Abstract

Pharmacokinetic and bioavailability studies of three enteric coated 50-mg sodium diclofenac tablets marketed in Indonesia (DR-50, FR-50 and VN-50) have been carried out in six healthy Indonesian volunteers with three-way crossover design. The results obtained showed that there were significant differences in the bioavailability parameters of the three products tested. The lag-time observed varied from 1 to 2 hours for DR-50, 0.5 to 2 hours for FR-50, and 1.5-2 hours for VN-50. Peak plasma levels of 1.16 + 0.59, 2.06 + 1.03, and 0.68 + 0.3 μg/ml were reached in 3.3 + 1.4, 1.9 + 0.9, and 3.8 + 1.2 hours following administration of DR-50, FR-50 and VN-50, respectively. The area under the curve given by each product were 3.81 + 1.60 μg/ml.hr for DR-50, 3.87 + 0.89 μg/ml.hr.for FR-50, and 3.13 + 1.33 μg/ml.hr for VN-50. The elimination half life of diclofenac observed varied from 2.1 to 6.9 hours with a mean value of 3.7 + 1.5 hours.

Abstract

Mucoadhesive strength test of some polymer excipients is importance for development of oral sustain release with mucoadhesive system to enchance bioavailability. Mucoadhesive strength of granules that prepared using single and combination of polymers i.e. from carboxymethylcellulose, gum arabic, and sodium alginate with Metolose 90SH-15.000 (Metolose K-15) and Metolose 90SH-100.000 (Metolose K-100) on rat stomach and intestinal had been determined by bioadhesion and wash off test. Results showed that single polymer and its combination could adhesive to rat stomach and intestine in 5 minuter. The granul prepared using Metolose K-15 and sodium alginat in single and combination form gave the strongest adhesive strength to stomach, while Metolose K-15 and gum arabic in a single form showed the strongest adhesive to intestine. The strongest adhesion to stomach and intestine was the granule combination of sodium alginat and Metolose K-15 (20 : 40) with the amount of granule adhesived to stomach and intestine was 100 % and 88 % respectively.

Abstract

Penghantaran senyawa hidrofilik secara transkutan memiliki keterbatasan karena permeabilitas rendah sehingga sulit dalam menembus lapisan stratum corneum (SC). Natrium askorbil fosfat (NAF) merupakan salah satu senyawa turunan vitamin C yang sangat hidrofil dan permeabilitas rendah terhadap lapisan kulit. Sebagaimana vitamin C, NAF dapat berfungsi sebagai antioksidan juga memiliki aktivitas sebagai antikerut karena dapat memicu pertumbuhan kolagen pada fibroblast. NAF dibuat dalam nanoemulsi, karena sediaan dengan ukuran globul yang kecil diharapkan dapat menembus lapisan SC dan membawa zat aktif berpenetrasi ke dalam lapisan kulit. Optimasi formulasi nanoemulsi dilakukan melalui pendekatan statistik design of experiment (DoE) yang terdiri dari desain faktorial dan respon surface methods (RSM hingga diperoleh hasil yang lebih efektif dan efisien. Komposisi minyak (VCO), surfaktan (Tween 80), ko surfaktan (PEG 400) dan proses pengadukan (waktu dan kecepatan) merupakan faktor yang berperan dalam optimasi formulasi. Penentuan faktor tersebut melalui desain eksperimen 2 level factorial dan dilanjutkan dengan metode Box Behnken menggunakan perangkat lunak Minitab 17. Parameter yang diamati adalah ukuran globul dan indeks polidispersitas. Berdasarkan hasil optimasi diperoleh nilai VCO 10% dan kombinasi surfaktan 24%, waktu dan kecepatan pengadukan 5 menit, 200 rpm memberikan ukuran globul ±180 nm dan indeks polidispersitas dibawah 0,5.
