Abstract

The objectives of this study is to decrease blood cholesterol levels from white male rats(Rattus novergicus L.) after giving infusion of soursop leaf (Annona muricata L). This studyused experimental laboratory method with complete randomized design. The subject of thisstudy are 15 white male rats weigth 100-160 gramme which were divided into 5 groups, 3rats for each group. The results were taken from 2 times measurement of blood cholesterolwhich are pre-test and post-test. Treatment began with a high-fat feeding for 7 days. The pretestwas done at 8th day. Then each group were given treatment, group A (C-) was givenaquadest, group B (C+) was given simvastatin and group C-E was given infusion of soursopleaf which each dose is 3, 5,and 10 soursop leaf. The post-test was done at 12 hours. Datawere analyzed by using ANOVA one way. The results of statistical analysis didn’t havesignificant differences between treatment groups. But consider from the average change,infusion dose of 5 soursop leaf gives the best decrease of blood choleterol levels.Key words: Blood cholesterol levels, infusion of soursop leaf, white male rats.

PENGARUH KONDISI KERJA DAN BUDAYA KERJA TERHADAP KINERJA KARYAWAN DENGAN MOTIVASI KERJA SEBAGAI VARIABEL INTERVENING(STUDI PADA TENAGA KEFARMASIAN PT. Ada beragam faktor yang mempengaruhi kinerja. Salah satu adalah motivasi yang berperan penting bagi karyawan dalam melakukan pekerjaan. Motivasi karyawan dalam bekerja dapat dipengaruhi oleh keadaan sekitarnya seperti kondisi kerja dan budaya kerja. Kondisi kerja merupakan kondisi yang dirasakan karyawan ditempat karyawan bekerja. Sedangkan budaya kerja merupakan cara atau perilaku karyawan dalam bekerja yang ditetapkan perusahaan. Tujuan penelitian ini adalah untuk mengetahui pengaruh kondisi kerja dan budaya kerja terhadap motivasi dan melihat dampaknya terhadap kinerja karyawan yang dalam hal ini adalah tenaga kefarmasian PT. Kimia Farma Apotek Unit Bisnis Manado. Kuesioner didesain dengan pertanyaan tertutup digunakan sebagai instrumen penelitian. Pengambilan sampel menggunakan metode stratified sampling yang dibagi dalam kelompok apoteker dan kelompok tenaga teknis kefarmasian dengan besar sampel adalah 22 apoteker dan 60 tenaga teknis kefarmasian dengan metode sensus. Berdasarkan kerangka konsep yang dibentuk, metode analisis regresi berganda dan analisis regresi sederhana digunakan sebagai teknik analisis data, program SPSS (Statistical Package For Service Solutions) versi 19 digunakan untuk menganalisis data yang diperoleh setelah penelitian. Penelitian ini menunjukkan bahwa dalam analisis regresi berganda pada kedua kelompok, secara parsial kondisi kerja berpengaruh terhadap motivasi dan budaya kerja berpengaruh terhadap motivasi. Demikian juga secara serempak pada kedua kelompok, kondisi kerja dan budaya kerja berpengaruh terhadap motivasi. Sedangkan dalam analisis regresi sederhana pada kedua kelompok diperoleh hasil bahwa motivasi berpengaruh terhadap kinerja. Berdasarkan koefisien determinasi diperoleh pengaruh kondisi kerja dan budaya kerja terhadap motivasi pada kelompok apoteker sebesar 70,1% dan pada kelompok tenaga teknis kefarmasian sebesar 36%. Untuk pengaruh motivasi pada kinerja, dari koefisien determinasi pada kelompok apoteker diperoleh sebesar 49,6% dan kelompok tenaga teknis kefarmasian sebesar 13,9%. Kata kunci: kinerja, motivasi, budaya kerja, kondisi kerja, apoteker, tenaga teknis kefarmasian  ABSTRACT Performance is a measure of the success of a company doing its business functions. There are many factors that influence the performance. One of the important factor supporting the employees doing their job is motivation. Employee’s motivation can be influenced by the environment such as working condition and work culture. The working condition is a conditions that perceived by the employees where they work. And the work culture is a manner or a behavior that specified by the company for employees to do their job. The aims of this research is to determine the influence of working conditions and work culture to motivation and see the impact on the employee's performance at PT. Kimia Farma Apotek Business Unit Manado. The questionnaire was designed with closed questions as a research instrument. Sampling used the stratified sampling method that be divided into pharmacists group and pharmaceutical technical personnel group with the sample 22 pharmacists and 60 pharmaceutical technical personnel with census method. Based on the conceptual framework that established, multiple regression analysis and simple regression analysis is used as data analysis, SPSS (Statistical Package For Service Solutions) version 19 is used to analyze the data obtained after research. This research shows that in a multiple regression analysis in both groups, partially working conditions can influence motivation and work culture can influence motivation. Likewise synchronously in both groups, working conditions and work culture can influence motivation. Whereas in the simple regression analysis in both groups showed that the motivation can influence performance. Based on the determination coefficient obtained the influence of working conditions and work culture to motivation of pharmacists group is 70.1% and in the pharmaceutical technical personnel is 36%. The influence of motivation to performance from determination coefficient in pharmacists group is 49,6% and in the pharmaceutical technical personnel is 13,9%. Keywords: performance, motivation, work culture, working condition, pharmacist, pharmaceutical technical personnel.

Abstract

Epidermal growth factor receptor (EGFR) is one of tyrosine kinase that overexpressed in many types of cancer. The ihbiition of EGFR activity has known as a rational target for cancer therapy. However,  several EGFR inhibitors have limitations related to resistance problems or reduced tumor response. Therefore, the effort to develop new inhibitors with greater efficacy is very important to overcome the problems. In the present study, we designed a computational method by combining ligand based pharmacophore modeling and molecular docking for screening the potential natural compounds as EGFR inhibitors. Validation result of ligand based pharmacophore modeling and docking molecular by ROC curves showed that both of these protocols had AUC ≥ 0.7. The Redocking erlotinib to EGFR in our docking protocols gave the RMSD < 2 Å. Therefore, our computational method could be applied for virtual screening. Initial screening based on pharmacophore modeling obtained 86 out of 179.816 compounds that could be mapped with the pharmacophore features.  Further screening through molecular docking showed two compounds had a lower docking score than erlotinib, i.e ZINC00941342 and ZINC72325782. However, by careful analysis of their interaction profiles, only ZINC00941342 which formed hydrogen bonds with Met769, which is known as an EGFR key residue. Additionally, it was also found to form hydrogen with Cys773. Therefore, we recommend ZINC00941342 as a potential EGFR inhibitor.
