Abstract

Bucks semen is easily damaged during freezing process, due to the formation of ice crystals. Consequently, semen quality decreases particularly the post-thaw sperm motility, viability, intack plasma membrane and intack acrosomal cap. The objectives of this research are to determine the optimum dose of glycerol in Tris extender in maintaining frozen semen quality of crossbred Etawah bucks. Four head of PE buck of 2 - 4 years old were used in this experiment. Doses of glycerol used were 5%, 6% and 7%. Semen was collected once a week using artificial vagina. Results of this experiment indicated that the mean percentage of pre-freezing motility, live sperm, sperm with intact plasma membrane and intact acrosomal cap in 5%, 6% and 7% glycerol were not significantly different (P>0.05). After freezing, the mean percentage of motility, live sperm, sperm with intact plasma membrane and intact acrosomal cap in 6% glycerol (52.60%, 65.03%, 45.63% and 47.54% respectively) were significantly higher (P<0.05) than in 5% (44.31%, 52.00%, 37.60% and 37.00% respectively) and in 7% glycerol (45.28%, 52.10%, 37.97% and 37.14% respectively). However, there were not significant differences in 5% and 7% glycerol for any parameter measured. It was concluded that supplementation of glycerol 6% in Tris extender effective to protect of sperm from various shock during the process of semen cryopreservation, so that it could maintain of frozen semen quality (sperm motility, viability, intack plasma membrane and intack acrosomal cap) that suitable used in AI program of crossbred Etawah bucks.   Key words : Glycerol, PE bucks, semen quality

Abstract

The objective of this study is to determine the best concentration of lactose in Tris extender in maintaining liquid semen viability of Saanen bucks stored at 5°C. Four heads of Saanen bucks of 2-4 years old were used as semen source. Semen was collected once a week using an artificial vagina. Variance analysis was conducted based on completely randomized design with three treatments, i.e 30, 60, and 90 mM lactose (L30, L68 and L90). Duncan test was used to observe the different between treatment. Results indicated that the mean percentage of motility, live sperm, intact plasma membrane, and intact acrosomal cap of liquid semen in treatment L30 (70.83; 78.20; 79.56 and 81.20% respectively) were significantly higher (P<0.05) than treatment L60 (67.08; 73.73; 75.01 and 76.87% respectively) and treatment L90 (61.50; 71.88; 69.08 and 71.33% respectively). After storage for 48 hours at 5°C apparently the mean percentage of motility, live sperm, intact plasma membrane and intact acrosomal cap in treatment L30 were significantly higher (P<0.05) than those of in treatment L60 and L90. Decreasing percentage of motility, live sperm, intact plasma membrane, and intact acrosomal cap during storage at 50C from 0 until 48 hour in treatment L30 (42.83; 19.75; 40.84 and 39.00% respectively) were significantly lower (P<0.05) than those of in treatment L60 (55.41; 23.06; 50.14; and 50.90% respectively) and treatment L90 (54.83; 29.93; 50.14 and 50.97% respectively). It was concluded that supplementation 30 mM lactose in Tris extender could maintain liquid semen viability in Saanen bucks.   Key words: Saanen bucks, lactose, liquid semen

Abstract

Bucks semen is easily damaged compared to bull semen during cryopreservation process. Consequently, frozen semen quality decrease especially motility, live sperm, intact plasma membrane, and intact acrosomal cap after thawing. Objectives of this research was to evaluate the effect of egg yolk concentration in Tris extender with and without seminal plasma in maintaing frozen semen quality of Saanen buck. Four heads of Saanen buck of 2-4 years old were used in this experiment. Semen was collected once a week using artificial vagina. Experimental design applied was factorial complete random desing 2x2, viz. A factor was seminal plasma (A1 = with seminal plasma and A2 = without seminal plasma), and B factor was concentration of egg yolk (B1 = 10% and B2 = 20%). Duncan test were applied to identity differences between treatment. Result of these study indicated that the mean percentage of motility (M), live sperm (LS), sperm with intact plasma membrane (IPM) and intact acrosomal cap (IAC) after dilution and equilibration were not significantly different (P>0.05) in all treatment. After thawing, the mean percentage of M, LS sperm with IPM and IAC in A1 treatment (41.43, 51.52, 56.63 and 50.35% respectively) were significantly higher (P<0.05) than A2 treatment (37.14; 48.67; 52.31 and 45.09% respectively). Likewise, the mean percentage of M, LS, sperm with IPM and IAC in B2 treatment (41.79; 51.32; 55.78 and 49.50% respectively) were significantly higher (P<0.05) than B1 treatment (36.79, 48.86; 53.16 and 45.94% respectively). There were a significant interaction between factors of seminal plasma and concentration egg yolk in Tris extender, where the increase of egg yolk concentration from 10% to 20% in unwashed seminal plasma treatment caused increase in percentages of M, LS, sperm with IPM and IAC. On the other hand in washed seminal plasma treatment there were a trend of decreasing frozen semen quality. It is concluded that the combination of 20% egg yolk in Tris extender with seminal plasma is effective in maintaining frozen semen quality of Saanen bucks. Abstract

In general, local goat productivity in Indonesia is relatively still lower than that from the temperate area. Yearling body weight in local goat about 14-17 kg. However, the local goat have superiority, such as adaptability and a good reproductive performance, which have litter size 1,49-1,61. Increasing local goat productivity may be done by breeding program (cross breeding and selection) and the environmental improvement. Both cross breeding program and selection that involve mating regulation will be efficient by reproductive technology application, such as artificial insemination (AI). Application of AI in goats is limited by four main factors that influence simultaneously, viz. poor quality of frozen semen, the variety of female reproductive performance, improper technical and insemination time, and poor management. To overcome these constraints his obligatory to pay more attention, so that AI will be able to be used as one of tools to increase the population and genetic quality in goats.   Key words: Goat, reproductive efficiency, artificial insemination

Abstract

Development of buffalo was relatively slow in the last five years (1993-1997), but the number of slaughtered animals was also increased in the same period. Factors limiting their development were extensive farming system, farming as a secondary enterprise, slow growth and low reproductive efficiency of the animals. Optimization of artificial insemination (AI) program in one of the alternative to improve buffalo productivity in Indonesia. AI is an effective tool toimprove the genetic quality and to increase the buffaloes population. Application of AI in buffalo has been conducted since 1975, but until now the results are not still significant. Within the last four years the calving average was 750 head/year, the implementation of targeted insemination doses reached less than 50%/year with S/C still above 2 and CR 38.32%. The limiting factors affecting this low performance are quality of semen, female fertility, skill of technician and zootechnical knowledge of farmers. Improvement of reproductive efficiency of buffaloes interrelated with optimization of AI program could be imposed through increasing of ability of farmers to detect oestrus, oestrus synchronization using hormones (PGF-2α and progesterone), improvement of quality of frozen semen with special attention on dilution materials used, control of equilibration time, and control of duration and temperature of thawing, as well as accuracy of insemination time.   Key words: AI, buffalo, semen quality, optimization, oestrus synchronization
