Abstract

Penggunaan bahan alam semakin meningkat dan masih menjadi andalan di beberapa negara berkembang. Penelitian kali ini menggunakan ekstrak etanol dari herba Phyllanthus niruri Linn. Dalam usaha menghambat pertumbuhan Staphylococcus aureus. S. aureus merupakan salah satu mikroflora normal di wajah yang berperan dalam infeksi jerawat. Penelitian ini merupakan penelitian eksperimental menggunakan Rancangan Acak Lengkap dengan 7 perlakuan dan 5 ulangan. Perlakuan yang digunakan adalah pemberian ekstrak etanol herba P. niruri Linn. untuk konsentrasi 5%; 25%; 50%; 75%; 100%, pengenceran menggunakan pelarut DMSO 10%. Kontrol positif menggunakan ciprofloxacin, sedangkan kontrol negatif menggunakan DMSO 10%. Zona hambat yang diperoleh dianalisis menggunakan uji Kolmogorov–Smirnov dan uji homogenitas Levene. Apabila variansi data homogen, maka dilanjutkan analisis dengan Anova yang dilanjutkan dengan uji Post Hoc. Dari perhitungan kontrol negatif, variasi konsentrasi (5%, 25%, 50%, 75%, 100%) dan kontrol positif diperoleh rata-rata zona radikal berturut sebesar 6 mm; 7,46 mm; 7,52 mm; 7,6 mm; 8,52 mm; 8,98 mm; 31,08 mm. Semua variasi konsentrasi ekstrak etanol herba P. niruri Linn. mampu menghasilkan zona hambat radikal dengan zona hambat terbesar tetap dijumpai pada kontrol positif. Hal ini menunjukkan bahwa konsentrasi ekstrak etanol herba P. niruri Linn. yang semakin besar akan berbanding lurus dengan peningkatan aktivitas penghambatan, namun belum seoptimum kontrol positif. The use of natural materials is increasing and is still a mainstay in several developing countries. This research was used ethanol extract from herb Phyllanthus niruri Linn. in an effort to inhibit the growth of Staphylococcus aureus. S. aureus is one of the normal microflora on the face that helps infection. This research is an experimental study used a completely randomized design with 7 preparations and 5 replications. The treatment used ethanol extract of P. niruri Linn herb. for a concentration of 5%; 25%; 50%; 75%; 100%, the dilution used 10% DMSO solvent. Positive controls used ciprofloxacin, while negative controls used DMSO 10%. The inhibition zone obtained was analyzed using the Kolmogorov-Smirnov test and the Levene homogeneity test. If the data variance is homogeneous, then the analysis is continued with ANOVA followed by the Post Hoc test. From the calculation of negative controls, various concentration (5%, 25%, 50%, 75%, 100%) and positive controls obtained an average radical zone of 6 mm; 7.46 mm; 7.52 mm; 7.6 mm; 8.52 mm; 8.98 mm; 31.08 mm. All variations in the concentration of ethanol extract of P. niruri Linn. herbs able to produce a radical inhibition zone with the largest inhibition zone still found in positive controls. This showed that the greater concentration of ethanol extract of P. niruri Linn. herb. will be directly proportional to the increase in inhibition activity, but not optimal as much as positive control.

