Abstract

Ethanol extracts of the fruits of three kinds of Capsicum showed similar potencies in their antimicrobial activities against Gram (+) and Gram (-) bacterias, and fungi, although they contained different level of capsaicin. Bioautographic tests demonstrated that capsaicin was the main antimicrobial component. At least two other non-polar components of ethanol extract also contributed in the antimicrobial activity and very likely that these compounds were responsible for the activity toward Pseudomonas aeruginosa.

Abstract

Antidiabetic activity of the ethanol extract of Andrographis paniculata Nees herbs had been tested using glucose tolerance test in rat and alloxan induced diabetic mice. The extract reduced rat blood glucose at the dose of 2.0 g/kg bw but there was no effect observed at dose of 0.50 g/kg bw and 1.0 g/kg bw. It is interesting that the extract at doses 2.1 g/kg bw and 2.8 g/kg bw, and the effect was much lower at 3.5 g/kg bw also showed the capacity to reduce blood glucose level of alloxane induced diabetic mice.

Abstract

Hypoglycemic activity of hexane, ethylacetate, ethanol and aqueous extract of Andrographis paniculata herbs had been tested using glucose tolerance test. Extraction was conducted consecutively using hexane, ethylacetate and ethanol. Aqueous extract was obtained directly from the crude drug similar to the traditional use. At the dose level of 0.5 g/kg bw, hexane and ethylacetate extract did not show the hypoglycemic effect. Significant activity was shown by ethanol and aqueous extract and the activity of the latter was higher than the former.

Abstract

Mucoadhesive strength test of some polymer excipients is importance for development of oral sustain release with mucoadhesive system to enchance bioavailability. Mucoadhesive strength of granules that prepared using single and combination of polymers i.e. from carboxymethylcellulose, gum arabic, and sodium alginate with Metolose 90SH-15.000 (Metolose K-15) and Metolose 90SH-100.000 (Metolose K-100) on rat stomach and intestinal had been determined by bioadhesion and wash off test. Results showed that single polymer and its combination could adhesive to rat stomach and intestine in 5 minuter. The granul prepared using Metolose K-15 and sodium alginat in single and combination form gave the strongest adhesive strength to stomach, while Metolose K-15 and gum arabic in a single form showed the strongest adhesive to intestine. The strongest adhesion to stomach and intestine was the granule combination of sodium alginat and Metolose K-15 (20 : 40) with the amount of granule adhesived to stomach and intestine was 100 % and 88 % respectively.

Abstract

Hypotensive and antihypertensive effect of fractions of ethanolic extract of “kucai” (Allium schoenoprasum L., Liliaceae) bulb had been tested on male Wistar rats. The ethyl acetate, n-buthanol and aqueous fractions of both fresh and dry bulb had hypotensive and antihypertensive effects. The aqueous fraction of fresh bulb at a dose of 25 mg/kg body weight showed the highest hypotensive and antihypertensive activities. The n-buthanol fraction of dry bulb at a dose of 50 mg/kg body weight showed the highest hypotensive activity. The ethyl acetate fraction of dry bulb at a dose of 50 mg/kg body weight showed the highest antihypertensive activity.

Abstract

One  of the plants  that grow  in Indonesia,  and  still does  not  use widely  is singawalang  (Petiveria  alliaceae)  from  Phytolaceae  family.  Traditional   herbal medicine  preparations  contained  of  Petiveria  alliacea  in  the  Caribbean,  Latin America,  West  Africa  and  other  regions  for  hundreds  years  has  been  used  to treat  pain,  colds,  inflammation,  tumor,  bacterial  and  fungal   infection,hyperlipidemia,  diabetes and  other  diseases.  This  study  aims to determine  the antibacterial  activity  of  singawalang  growth  in  Indonesia  against  resistant bacteria  to  antibiotics.  The  minimum  inhibitory  concentration   (MIC)  and minimum  bactericidal  concentration(MBC)  were  determined  by  microdilution broth method and type of antibacterial action were determined by observing the profile of bacterial growth curve. Ethanol extract of dried leaves showed activity against bacteria i.e. MRSA (Methicillin-Resistant Staphylococcus aureus), MRCNS (Methicillin-Resistant  Coagulase-Negative  Staphylococci),  VRE  (VancomycinResistant  Enterococcus).   Further  studies  conducted  by  reviewing  the  type  of work  by  looking  at the  growth curve  and  the observed changes  in cell walls by scanning  electron  microscopy  (SEM)  at  MRCNS  (Methicillin-Resistant Staphylococcus  coagulase-Negative).  The  results  showed  that  ethanol  extracts and  all  fractions  showed  good  antibacterial  activity  against  resistant  bacteria, are  bacteriostatic  hexane  fraction  until  the  concentration  of  8  MIC  and  change the morphology of the cell wall of MRCNS.Key words:Singawalang, Antibacteria, Broth Microdilution, bacterial growth curve, SEM

