Abstract

In vitro conservation needs highly efficient micropropagation protocol. The objective of the research was to obtain an efficient and reproducible protocol for pummelo (Citrus maxima(Burm.) Merr.) micropropagation through direct shoot formation. The experiment was arranged in a completely randomized design with two factors and 20 replications. The 1stfactor was type of explant, i.e. cotyledon and epicotyl segments of Pummelo cv. Cikoneng, while the 2ndfactor was the media composition as follow (1) MS + 1.0 mg BAP L-1 + 0.5 mg Kinetin L-1 + 0.5 mg NAA L-1; (2) MS + 2.0 mg BAP L-1 + 0.5 mg Kinetin L-1 + 0.5 mg NAA L-1; (3) MS + 1.0 mg BAP L-1 + 1.0 mg Kinetin L-1 + 0.5 mg NAA L-1; (4) MS + 2.0 mg BAP L-1 + 0.5 mg Kinetin L-1+ 1.0 mg NAA L-1; (5) MS + 2.0 mg BAP L-1+ 1.0 mg Kinetin L-1 + 1.0 mg NAA L-1. Observation was conducted on days to shoot induction, number of explant forming shoots, shoot height, number of shoots, leaves, and roots. The results showed that adventitious shoots emerged from callus in epicotyl (6-8 WAP), but adventitious shoots could emerge directly without an intervening callus phase from cotyledon (4-5 WAP). Shoots emerged from epicotyl were weak and vitrous due to hyperhydricity, thus they can not be used for micropropagation. Cotyledons cultured in media MS + 1.0 mg BAP L-1 + 0.5 mg Kinetin L-1 + 0.5 mg NAA L-1or media MS + 1.0 mg BAP L-1 + 1.0 mg Kinetin L-1 + 0.5 mg NAA L-1gave the highest percentage of explant forming adventitious shoot (38.8 and 26.3%), highest efficiency of shoot formation (62.5 dan 72.5%), and highest numbers of leaves (value of 1.9 leaves shoot-1) and roots (1.1 roots shoot-1) compared to other media. Since shoot height and number of leaves and root were not significantly different in both media, thus cotyledon and media MS + 1.0 mg BAP L-1 + 0.5 mg Kinetin L-1+ 0.5 mg NAA L-1which was less in Kinetin is suggested to be used for pummelo micropropagation.Keywords: Citrus maxima, epicotyl, cotyledon, regeneration, pummelo

Abstract

ABSTRAK Development of black rice is quite rapid in Indonesia. The objective of this study was to obtain doubled haploid black rice lines with good agronomic characters through anther culture. Materials used in this experiment were 6 F1 derived from black rice landrace (Melik) and two cultivated varieties (Inpari 13 and Fatmawati): Melik/Inpari13//Inpari13, Melik / Inpari13// Melik, Melik/Fatmawati //Fatmawati///Fatmawati, Melik/ Fatmawati//Fatmawati// /Fatmawati, Melik/Fatmawati, and Melik/Fatmawati//Melik. The experiment was performed using a completely randomized design consisting of 12 replications. An experimental unit was a petri dish containing ± 150 anthers. Results of the anther culture varied widely between crosses. Crosses Melik/Inpari13//Melik, Melik/Fatmawati//Melik, and Melik/Inpari13 //Inpari13 showed the best response on callus induction and plant regeneration compared to the other crosses. Melik / Inpari13 // Melik produced  the highest number of acclimatized plants, the highest number and percentage of doubled haploid: 63 plants, 42 plants and 91.3% respectively. The highest number of doubled haploid having black aleuron layer were obtained by Melik/Inpari13//Melik and Melik/Fatmawati//Melik.Keywords: anther culture, black rice, doubled haploid, regeneration

Abstract

Anther culture is useful tool in rice breeding. The technique shortens the time to obtain full homozygous plant in just one generation. Information related to genetic variability among the regenerants is important for further analysis including selection activities. The objective of this study was to analyze agronomic character variability and yield potential among doubled-haploid lines (DH0) derived from anther culture. This study was carried out in Center for Agricultural Biotechnology and Genetic Resources Research and Development (BB Biogen) from October 2016 to February 2017. The experiment was arranged in randomized complete block design (RCBD) with three replications. Forty eight doubled haploid lines derived from anther culture and three check varieties were evaluated. The results showed high genetic variability as well as broad sense heritability (more than 90%) for all variables tested. Several lines showed superior agronomic characters compared to check (Ciherang, Inpari 18, Inpago 10). Anther culture could generate high genetic variability for further selection.Keywords: anther culture, broad sense heritability, haploid technology

