Abstract

Trigonella  foenum-graecum  (TFG)  is  one  of  medicinal  plants  contains  some steroidal sapogenin  such as diosgenin, yamogenin, gitogenin, tigogenin and trigoneoside, also  alkaloid  trigonellin,  which  is  have  many  activity  as  antidiabetic,  estrogenic  and  also as  anti  cancer.    This  experiment  was  done  to  explore  the  activity  of  some  extract  of TFG on some cell lines such as MCF7 (Human Breast Cancer Cell-line), T47D (Human Breast  Cancer  Cell-line),  PC3  (Human  Prostate  Cell-line)  and  SKOV  (Human  Ovarian Carcinoma Cell-line).  This assay was done using MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium)  methods.    Results  showed  that  ethyl  acetate  fraction  gives  the lowest  IC50  than  another  extracts.  IC50  for  PC3  is  66.24  ppm,  IC50  for  MCF7  is  41.81 ppm,  IC50  for  T47D  is  58.63  ppm.    These  datas  can  be  used  for  further  research  to isolate the active compound from TFG. Abstract

In this work we have investigated mixing in a modified orbiting screw mixer  (MOSM)  designed  for  solid-solid mixing.  Mixing  was  carried  out  using urea powder and natural zeolite powder (UZ) of three varying particle sizes (50-60, 60-80 and 80 mesh). Power consumption was calculated from the measured torque of orbit and screw, obtained from computerized records. It was found that the  mixing  process  in  the  modified  orbiting  screw  mixer  with  air  injection required a lower power consumption for each particle-size group when compared to mixing without air injection. With UZ mixing in MOSM with  air injection, the lowest E was obtained for  the 60-80 mesh particle-size group  (4,297 Joule/kg-1),whereas when mixing without  air injection, the value was  10,296 J/kg. The best mixing  operation in this experiment was achieved at NFr  =  1.18x10-3 and in the range  of  values  NRe≈  8.77x107 to  2.63x108.  Moreover,  in this  study,  we  have developed  an  equation  to  estimate the  power  consumption required  for  mixing and determined its correlation with dimensionless numbers.

Abstract

Isolation of the main compound of sponge Axynissa aplysinoides collected from Lombok seawater, has been conducted. Methanol extract of dried sponge was fractionated into nonpolar, semipolar and polar fractions using hexane, ethyl acetate and buthanol. Identification and final purification of the main compound was done using column chromatography and high performance liquid chromatography (HPLC). Structure elucidation of the main compound was performed using mass spectrometry (EIMS), 1H- and 13C-NMR spectroscopies. The main compound was identified as (E)-(4-hydroxystyryl) trimethylammonium.Key words : isolation, structure elucidation, Axynissa aplysinoides