Abstract

Penyakit kulit terinduksi obat atau disebut juga drug eruption merupakan reaksi toksik yang menyebabkan gangguan pada kulit setelah penggunaan obat. Drug eruption merupakan reaksi obat yang paling umum ditemukan. Obat yang banyak menginduksi reaksi ini adalah antibiotik dan AINS (antiinflamasi non-steroid). Penelitian ini bertujuan untuk mengidentifikasi pola kejadian penyakit kulit terinduksi obat, menentukan obat yang paling banyak diduga sebagai penyebab, dan menentukan jenis penyakit kulit terinduksi obat yang paling banyak terjadi pada tahun 2009-2011 di salah satu rumah sakit pemerintah di Kota Bandung. Penelitian ini dilakukan secara retrospektif dengan mendata laporan Monitoring Efek Samping Obat (MESO) dan rekam medik pasien rawat inap kelas III bagian kulit. Berdasarkan hasil penelitian, dapat disimpulkan bahwa pada periode 2009–2011 terdata 250 kasus drug eruption dengan adanya peningkatan jumlah setiap tahunnya. Obat yang paling banyak diduga menjadi penyebab adalah antibiotik terutama amoksisilin (oral) dan jenis penyakit kulit terinduksi obat yang banyak terjadi adalah drug eruption yaitu reaksi kulit dengan manifestasi klinik berupa kemerahan dan makulopapular.Kata kunci: penyakit kulit terinduksi obat, drug eruption, amoksisilin, studi retrospektifAbstractDrug induced skin disease or also known as drug eruption is a toxic reaction that causes skin disorder after drug administration. Drug eruption is the most common drug reaction. Antibiotic and NSAID (non-streoidal anti-inflammatory drug) were drugs that often induce this reaction. This study was made to identify the pattern of drug eruption incidence, to determine the most suspected drug, and to determine the most occurred type of drug eruption in year 2009-2011 in one of the government hospital in Bandung. This study was done retrospectively by using MESO report and medical records of dermatology patients in third class wards. According to this study, it can be concluded that in year 2009 – 2011, there were 250 drug induce skin disease cases with the increasing number every year. The most common suspected drug was antibiotic especially amoxicillin (oral) and the most occurred type of drug induced skin disease is drug eruption which clinical manifestation is redness and maculopapular.Keywords: Drug Induced Skin disease, drug eruption, amoxicillin, retrospective study.

Abstract

Kinin adalah senyawa alkaloid dari kulit batang Cinchona sp. yang digunakan sebagai antimalaria. O-desmetil kinin (C19H22N2O2) adalah senyawa turunan kinin baru yang diperoleh dengan menghilangkan gugus metil pada kinin. Selain sebagai antimalaria, kinin juga dapat digunakan untuk mengobati kram kaki. Karena efek samping yang besar, FDA (2009) melarang penggunaan kinin oral dalam pengobatan kram kaki. Oleh karena itu, penelitian ini dilakukan untuk menguji efek kinin dan pada rute pemberian oral dan topikal serta mengetahui efek o-desmetil kinin pada terapi kram kaki. Mencit dibagi menjadi 5 kelompok yaitu kelompok normal, kelompok kinin oral dosis 26 mg/kg bb, kelompok kinin topikal dosis 52 mg/kg bb, kelompok o-desmetil kinin oral dosis 26 mg/kg bb, dan kelompok o-desmetil topikal dosis 52 mg/kg bb. Mencit diletakkan di atas rotarod dengan kecepatan 10-16 rpm selama 4 menit. Waktu yang dapat ditempuh mencit selama berada di atas rotarod diukur. Perlakuan yang sama diulang dengan durasi 45 menit selama 4,5 jam. Setelah menit ke 180, kelompok uji mengalami peningkatan waktu ketahanan di atas rotarod sedangkan kempok normal mengalami penurunan. Pada menit 270, kelompok oral dan topikal memberikan perbedaan yang bermakna dibandingkan dengan kelompok normal. Pemberian kinin dan odesmetil kinin pada rute pemberian oral dengan dosis 26 mg/kg bb dan topikal dengan dosis 52 mg/kg bb sebagai antikram memberikan hasil yang bermakna dibandingkan dengan kelompok normal (p<0,05). O-desmetil kinin memberikan efek yang tidak berbeda bermakna dari kinin base (p<0,05).Kata kunci: kram kaki, kinin, o-desmetil kinin, rotarod.AbstractQuinine is an alkaloid from the bark of Cinchona sp. as an anti-malaria. O-desmethyl quinine (C19H22N2O2) is the new quinine derivative, by losing its methyl. Beside the function as anti-malaria, quinine is used for leg cramps treatment. Because of the great side effects, FDA (2009) banned the use of quinine oral for leg cramps therapy. Therefore, this study was aimed to determine the effect of quinine by oral and topical administration and to determine o-desmethyl quinines effect for leg cramps therapy. Mice were divided into normal group, quinine oral dose 26 mg/kg bw, quinine topical dose 52 mg/kg bw, o-desmethyl quinine oral dose 26 mg/kg bw, and o-desmethyl topical dose 52 mg/kg bw. Mice were taken on the rotarod at 10-16 rpm for 4 minutes. Time to remain on the rotarod was measured. The same treatment was repeated after 45 minutes for 4.5 hours. Over the 180 minutes, the endurance of the test groups were increased on the contrary, the normal group is decreased. After 270 minutes of observations, the oral and topical groups have significant difference compared by the normal group. The administration of quinine by oral dose 26 mg/kg bb and topical dose 52 mg/kg bb as an anti-cramps showed significantly difference compared to the normal group (p<0.05). O-desmethyl quinine has no significantly difference compared by quinine (p<0.05).Keywords: leg cramps, quinine, o-desmethyl quinine, rotarod.