Abstract

Tantangan pemulia padi ke depan bertambah seiring dengan terjadinya perubahan iklim yang menyebabkan peningkatan salinitas lahan pertanian. Salinitas dapat mengakibatkan penurunan pertumbuhan dan kehilangan hasil yang besar dalam produksi padi. Tujuan penelitian ini ialah mengevaluasi toleransi salinitas galur-galur harapan padi sawah doubled-haploid /dihaploid (DH) hasil kultur antera pada fase bibit. Penelitian dilaksanakan di rumah kaca dari bulan Desember 2018 sampai Januari 2019. Penelitian dilakukan secara hidroponik dan disusun berdasarkan split plot rancangan kelompok lengkap teracak dengan empat ulangan. Petak utama ialah konsentrasi NaCl (0 mM dan 120 mM) dan anak petak berupa genotipe padi yang terdiri atas 14 galur DH, 4 VUB, pembanding toleran (Pokkali), dan pembanding peka (Inpara 4). Evaluasi pertumbuhan bibit dan skoring kerusakan visual bibit dilakukan 14 hari sesudah pemberian NaCl. Hasil penelitian menunjukkan konsentrasi NaCl 120 mM menyebabkan penurunan tinggi bibit, panjang akar, jumlah daun, jumlah anakan, bobot basah tajuk, bobot basah akar, bobot kering tajuk, bobot kering akar, bobot basah biomassa, dan bobot kering biomassa. Penurunan tinggi bibit dapat dipertimbangkan sebagai karakter penting dalam penapisan toleransi salinitas pada fase bibit. Hasil skoring terhadap kerusakan visual menghasilkan 11 galur DH dan 2 VUB toleran serta 3 galur DH dan 2 VUB moderat toleran. Kata kunci: hidroponik, kultur antera, penapisan, toleransi

Abstract

Mutu beras dan nasi yang mencakup mutu fisik dan kimia (fisikokima) merupakan preferensi konsumen yang perlu dipertimbangkan. Karakter kualitas gabah dan beras meliputi penampilan, tekstur dan rasa. Penelitian bertujuan untuk melakukan pengukuran atau identifikasi secara kuantitatif terhadap karakter fisikokimia beras galur-galur dihaploid padi hitam. Percobaan dilakukan di Laboratorium Mutu, Kebun Percobaan Muara, Balai Besar Penelitian Tanaman Padi (BB Padi) pada bulan Agustus hingga September 2021. Penelitian menggunakan rancangan acak lengkap dengan ulangan atau sampel sebanyak 10 per genotipe. Genotipe yang digunakan terdiri atas 16 (14 galur dihaploid padi hitam dan dua varietas pembanding yaitu Aek Sibundong dan Jeliteng). Pengamatan mutu fisik meliputi kadar air, beras pecah kulit, beras kepala, beras giling, bentuk beras, panjang beras dan butir kapur beras. Karakter kimia beras yang diamati meliputi kadar amilosa, suhu gelatinisasi dan organoleptik. Hasil menunjukkan bahwa galur AW6 memiliki tingkat kemiripan yang dekat dengan varietas pembanding yang ditunjukkan dalam analisis gerombol di Grup 1. Karakter penciri pada Grup 1 ialah kadar air 13.6%, beras kepala 88.7%, beras pecah kulit 76%, beras giling 70%, skor alkali 1-4, suhu gelatinisasi sedang hingga tinggi, bentuk ramping, rasa gurih dan pengapuran kecil. Galur AW6 termasuk kedalam Grup 1 karena memiliki karakter yang mirip dengan varietas pembanding. Kata kunci: amilosa, beras hitam, fisikokimia

Abstract

Pemulia tanaman melakukan seleksi multi-karakter dengan metode yang sesuai, salah satunya adalah indeks seleksi. Analisis multivariat dapat digunakan dalam menyusun model indeks seleksi. Percobaan ini bertujuan untuk menyeleksi galur-galur dihaploid dengan potensi hasil yang tinggi dan memiliki penampilan agronomi baik berdasarkan indeks seleksi. Percobaan dilakukan pada bulan Agustus 2018 sampai dengan September 2019 menggunakan rancangan acak lengkap dengan tiga ulangan dan satu faktor yaitu genotipe. Genotipe yang diuji ialah 89 galur dihaploid padi sawah hasil kultur antera ditambah 4 varietas pembanding (Ciherang, Inpari 30 Ciherang Sub1, Inpari 41, dan Inpari 35 Agritan). Unit percobaan adalah satu bibit per pot. Model indeks seleksi disusun dengan mempertimbangkan hasil analisis ragam, analisis korelasi, analisis lintas, analisis komponen utama, dan ideotipe tanaman yang dituju. Hasil studi ini mengungkapkan bahwa terdapat keragaman sifat agronomi yang diuji dari galur-galur dihaploid. Model seleksi indeks yang disusun adalah sebagai berikut: I (nilai indeks) = 3*0.75 hasil + 0.85 jumlah gabah total per malai – 0.85 tinggi tanaman. Berdasarkan model tersebut, diperoleh 85 galur dengan nilai indeks dan produksinya lebih tinggi dibandingkan Inpari 41. Galur-galur dihaploid terpilih diikutsertakan pada pengujian berikutnya di lapangan. Kata kunci: heritabilitas, korelasi, sidik lintas, variabilitas