Abstract

ABSTRACTMillennial moms have different behaviors in shopping for children's items, and the use of makeup is considered as a trend in childre1. Unfortunately, there are currently no special children's makeup products tailored to the needs of children. This study aims to explore how market acceptance of the use of makeup in children, then further translated into a marketing strategy concept that is suitable for children's makeup products so that they can be well received by target consumers, namely Millennial moms who have 3- to 6-year-old girls. 11 years old. The research method used is descriptive verification with market acceptance independent variables (X1) and marketing mix strategies (X2), as well as the dependent variable purchase interest (Y). Data obtained through surveys using questionnaires for 250 millennial mom respondents, who lived in the DKI Jakarta area, which were then analyzed by correlation test and multiple regression test. The results showed that the market acceptance of children's makeup products was positive so that it was potentially good to be marketed with a marketing mix strategy that was in line with the target market for children's makeup products, namely girls aged 3-11 years. As for the verification study shows that there is an influence of market acceptance and marketing mix strategies both partially and simultaneously on the interest in buying children's makeup products. Keywords : market acceptance, marketing mix strategy, buying intention, makeupfor kid, millennial mom. ABSTRAK Millennial mom memiliki perilaku berbeda dalam berbelanja barang-barang anak, dan penggunaan makeup maka pada anak dianggap sebagai suatu tren. Namun sayangnya saat ini belum tersedia produk makeup khusus anak yang disesuaikan dengan kebutuhan anak. Penelitian ini bertujuan untuk menggali bagaimana penerimaan pasar terhadap penggunaan makeup pada anak, untuk selanjutnya diterjemahkan lebih jauh menjadi sebuah konsep strategi pemasaran yang sesuai untuk produk makeup anak tersebut sehingga bisa diterima dengan baik oleh target konsumen yaitu para Millennial mom yang memiliki anak perempuan berusia 3-11 tahun. Metode penelitian yang digunakan adalah deskriftif verifikatif dengan variabel bebas penerimaan pasar (X1) dan strategi bauran pemasaran (X2), serta variabel terikat minat pembelian (Y). Data diperoleh melalui survey menggunakan kuesioner pada 250 responden millennial mom, yang tinggal di wilayah DKI Jakarta yang selanjutya dianalisis dengan uji korelasi dan uji regresi berganda. Hasil penelitian menunjukkan bahwa penerimaan pasar produk makeup anak positif sehingga berpotensi baik untuk dipasarkan dengan strategi bauran pemasaran yang sesuai dengan target pasar produk makeup anak yaitu anak perempuan usia 3-11 tahun. Adapun untuk studi verifikatif menunjukkan bahwa ada pengaruh penerimaan pasar  dan strategi bauran pemasaran baik secara parsial dan secara simultan terhadap minat pembelian produk makeup anak.Kata kunci : penerimaan pasar, strategi bauran pemasaran, minat pembelian, makeup anak, millennial mom

Abstract

Aspergillus terreus is a Saprophyte fungus that produces several secondary metabolites as lovastatin (anti-cholesterol drug) and itaconic acid (a polymer material). Lovastatin is one of the statin class of drugs that have efficacy as antihypercholesterolemic. Plasmid transformation is the introduction and incorporation of exogenous plasmid into cells orprotoplast. In this study, pTRLI plasmid (pTRI inserts containing lovE gene as a regulator gene in the biosynthesis of lovastatin) will be transformed by electroporation transformation. The purpose of this research is transformation of pTRLI plasmid into protoplasts of Aspergillus terreus by electroporation and obtain stable transformants. The research was initiated by isolation of pTRLI plasmid. Then pTRLI plasmid was determined purity and concentration by nanodrop. Furthermore, Protoplasts of Aspergillus terreus were isolated enzymatically by adding an enzyme which can degrade the cell wall of Aspergillus terreus which contains chitin and cellulose. PTRLI plasmid were transformed into protoplasts of Aspergillus terreus by electroporation. These transformants were grown in Czapek-Dox medium containing pyrithiamine agar and the number of transformants mg-1 of pTRLI plasmid was calculated. Transformants were selected to grow in Czapek-Dox medium containing piritiamin 1 mg l-1. The number of transformants produced 187 transformants mg-1 of PTRLI plasmid. Transformants are stable up to five generations by growing the transformants in Czapek-Dox medium agar containing piritiamin 1 mg l-1. The success of the transformation indicated by ptrA gene in transformants that can be amplified by PCR. The size of fragment DNA is 801 bp.