Abstract

Pisang kepok (Musa paradisiaca L.) adalah buah yang mudah ditemukan, akan tetapi kulit buahnya selama ini jarang dimanfaatkan. Pada kulit buah M. paradisiaca L. terkandung flavonoid, alkaloid, tanin, saponin, dan terpenoid yang berperan sebagai senyawa antibakteri. Penelitian ini bertujuan untuk mengetahui apakah ekstrak etanol limbah kulit M. paradisiaca L. mampu menghambat pertumbuhan Klebsiella pneumoniae. Metode penelitian yang digunakan adalah analitik eksperimental, sampel kulit M. paradisiaca L. diperoleh dari pedagang keripik pisang kepok di Kabupaten Karanganyar. Ekstrak etanol 96% kulit M. paradisiaca L. dibuat dengan menggunakan metode perkolasi, sedangkan uji aktivitas antibakteri dilakukan dengan metode Kirby-Bauer. Hasil penelitian menunjukkan bahwa ekstrak etanol 96% limbah kulit M. paradisiaca L. mampu menghambat pertumbuhan bakteri K. pneumoniae, pada konsentrasi 20%; 40%; 60%; 80% dan 100%, dengan rata-rata zona hambat berturut-turut 6,10 mm; 6,35 mm; 6,95 mm; 7,35 mm; dan 7,80 mm. Ekstrak etanol 96% kulit M. paradisiaca L. mampu membentuk zona hambat radikal pada semua konsentrasi terhadap pertumbuhan K. pneumoniae, namun tidak mampu menghasilkan zona hambat radikal setara dengan kriteria sensitif antibiotik ciprofloxacin 5 µg berdasarkan CLSI 2018 terhadap pertumbuhan K. pneumoniae. Banana kepok (Musa paradisiaca L.) is a fruit that is easily found, but the peel of the fruit has rarely been used. The peel of M. paradisiaca L. contains flavonoids, alkaloids, tannins, saponins, and terpenoids which act as antibacterial compounds. This study aims to determine whether the ethanol extract of M. paradisiaca L. peel can inhibit the growth of Klebsiella pneumoniae. The research method used was an experimental analytic, M. paradisiaca L. peel samples were obtained from banana chips traders in Karanganyar. Ethanol extract 96% of the skin of M. paradisiaca L. was prepared using the percolation method, whereas the antibacterial activity test was carried out by the Kirby-Bauer method. The results showed that the 96% ethanolic extract of M. paradisiaca L.peel was able to inhibit the growth of K. pneumoniae in 20%; 40%; 60%; 80% and 100% concentrations, with mean inhibitory zones in order 7,46 mm; 7,52 mm; 7,60 mm; 8,52 mm; dan 8,98 mm. 96% of ethanolic extract M. paradisiaca L. skin was able to form radical inhibitory zones at all concentrations of K. pneumoniae growth, but was unable to produce radical inhibitory zones equivalent to the 5µg ciprofloxacin antibiotic sensitive criteria based on CLSI 2018 on the growth of K. pneumoniae.

Abstract

Kasus diare di Indonesia pada umumnya disebabkan oleh masalah kebersihan lingkungan, makanan, dan infeksi mikroorganisme. Salah satu mikroorganisme penyebab diare adalah bakteri Shigella dysenteriae. Kulit pisang diketahui mempunyai sifat anti bakteri. Tujuan penelitian ini untuk mengetahui kemampuan ekstrak etanol 96% limbah kulit pisang kepok (Musa paradisiaca L.) membentuk zona radikal terhadap Shigella dysentriae berdasarkan variasi konsentrasi. Penelitian ini menggunakan desain penelitian analitik eksperimental dengan pendekatan post test with control. Ekstrak etanol 96% kulit Musa paradisiaca L. dibuat dengan variasi konsentrasi 20%, 40%, 60%, 80%, dan 100%. Aktivitas antibakteri diuji dengan metode cakram dengan kontrol negatif DMSO 10% dan kontrol positif ciprofloxacin 5 µg, dan dilanjutkan dengan analisis Anova satu jalur. Hasil dari penelitian ini menunjukkan terbentuknya zona hambat pada konsentrasi 20%, 40%, 60%, 80%, dan 100% berturut-turut 6,09 mm, 6,26 mm, 6,58 mm, 7,58 mm, dan 9,00 mm. Hal ini menunjukkan semakin tinggi konsentrasi zat antibakteri maka semakin besar juga zona hambat radikal yang terbentuk. Uji Anova satu jalur diperoleh nilai p ≤ 0,05, sehingga dapat disimpulkan bahwa ekstrak etanol 96% kulit Musa paradisiaca L. mampu menghambat pertumbuhan Shigella dysenteriae. Diarrhea cases in Indonesia are generally due to environmental problems, food hygiene, and microorganism infections. One of the microorganisms that cause diarrhea is Shigella dysenteriae. Banana peels are known have antibacterial properties. The purpose of this project was to study the antibacterial activity of 96% ethanol extract of Kepok banana (Musa paradisiaca L.) peel waste with variations concentration on the growth of Shigella dysenteriae. This study uses an experimental analytic research design by post test with control. Ethanol extract 96% of the peels of Musa paradisiaca L. was made with a composition variation of 20%, 40%, 60%, 80%, and 100%. Antibacterial activity was tested by disc-method with DMSO as negative control and ciprofloxacin as positive control. The data obtained were analyzed with one way Anova. The results of this study indicate the formation of inhibition zones at concentrations of 20%, 40%, 60%, 80%, and 100% collected 6.09 mm, 6.26 mm, 6.58 mm, 7.58 mm, and 9,00 mm. This data shows that the greater the concentration of antibacterial substances, the greater the zone of radical inhibition formed. Anova one path test obtained p value 0.001, so it can conclude that ethanol extract 96% of the skin of Musa paradisiaca L. able to inhibit the growth of Shigella dysenteriae.

