Abstract

Latar belakang: Tanaman Upo-upo (Phyllodium pulchellum (L.) Desv) adalah salah satu jenis tanaman yang dikategorikan dalam kelompok tanaman obat atau tanaman herbal. Tanaman Upo-upo tumbuh khususnya di dataran tinggi dan banyak digunakan sebagai tanaman obat tradisional. Tujuan: Untuk mengetahui aktivitas ekstrak etanol daun Upo-upo (Phyllodium pulchellum (L.) Desv) terhadap Bakteri gram positif Streptococcus pyogenes, Staphylococcus epidermidis, Bacillus cereus dan bakteri Gram Negatif Salmonella typhi, Vibrio cholerae, Escherichia coli. Metode: Penelitian ini menggunakan metode ekstraksi maserasi terhadap bahan uji dan metode Disc diffusion menurut Kirby & Bauer untuk menentukan diameter hambatan dengan konsentrasi ekstrak 1% b/v, 3% b/v, 5 % b/v, 7% b/v. Hasil: Menunjukkan bahwa konsentrasi ekstrak Etanol Daun Upo-upo (Phyllodium pulchellum (L.) Desv) 1% b/v, 3% b/v, 5 % b/v, 7% b/v menunjukkan daya hambat pertumbuhan bakteri Gram positif dan bakteri Gram negatif. Rata-rata diameter zona hambat yang terbentuk pada kelompok konsentrasi ekstrak tertinggi 7% dengan masa inkubasi 1x24 jam terhadap bakteri Gram positif adalah Staphylococcus epidermidis (23,35mm), Streptococcus pyogenes (20,36mm), Bacillus cereus (15,73mm) sedangkan diameter zona hambat pada bakteri Gram negatif adalah Salmonella typhi (25,24mm), Vibrio cholerae (13,93mm) dan Escherichia coli (11,92mm). Rata-rata diameter zona hambat pada kelompok konsentrasi 7% dengan masa inkubasi 2x24 jam terhadap bakteri Gram positif adalah Staphylococcus epidermidis (22,15mm), Streptococcus pyogenes (18,96mm), Bacillus cereus (14,92mm) sedangkan diameter zona hambat pada bakteri Gram negatif adalah Salmonella typhi (24,64mm), Vibrio cholerae (12,92mm) dan Escherichia coli (10,92mm). Kesimpulan: ekstrak Etanol Daun Upo-upo (Phyllodium pulchellum (L.) Desv) mempunyai aktivitas antibakteri terhadap pertumbuhan Streptococcus pyogenes, Staphylococcus epidermidis, Bacillus cereus, Salmonella typhi, Vibrio cholerae, Escherichi coli dan bersifat bakteriostatik.

Abstract

Research about antibacterial effectiveness of ethanol extract of purple sweet potato tuber skin (Ipomoea batatas L.) against Staphylococcus aureus and Salmonella typhi has been conducted. This study aims to determine the antibacterial effectiveness of ethanol extract of Purple Sweet Potato Tuber Skin against the growth of Staphylococcus Aureus and Salmonella Typhi and the most effective concentration. This research is expected to provide additional information about alternative medicine that has proven effective in laboratory testing for the community as an antibacterial agent. This type of research uses the agar diffusion method, Staphylococcus Aureus, namely with an average zone of resistance for concentrations of 2% w/v, 4% w/v, 6% w/v and 8% w/V of 12 mm, 16.26 mm, 24.26 mm, 27.1 mm and for positive control 31.16 mm, negative control around 6 mm. For the growth of Salmonella typhi, namely with an average zone of inhibition for concentrations of 2% w/v, 4% w/v, 6% w/v and 8% w/V of 12.83 mm, 17 mm, 25 mm, 26, 6 mm and for positive controls 32.5 mm, negative controls around 6 mm. Statistical tests using SPSS 24 (Statistical Product and Service Solutions) based on Analysis of Variance (ANAVA) showed that there were significant differences between each treatment and the positive control. Conclusion Purple Sweet Potato Tuber Skin is effective as an antibacterial.