Abstract

Along with the increasing activity of people, especially in urban areas, and thenumber of instant products that are fast-paced and practical, an innovativewaterless hand sanitizer product has emerged, known as antiseptic hand sanitizeror hand sanitizer. This hand sanitizer product contains an antiseptic used to killgerms on hands, which consists of alcohol and triclosan. Based on data from2015 - 2020 which is the result of a Top Brand survey of hand sanitizer users inIndonesia, it is found that the Antis and Dettol brand hand sanitizers always havean index that places them in the top two positions in that period. In this study, twoindependent variables were selected including brand image and perceivedquality, with the assumption that the independent variables were proven to havea positive relationship with trust and purchasing decisions. This research wasconducted in five areas of DKI Jakarta (West Jakarta, East Jakarta, CentralJakarta, North Jakarta, and South Jakarta) in November - December 2020. Thesample used was 200 respondents. The data analysis technique used StructuralEquation Model (SEM), in this case, Liserel. The results of the study concluded:1) Brand Image affects consumer confidence in antis products; 2) Quality affectsconsumer confidence in antis products; 3) Brand Image affects consumerdecisions to buy antis products; 4) Quality influences consumer decisions to buyantis products; 5) Trust affects consumer decisions to buy antis products; 6)Brand image and quality can explain 86% of the trust variance, while theremaining 14% is explained by other factors. Brand image, quality, and trust canexplain 92% of the variance of purchasing decisions while the remaining 8% isexplained by other factors; and, 7)There are direct and indirect effects of brandimage and quality variables on the decision to buy antis products. The indirecteffect is through the variable of consumer confidence in antis products.

Abstract

Hasil operasi dan telaah agen Badan Pengawas Obat dan Makanan (BPOM) memperlihatkan semenjak pandemi COVID 19 terjadi lonjakan perdagangan penawar dan pangan tidak sah melintasi perangkat daring hingga 480 persen.1 Kosmetik tidak sah juga dipasarkan dengan sistem daring dengan kuantitas klien mencapai lima ribu orang.2 Sedangkan data dari ZAP Beauty Index 2018 memperlihatkan pada 17.889 Wanita Indonesia memperlihatkan, mereka mendalami make up semenjak berusia 13-15 tahun (41,9%) dan hanya 36,4% yang melakukan perawatan di klinik kecantikan.3 Hal ini memperlihatkan bahwa Wanita Indonesia harus lebih berhati-hati memilih produk kosmetiknya, dan pemilik kecantikan harus memajukan mutu produk maupun jasanya supaya dapat bersaing dengan produk daring, terutama yang illegal. Data awal berkaidi klinik dr Jo yang sudah menjalankan perdagangan produk secara daring terjadi kenaikan dari rata-rata perdagangan dari Rp 48.663.667,- menjadi Rp 66.581.592,- sedangkan kuantitas lawatan penderita di Klinik Kecantikan dr. Jo, Klinik Estetika dan Klinik dr. Fauzyah menyusut. Riset ini bermaksud untuk memahami dampak mutu produk dan jasa atas kemudahan dan kesetiaan penderita di tiga Klinik Kecantikan Kota Bekasi. Desain riset yang dimanfaatkan adalah riset deskriptif kuantitatif dengan melakukan peninjauan kepada 225 klien Ketiga klini yang dialokasikan secara wajar, setelahnya ditelaah SEM memakai Lisrell 8.0. Hasil Data memperlihatkan Mutu Produk berdampak penting atas Kemudahan Penderita (t pengamatan 4,357 > H01 1,96) tapi tidak berdampak penting atas Kesetiaan (t pengamatan 0,684< H03 1,96). Mutu Jasa Perawatan berdampak secara penting atas Kemudahan Penderita (t pengamatan 2,071 > H02 1,96) tapi tidak berdampak penting atas Kesetiaan Penderita (t pengamatan -0,439 < H04 1,96). Kemudahan Penderita berdampak langsung secara penting atas kesetiaan penderita (t pengamatan 4,095 > H05 1,96). Sebagaimana data ini dapat ditanggapi kesetiaan penderita ketiga klinik kecantikan ini akan didapat jika penderita mendapatkan kemudahan. Kemudahan akan diperoleh dengan memajukan mutu produk dan mutu jasa diklinik kecantikan ini.

