Abstract

The one of  genes that play an important role in the body metabolism that is closely related to productivity is the Leptin gene. This study aims to determine the assosiation polymorphism of exon 2 Leptin  gene  based on Single Nucleotide Polymorphism (SNP) with birth weight of Balinese cattle. This study used 47  DNA of Balinese cattle, their  birth weight (BW), average daily gain (ADG) and yearling weight (YW) data, that were obtained from BPTU HPT Pulukan Bali. The isolation of DNA using a high salt concentrated method and DNA target was duplicated by  PCR machine. Analysis genetic mutation of Leptin genes was performed by evaluating SNP in Exon 2 at R25C  and R25H, from direct sequencing results, using Bioedit and MEGA 5.2 program. The association of Leptin gene polymorphism with BW, ADG and YW were  analyzed using variance analysis. The result showed that there were For genotypes based on SNP R25C and R25H, ie CC, CT, CA, and AA. There were  no assosiation between Leptin gene polymorphism with birth weight (BW), average daily gain (ADG) and yearling weight (YW) on Balinese cattle.

Abstract

Mutation on leptin gene affect leptin function as  one of the hormones  which contribute to body fat deposition.  This research aims to determine the effect of R25C and R25H mutations on the leptin gene toward  back fat thickness and Body Condition Score (BCS) in Ongole Grade cattle. The study was conducted with a quasi-experimental method, using completely randomized design. The treatments in this study were genotypes differences was, namely CC, TT and AA.  This research used  sixthteen heads of Ongole Grade Cattle  aged ± 2 years, and was reared for 60 days intensively. The feed  consisted of forage and concentrate with a ratio of 25: 75, as much as 3% dry matter of body weight and drinking water was given ad libitum. The backfat thickness was estimated using ultrasound performed on 4.5-6.5 MHz frequency, with depth of 8.8-13 cm, BCS was assessed visually by using 1 – 5 score.  The backfat thickness data were analyzed by variance component and BCS data was analyzed by Kruskal Wallis. The results showed average of backfat  thickness  CC, TT and AA genotype were  3.14 mm,  2.9 mm and 3.57 mm, respectively. The  body condition score for CC, TT and AA genotype were  3.54, 3.63 and 3.46  respectively.  The Mutation of leptin gene on  R25C and R25H have not effect to backfat thickness and body condition score in PO cattle.

Abstract

Itik lokal merupakan salah satu sumberdaya genetik atau plasma nutfah ternak unggas di Indonesia sebagai penghasil daging dan telur. Sebagian besar masyarakat Indonesia, itik dipelihara sebagai penghasil telur. Tujuan dari penelitian ini yaitu untuk mengetahui korelasi antara lebar yolk, tinggi yolk dan indeks yolk dengan haugh unit , untuk mendapatkan besaran nilai pengaruh lebar yolk, tinggi yolk dan indeks yolk terhadap haugh unit telur Itik lokal. Data yang digunakan dalam penelitian ini dianalisis menggunakan Path Analysis dan diuji normalitas juga linieritas. Data penelitian telah berdistribusi normal atau (Sig) > 0,05. Hasil penelitian ini menunjukkan adanya korelasi yang signifikan (P< 0,01) antara tinggi yolk dan indeks yolk dengan haugh unit sedangkan lebar yolk tidak signifikan dengan haugh unit (P>0,01). Berdasarkan scatter plot dan analisis varian diperoleh hasil bahwa lebar yolk tidak linier terhadap haugh unit sedangkan tinggi yolk dan indeks yolk linier. Besarnya pengaruh tinggi yolk terhadap haugh unit yaitu 1,42 % dan indeks yolk terhadap haugh unit sebesar 2,09 %. Kata kunci : Haugh Unit, Indeks Yolk, Itik lokal, Path Analysis, Telur Itik

Abstract

Haugh unit is one of the indicators that determine albumen quality. This study aims to know the correlation and the magnitude of the effect of albumen width, albumen height, and albumen index on the haugh unit of sentul chicken eggs. The research method used was correlational method. This research was using secondary data of 119 sentul chicken eggs and tested with normality test, linearity test, correlation, and path analysis used SPSS 26. The result showed that there was a correlation between albumen width, albumen height, and albumen index on the haugh unit of sentul chicken eggs and the effect of albumen height on the haugh unit of sentul chicken eggs is 83.13% and the effect of albumen index on the haugh unit of sentul chicken eggs is 9.78%.