Abstract

Latar belakang: Extended-Spectrum Beta Lactamase (ESBL) merupakan enzim yang diproduksi oleh bakteri gram negatif salah satunya adalah E coli. Dimana bakteri jenis ini mampu menghasilkan enzim betalaktamase yang dapat melawan jenis antibiotik beta laktam. Prevalensi terjadinya infeksi oleh bakteri penghasil ESBL sebesar 50,60%. Tujuan: penemuan bahan alami yang dapat menjadi alternatif baru untuk mengatasi masalah infeksi Escherichia coli ESBL. Metode: Penelitian ini menggunakan jenis penelitian analitik eksperimental dengan melakukan uji aktivitas antibakteri untuk mengetahui adanya perbandingan daya hambat ekstrak etanol 70% dan etanol 96% biji pepaya (Carica papaya L) terhadap pertumbuhan Escherichia coli ESBL. Penelitian ini menggunakan sampel biji Carica papaya L yang diambil pada buah pepaya matang, segar dan kemudian dikeringkan. Biji Carica papaya L diekstraksi menggunakan cara perkolasi dengan pelarut etanol 70% dan etanol 96% yang kemudian dibuat dalam konsentrasi 100.000 ppm, 200.000 ppm, 300.000 ppm, 400.000 ppm, dan 500.000 ppm. Aktivitas antibakteri menggunakan metode difusi cakram dengan media MHA. Hasil: Pada penelitian ini didapatkan hasil dengan uji Kruskal Walis dengan nilai sig sebesar <0,001 dan dapat disimpulkan bahwa terdapat perbedaan yang bermakna pada daya hambat ekstrak etanol 70% dan etanol 96% biji pepaya (Carica papaya L) terhadap pertumbuhan Escherichia coli ESBL Background: Extended-Spectrum Beta Lactamase (ESBL) is an enzyme produced by Gram-negative bacteria, one of which is E coli. This kind of bacteria is able to produce beta-lactamase enzymes that can fight against beta-lactam antibiotics. The prevalence of infection by ESBL-producing bacteria is 50.60%. Objective: To discover a natural ingredient that can be a new alternative to overcome the Escherichia coli ESBL infection. Method: This study was an experimental analytical research that tested antibacterial activity to determine the comparison of inhibition of ethanol 70% extract and ethanol 96% extract of papaya seeds (Carica papaya L) on the growth of Escherichia coli ESBL. This study used samples of Carica papaya L seeds taken from ripe and fresh papaya, that was then dried. Carica papaya L seeds were extracted using percolation method with ethanol 70% and ethanol 96% solvent which were then made in concentrations of 100,000 ppm, 200,000 ppm, 300,000 ppm, 400,000 ppm, and 500,000 ppm. Antibacterial activity used disc diffusion method with MHA media. Results: The result of this study showed that sig value of Kruskal Walis test was <0.001. Thus, it can be concluded that there is a significant difference in the inhibition of ethanol 70% extract and ethanol 96% extract of papaya seeds (Carica papaya L) on the growth of Escherichia coli ESBL.

Abstract

Tuberculosis (TB) is an infectious disease caused by the bacterium Mycobacterium tuberculosis that can affect the lungs and other organs. Indonesia has the second highest case of pulmonary TB in the world after India. Tuberculosis cases in the Wonogiri Regency area in 2020 amounted to 2,062 patients. There are several groups of people who have a higher risk of developing TB disease, one of which is smokers. Indonesia is the third largest cigarette consumer in the world. The purpose of this study was to determine the relationship between smoking behavior and the results of microscopic examination of Acid Resistant Bacteria at Maguan Husada Wonogiri General Hospital. This research method is an observational analytic study with a cross sectional design. The sample in this study amounted to 43 respondents obtained using purposive sampling technique. The study was conducted using sputum samples with the Ziehl Neelsen staining method. The statistical test used was Chi Square. The results of the Chi Square test obtained a value of p = 0.045 can be concluded that there is a relationship between smoking behavior and the results of microscopic examination of Acid Resistant Bacteria at Maguan Husada Wonogiri General Hospital.