Abstract

Ketapang (Terminalia catappa L.) banyak digunakan sebagai obat tradisional. Informasi mengenai aktivitas antimikroba dari kulit batang ketapang sejauh ini belum diketahui. Penelitian ini bertujuan untuk menguji aktivitas antimikroba ekstrak etanol kulit batang ketapang. Kulit batang diekstraksi dengan metode refluks menggunakan pelarut etanol. Ekstrak difraksinasi dengan metode ekstraksi cair-cair menggunakan pelarut n-heksan, kloroform, dan etil asetat. Pengujian aktivitas antimikroba dilakukan dengan metode difusi agar. Ekstrak etanol simplisia kering kulit batang ketapang menunjukkan aktivitas antimikroba terhadap Staphylococcus aureus dan Candida albicans dengan konsentrasi hambat minimum (KHM) masing-masing 200 μg/cakram dan 300 μg/cakram. Fraksi etil asetat merupakan fraksi yang paling aktif dengan KHM 50 μg/cakramterhadap S. aureus dan C. albicans. Satu mikrogram fraksi etil asetat setara dengan 1,27x10-3 μg tetrasiklin HCl dan setara dengan 2,1x10-4 μg ketokonazol. Aktivitas antimikroba ekstrak etanol simplisia kering lebih tinggi daripada ekstrak etanol simplisia segar. Fraksi etil asetat memiliki aktivitas antimikroba paling tinggi dan semua fraksi menunjukkan aktivitas antimikroba yang lebih rendah daripada antibiotik pembanding.Kata kunci : ketapang, antimikroba, fraksi etil asetat, Candida albicans, Staphylococcus aureus “Ketapang” (Terminalia catappa L.) had been widely used as a folk medicine. Antimicrobial activity of “ketapang” bark had not been reported. The aim of this research was to investigate antimicrobial activity of ethanol extract of “ketapang” bark. The bark was extracted by refflux method, using ethanol as solvent. The extract was fractionated by liquid-liquid extraction method, using n-hexane, chloroform, and ethyl acetate as solvents. Antimicrobial activity was tested by agar disc diffusion method. Dried crude drug ethanolic extract of “ketapang” bark showed antimicrobial activity against Staphylococcus aureus and Candida albicans with minimum inhibition concentration (MIC) of 200 μg/disc and 300 μg/disc, respectively. The ethyl acetate fraction had highest antimicrobial activity against S. aureus and C. albicans with MIC of 50 μg/disc. A microgram of ethyl acetate fraction was equivalent to 1.27 x 10-3 μg tetracycline hydrochloride and 2.1 x 10-4 μg ketoconazole, respectively. Antimicrobial activity of dried crude drug was higher than fresh crude drug ethanolic extract. The ethyl acetate fraction had highest antimicrobial activity and all fractions showed lower activity than standard antibiotics.Keywords: ketapang, antimicrobial, ethyl acetate fraction, Candida albicans, Staphylococcus aureus

Abstract

Hisbiscus surattensis L the local name “tamoenju” is utilized by ethnic Sumari (Central Sulawesi) as an antidiabetic herbal, but de nitive studies on ef cacy and safety are lacking. Therefore, this study was aimed to investigate the antidiabetic activity of ethanolic extract of the leaves and fractions using in vitro and in vivo models. Ethanol extract (EE) of the leaves of H. surattensis L was prepared by maceration. The ethanol extract was separated into three fractions by liquid/liquid extractions to yield n-hexane (HF), ethyl acetate (EAF) and water fraction (WF). Firstly, the extract and fractions were tested for its inhibitory activity on α-glucosidase in vitro. The results showed IC50 EE, HF and WF more than 5000 ppm while the IC50 fraction of EAF 3888.34 ppm. Then, oral glucose tolerance test (OGTT) of the extract/fractions were carried out in normal mice. On OGTT, the ethanol extract, EAF, and WF at doses of 150 and 300 mg/kg BW were able to inhibit increasing the level of glucose in the blood. The results showed that the extract and fraction of H. surattensis L leaves in vitro had no effect as α-glucosidase inhibitor but in vivo had the hypoglycemic effect especially on the fraction of ethyl acetate and the water fraction.
