Abstract

Rhizoctonia solani is a soil pathogen that causes diseases in wide range of hosts of agricultural, horticultural and flower crops. Biological control is the most promising way for the diseases management and it is environment friendly too. The objective of this study was to isolate and screen the potency of soil bacteria as biological control from various local compost and peat soil of tropical peat swamp forest in Kalampangan Zone, Central Kalimantan. Forty seven isolates from peat soil and compost were screened for biocontrol agent of Rhizoctonia solani . R. Solani Seven out of thirteen peat soil isolates, and six out of thirty three compost isolates showed antagonistic activity against in Potato Dextrose Agar. The cultivation of the antagonistic isolates in Trypticase Soy Broth (TSB) was extracted and analysed by high performance liquid chromatography (HPLC) column. The HPLC analyzes indicated that the antagonistic isolates produce an antifungal iturin A. Macroscopic observation of isolates colonies showed that form of their colonies were amuboid, myceloid, curled, circular, rhizoid, irregular and filamentous. These achievement indicate peat swamp forest not only offer a potential biocontrol agents of damping off but also provide a new source for production of antibiotics.

Abstract

Dua biakan micro-Rhizopus ditumbuhkan pada substrat minyak nabati untuk meneliti kemampuan memproduksi lipase ekstraselular. Medium basal ditambahkan minyak olive, kelapa, jagung, bunga matahari dan minyak kedelai. Aktivitas lipase diuji dengan metode titrasi. Aktivitas lipase tertinggi pada substrat minyak olive pada R. microsporus var. oligosporus UICC 550 (16,23 U/g Biomassa) dan minyak kelapa pada R. microsporus var. rhizopodiformis UICC 520 (30,5 U/g Biomassa) setelah 48 jam inkubasi. Nilai pH mengalami kenaikan selama berlangsungnya proses fermentasi.

Abstract

Telah dilakukan penelitian tentang Palm Kernel Meal (PKM) yang difermentasi selama 7 hari secara alami tanpa penambahan inokulum yang hasil fermentasi disebut dengan PKMK. Produk fermentasi ditambahkan telur Hermetia illucens L. (maggot) diinkubasi selama 14 hari. Hasil penelitian menunjukkkan adanya kenaikan berat dan panjang maggot. Analisis proksimat menunjukkan kenaikan berat kering, nitrogen total dan serat, tetapi terjadi penurunan lemak dan abu.

Abstract

Effect of nitrogen and carbon source on the production of extracellular lipase by R.microsporus var. oligosporus UICC 550 was studied. The enzyme activity was alsocharacterized in terms of temperature, pH, stability at room temperature, and effectof divalent ion. The amount of 2.5% (w/v) of olive oil as carbon sources and 5% (w/v)peptone as nitrogen sources were the optimum for production of lipase enzyme.Partial purifications using ammonium sulfate followed by dialysis showed that at pH6.5 and temperature 35oC was the optimum condition, respectively. The stability(remaining up to 80% of the optimum enzyme) was recorded at pH 6.5 after 24 hoursincubation at room temperature. The optimum activity remained 40% of the after onehour incubation at 35oC. Divalent ions concentration at 1 mM, Zn2+, Cu2+, Mg2+ andFe2+ inhibited the lipolytic activity.

Abstract

A total of 236 yeast isolates from mollusc, litter, and plant samples from Gunung Halimun National Park were screened for cellulolytic activity based on Smith method by using 0,2% (w/v) cellulose-azure for 30 days. The results showed that 12 isolates (9 isolates from plants, 2 isolates from molluscs, dan 1 isolate from litter) have cellulolytic activity. These isolates were further screened based on Teather and Wood method for six days to determine their cellulases components by using specific substrates. Carboxymethyl cellulose (CMC) 0,1% (w/v) was used as a specific substrate to determine endoglucanase activity. Avicel 0,1% (w/v) was used as a specific substrate to determine exoglucanase activity. Cellobiose 0,1% (w/v) was used as a specific substrate to determine β-glucosidase activity. The results showed that 6 isolates from plant have β-glucosidase activity, and 1 isolate from plant have β-glucosidase and endoglucanase activities. Five isolates (2 isolates from plants, 2 isolates from molluscs, and 1 isolate from litter) showed no cellulase activity on specific substrates after six days incubation.