Abstract

Tuberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis bacteria is still a health problem in the world, including Indonesia. One way to diagnose this disease is by microscopic examination in the form of Acid Resistant Basil (BTA) staining by the Ziehl-Neelsen method using sputum specimens. This check in Indonesia is very often used because the material price is quite affordable and easy to get. Sodium hypochlorite is a disinfectant solution that can be beneficial as a sputum thinner. The purpose of this study was to analyze the degree of positivity of BTA coloring results with the use of NaOCl solution of 0.25%, 0.5% and 1%. The Research Design used in this descriptive descriptive research is Observational. The subject of this study was a suspected sputum sample of TB at Aisyiyah Klaten RSU on February 10, 2025 to February 24, 2025 with a total of 10 samples. From the results of the study obtained degrees of positivity of microscopic TB diagnostics between without use of NaOCl solutions compared with the use of NaOCL 0.25%, 0.5% and 1%. Conclusion: The use of Sodium Hypochlorite (NaOCl) in sputum samples has been shown to increase the degree of positivity of microscopic diagnostic tuberculosis. The number of positive samples increased from 6 samples in the group without NaOCl to 6 postive samples and 2 scanty samples in the group with NaOCl 1%. In addition, there was an increase in positivity score from +1 to +2 in 4 samples and from negative to scanty in 2 samples after the use of NaOCl of 1%. NaOCl concentration 1% is the best concentration capable of increasing the average value of the number of Mycobacterium tuberculosis cells. The average number of BTA cells increased from 809 cells in the group without NaOCl to 2,168 cells in the group with 1% NaOCl. This concentration also converts 2 samples that were initially negative into scanty. In addition, the addition of this bleach solution can also improve the clarity of the visibility of the microscope.

Abstract

Acinetobacter baumannii (Carbapenem Resistant Acinetobacter baumannii CRAB) is a bacterium that is resistant to almost all antibiotics and is difficult to eliminate from the environment. Some CRABs can carry genes that allow bacteria to make carbapenemase enzymes that destroy carbapenem antibiotics. These carbapenemase genes can move between bacteria, increasing the possibility of spreading resistance to various drugs. Commonly detected genes are the oxacillinase group such as OXA-23, ΟΧΑ-24/40, ΟΧΑ-51 and OXA-58. A small portion of CP-CRAB has a mobile gene that encodes carbapenemase which is often found in other gram-negative bacteria, such as Enterobacterales. This study aims to detect whether the OXA-51 and OXA-58 genes are detected in Acinetobacter baumannii isolates, which are genes encoding molecular indicators of resistance to the carbapenem group. The study design was an observational analytical study with a cross-sectional approach. The target population was all Acinetobacter baumannii cultures. From the research results, the sensitivity test showed that all samples were resistant to meropenem and imipenem antibiotics, while the DNA isolation process that had been carried out needed to be optimized to obtain the right isolation procedure in order to obtain clear DNA band visualization results and visualization of the OXA-51 gene showed optimal results, while detection of the OXA-58 gene still needed optimization to be more sensitive and specific. The output of the research for publication has cannot been done yet because the research will be continued to the next stage.

Abstract

Diabetes merupakan gangguan metabolik kronis yang terjadi akibat produksi insulin yang tidak mencukupi atau penurunan efektivitas kerja insulin, yang berujung pada peningkatan kadar glukosa darah (hiperglikemia). Pada tahun 2024, di Indonesia tercatat sebanyak 20.426.400 orang dewasa  menderita  diabetes,  dengan  prevalensi  sebesar  11,3%  dari  total populasi  dewasa  yang  berjumlah  185.217.400  jiwa. Salah - satu efek kronis dari diabetes melitus adalah ulkus kaki diabetik (UKD) yang  merupakan  penyebab  komplikasi  mikroangiopati  neuropati perifer. Tujuan dari  penelitian ini adalah untuk melihat gambaran resistensi Staphylococcus aureus dan Klebsiella pnuemoniae terhadap antibiotik seftriakson dan meropenem pada pasien ulkus kaki diabetik. Penelitian ini merupakan penelitian deskriptif dengan pendekatan retrospektif. Pendekatan retrospektif digunakan untuk mendapatkan data-data spesies bakteri dan hasil uji kepekaan antibiotik bakteri melalui data sekunder yang diambil secara total sampling periode bulan Januari-Desember Tahun 2023-2025. Sampel pada penelitian ini merupakan hasil pemeriksaan kultur pus dan resistensi pada pasien UKD positif bakteri Staphylococcus aureus dan Klebsiella pneumoniae. Hasil penelitian ini didapatkan bahwa terdapat resistensi Staphylococcus aureus terhadap antibiotik seftriakson pada tahun 2025 (20%). Sementara itu, Klebsiella pneumoniae menunjukkan kecenderungan resisten (100%) terhadap seftriakson selama periode penelitian. Seluruh isolat bakteri, baik Staphylococcus aureus maupun Klebsiella pneumoniae tidak menunjukkan resistensi terhadap antibiotik meropenem selama periode 2023–2025.
