Abstract

Curcumin has been reported with an in vitro the cytotoxic effect on several human cancer cells. However, reports on the mode of action and detail mechanism of curcumin in breast cancer disease are limited. Hence, curcumin’s effect on the human breast cancer cell line MCF-7 and MDA-MB-468 was investigated. The MCF-7 and MDA-MB-468 breast cancer cells line were given curcumin in several doses. The anti-proliferation activity of curcumin was determined using the MTS cell viability test and caspase-3 activity was used to detect apoptosis using flowcytometry. The expression of Ras-association domain family 1 isoform A (RASSF1A) and Bax protein in cells was evaluated by ELISA analysis. Kruskal-Wallis followed by the Mann-Whitney test and the Spearman correlation tests were used to asses correlation among RASSF1A, Bax, and caspase-3. Cytotoxicity of curcumin on MCF-7 was lower than that of MDA-MB-468 (75.73 μg/mL and 380.79 μg/mL). The concentration of curcumin at 80 μg/mL induced apoptosis mainly through the intrinsic pathway by caspase-3 activation. Curcumin also showed an anti-proliferative activity as shown by the increase of RASSF1A and Bax protein. Curcumin mediates anti-proliferative and apoptotic effect through the activation of RASSF1A and Bax. Our research data adds information about the role of curcumin in epigenetic events through RASSF1A protein.Keywords: Bax, caspase-3, curcumin, MCF-7, MDA-MB-468, RASSF1A

Abstract

Background : Mobile Phone (MP) is a wireless communication tools that nowadays used by people with many features that can ease the user. Cellphone is releasing electromagnetic radiation that can causes side effect for the user itself, one example of the side effect is disturbing the melatonin hormone synthesis that can cause the abnormal sirkadian body rythm, so the sleep patterns are disrupted, causing insomnia, islam is teaching us to keep our health, for example is to keep the rythm of our bedtime regularly. Objective : Knowing the relation between exposure duration of mobile phone electromagnetic radiation to melatonin level of Wistar albino rats observed by medicine and islam perspectives. Methods : This study was experimental study, total sample was determined by frederer theory and analyzed using IBM SPSS Statistic 21 for Windows. Results : The results showed that melatonin levels in the positive control group (dark control group) 13,705 ± 0,500 were compared with the group which exposed for 3 hours 13,375 ± 0,654 with (p=0,299) and in the positive control group (dark control group) 13,705 ± 0,500 compared with the group which exposed for 6 hours 13,223 ± 0,529 with (p=0,116). Conclusions : There is no significant relation between exposure duration of mobile phone electromagnetic radiation to melatonin levels in Wistar albino rats.

Abstract

Introduction: Curcumin is a polyphenol that has pharmacological activity that can inhibit tumor cell growth and induce apoptosis through various mechanisms. However, the specific mechanism of curcumin cytotoxicity remains controversial because of many anti- and pro-apoptotic signaling pathways in various cell types. This study aims to examine the relationship among curcumin on RASSF1A, Bax protein levels, and caspase-3 activity in supporting the apoptotic mechanism in CSA03 breast cancer cells. Method: Curcumin administration to cancer cells is based on differences in dosage with 24-hour incubation. Cytotoxicity after curcumin administration was determined using MTS. RASSF1A and Bax protein levels were tested through ELISA. Caspase-3 activity was used to determine apoptosis and was tested using flow cytometry. Results: The results indicated that curcumin had a cytotoxicity effect of 40.85 µg/mL. At concentrations of 40 µg/mL and 50 µg/mL, curcumin increases levels of protein RASSF1A (∆ = 26.53% and 47.35%, respectively), Bax (∆ = 48.79% and 386.15%), and caspase-3 (∆ = 1,678.51% and 1,871.889%) significantly. Conclusions: Curcumin exhibits anti-proliferative activity and apoptotic (Caspase-3) effects through activation of RASSF1A and Bax.

Abstract

COVID-19 is a widespread global health problem with a rapid and easy way of transmission. With a high death rate, the government implements a lock down that indirectly reduces the space for people to move. This then relate to the health and productivity of the elderly in achieving healthy aging in the elderly. : This study aims to see if there is a healthy aging movement carried out by the elderly in Sukarame Village, Bandar Lampung during the COVID-19 pandemic. The method in this research is qualitative phenomenology. The study was conducted through interviews with respondents with criteria for healthy and active elderly in Sukarame Village, Bandar Lampung. Samples were determined by means of Purposive Sampling. The data were analyzed using three stages, namely data reduction, data presentation, and drawing conclusions using correlation test bivariate Spearman's Rho. In the study, it was found that 87% of respondents were still physically active. As many as 97% of respondents have a good and sufficient appetite. It was found that 75% of the respondents were non-smokers. Of the total respondents, there are 20% of respondents who are still working. All respondents are still involved in environmental and social activities. All the healthy aging carried out by the respondents are very strongly and strongly related to each other. The mood of the elderly has a significant impact on the satisfaction that the elderly feel. The healthy aging movement was found in respondents in Sukarame Village, Bandar Lampung.

Abstract

The aging process that feeled by elderly will cause a decrease in functional and cellular functions. The factors that influence this process are divided into two factors, internal factors such as free radicals and external factors such as lifestyle, environment, and food. Free radicals in the body can be neutralized by antioxidants that come from a good lifestyle and foods that contain antioxidants. This study uses a qualitative ethnographic method by conducting interviews using a questionnaire combined with respondent observations. The questionnaire used is divided into three analyzes, there are symptom analysis, lifestyle analysis, and food analysis. Then the scores on the three analyzes will be added up and produce qualitative conclusions. The sample was determined by purposive sampling with analysis of the relationship between each of independent variables using a correlation test using SPSS application. The symptoms and the lifestyle lived by the elderly in Kelurahan Sukarame, Bandar Lampung play an important role in the personal antioxidant profile while the food intake chosen by the elderly has a lower role than symptoms and lifestyle. Physical activities such as exercise and morning walks and minimizing UV exposure with a combination of taking vitamin supplements routinely will maximize the aging process of the elderly in Kelurahan Sukarame, Bandar Lampung.

Abstract

Corona Virus Disease (COVID-19) teridentifikasi di Indonesia pada awal Maret 2020. Salah satu golongan yang terdampak pandemi COVID-19 adalah lansia. Penurunan kinerja fisik dalam melakukan aktivitas seiring bertambahnya usia dapat mengakibatkan menurunnya harga diri pada lanjut usia. Penelitian ini bertujuan untuk menganalisa apakah terdapat hubungan kualitas hidup terhadap harga diri lansia selama pandemi COVID-19. Jenis penelitian yang digunakan adalah deskriptif korelasi dengan pendekatan cross sectional. Populasi penelitian ini yaitu lansia 60 tahun ke atas yang tinggal di Kelurahan Pulo Gadung Jakarta Timur. Total 50 responden yang dipilih secara acak dengan penetapan sampel menggunakan cluster random sampling. Peneliti merandom dari jumlah populasi lansia berdasarkan daerah populasi untuk dijadikan sampel penelitian. Variabel independen adalah kualitas hidup dan variabel dependen adalah harga diri. Analisa data dilakukan secara analisa bivariat dengan alat bantu program SPSS. Hasil penelitian menunjukkan terdapat hubungan yang signifikan antara kualitas hidup terhadap harga diri rendah pada lansia selama pandemi COVID-19 dengan nilai p 0,039
