Abstract

The purpose of this research study to isolate and identify bacterial soil and water producing IAA (Indole-3-Acetid Acid) Situgunung, Sukabumi potential biofertilizer. This research study method consists of six stages: soil and water sampling, manufacturing pure isolates, gram stain for bacterial identification, testing bacteria producing IAA (Indole-3-3Acetid Acid) with the Salkowski reagent, IAA standard curve creation, and storage of bacterial isolates obtained in IPB Culture Collection. Soil samples taken from the root zone 5 (five) trees Rasamala (Altingia excelsa) around the hiking trail to the waterfall Cimaracun, Sawer, Kembar, Situgunung, and Situgunung lake, of HM 0 HM 2, HM 5, HM 6, HM 9. Isolation of auxin-producing bacteria using media Nutrient Broth (NB) and Nutrient Agar (NA), which both added 0.0204 grams of L-tryptophan (Trp) per 100 mL of distilled water. The results of the purification of pure isolates obtained 26 NA + Trp medium, 15 isolates from soil samples, while 11 isolates from water samples. Has been fully tested with Salkowski reagent and measurement accuracy of a standard curve with a value of 0.972. The test results showed that 9 out of 10 isolates produced IAA exogenous to the value of the highest concentrations of 9.92 ppm produced by isolates A4.01. Keywords.
