separation; has type M/A; plastic thixotropic flow properties; has an increasing value including viscosity, spreadability, globule size, and pH, which are respectively (455000-620000)cPs, (2695.82-3545.83)mm2, (54.66-66.27)μm , and (4.44-5.04); tyrosinase inhibition activity in formula 1, 2 and 3 were stored at weeks 0, 2nd and 4th were respectively (1.78; -25.74; 22.04)%, (6.74; 6, 12; 4.49)%, and (-28.78; 53.06; 20.32)%. It can be concluded that the cream formula containing liquorice root extract with a concentration of 1.01% is the best formula so it can be used as skin whitening.