Abstract

PT. KLM merupakan perusahaan air minum yang ada di Bogor Jawa Barat, memproduksi air minum pH 8+ atau air minum alkali, yang berfungsi bukan sekedar menghilangkan rasa dahaga tetapi juga memberikan manfaat kesehatan lainnya. Tujuan penelitian adalah mengidentifikasi faktor internal dan eksternal produk air minum alkali PT. KLM, menggunakan analisa SWOT (Strengths, Weaknesses, Opportunities and Threats) dari beberapa faktor yang diperoleh untuk menentukan strategi bisnis PT KLM dalam menghadapi persaingan industri. Hasil analisis SWOT dan IFE EFE, didapatkan kekuatan dan peluang memiliki skor lebih besar dari kelemahan dan ancaman. Pada faktor internal, kekuatan memiliki skor 2.13 dan kelemahan dengan skor 1.99 sehingga didapatkan nilai x adalah 0.14. Faktor eksternal, peluang mendapat skor 2.17 dan ancaman yang mendapat skor 1.90, sehingga didapatkan nilai y adalah 0.27. Dilihat dari titik x dan y (0.14, 0.27), posisi berada pada kuadran 1 yaitu pertumbuhan agresif. Langkah implementasi dari Strategi pertumbuhan agresif adalah melakukan penetrasi pasar, mengembangkan market melalui media promosi dan meningkatkan kualitas mesin

Abstract

Trypsin is an enzyme that has a unique mechanism of cutting peptide bonds specifically at the carboxyl side of lysine or arginine amino acids, with another amino acid. This study aims to analyze a trypsin-like protease (TLP) found in Lactobacillus plantarum FNCC 0270,  by performing partial proteomic tests, i.e. MALDI-TOF/TOF, and standard bioinformatics tools. SDS-PAGE analysis showed 4 protein bands. Two bands of the (P1 and P2) showed molecular weights equivalent to 47.35 and 38.42 kD, each generating 8 and 11 peptide fragments respectively. According to information in www.ncbi.nlm.nih.gov/genbank/structures, the structure  of serine protease HtrA (subs. plantarum L. plantarum ST–III) consists of three domains. Using Clone Manager® software by aligning two sequences we obtained eleven. The Lactobacillus produces of the trypsin-like serine protease has 40-90% similarity. Using the Clustal W2 software we passed the 11 sequences through multiple alignments, and found that the isolate L. plantarum is closely related to L. buchneri, L. brevis, and L. malefermentans on the phylogenetic tree. Alignment analysis results showed that all 8 peptide fragments of band 1 and 11 peptide fragments of band 2,  of the SDS-PAGE, were located in the active domain region of  the fourth trypsin-like serine protease producing Lactobacilli.

Abstract

The most common skin disorder is acne (Acne vulgaris) which is a chronic inflammatory disease that occurs in the pilosebaceous unit. Infection can be caused by Propionibacterium acnes and Staphylococcus aureus. Moringa leaves are known to inhibit acne growth. Piper crocatum leaves are also thought to have antibacterial activity. The study aims to determine the effect of the combination of the two extracts in inhibiting the growth of acne-causing bacteria, also tested the gel formula to meet physical and chemical parameters as well as stability to temperature and storage, also to test the acute dermal irritation against rabbit. Each extract with a concentration of 0.3%; 0.6%; 1.25%; 2.5%; 5%; 10%; 20%, incubated then measured the inhibition zone, then determined the minimum inhibition zone concentration of the two extracts to determine the lowest levels of the extracts which still gave antibacterial activity against the tested bacteria by the well diffusion method. The extract combination was formulated in a gel preparation with the excipients Carbopol 940, PG, Phenoxyethanol, TEA, CMC, and Aquadest. Antibacterial activity test for gel preparations used the well method with positive control Medi-Klin® gel. Then the formula evaluation includes physical, chemical, and microbiological evaluations. The results showed that the combination gel form had activity against P. acne and S. aureus at concentrations of 2.5% : 2.5%. The combination gel preparation of the two extracts also has synergistic effect in inhibiting acne bacteria. The formula can meet physical and chemical parameters and is stable to temperature and storage.
