Abstract

Diabetes mellitus (DM) is a chronic condition which caused by insulin secretion disorder, insulin resistance or both. High number of diabetics per year has led many researchers to conduct research to find the most safe and effective medicine for DM treatment. Flavonoid are known to act as inhibitors of α-amylase as a parameter of pancreatic damage and help lower blood glucose levels. One source of flavonoids is purslane. The aim of this study was to examine the effect of purslane ethanol extract on glucose and α-amylase levels in rat. Twenty rats Rattus novergicus L. strain Wistar are divided into 5 groups; control, positive control, and three test groups, each was given purslane ethanol extract of 100 mg/kgBW, 200 mg/kgBW, and 300 mg/kgBW. Blood glucose and body weight were measured before alloxan induction, after induction, as well as day- 7, 14, 21, and 28; while feed and drink consumption were measured everyday starting with the administration of purslane ethanol extract. The α-amylase test was carried out on the 28th day using enzymatic UV determination of degradation product of maltose and glucose method. The analysis result showed that rats blood glucose and α-amylase decreased in each group with purslane extract. The antioxidant content of flavonoids in purslane ethanol extract has been shown to have anti-diabetic effects by repairing pancreatic function.

Abstract

Suku Madura merupakan salah satu etnis di Indonesia yang hingga kini mempertahankan pengetahuan tradisional dalam pemanfaatan tumbuhan sebagai obat. Masyarakat Madura, khususnya di Sumenep dan Pamekasan, menjaga kearifan lokal dalam penggunaan tanaman obat untuk mengatasi diabetes, seperti jamu kencing manis yang dikenal di Kecamatan Pamekasan. Jamu ini memanfaatkan beberapa jenis tumbuhan, antara lain daun insulin (Tithonia diversifolia), sirih merah (Piper crocatum), kayu manis (Cinnamomum burmannii), ketumbar (Coriandrum sativum), dan temulawak (Curcuma xanthorrhiza). Penelitian ini bertujuan menganalisis potensi jamu kencing manis madura terhadap struktur limpa tikus diabetes melitus. Penelitian menggunakan 30 ekor tikus wistar jantan dibagi menjadi enam kelompok perlakuan: kontrol P0 (akuades), P1 (diabetes + metformin), P2 (diabetes + 5 g jamu), P3 (diabetes + 10 g jamu), P4 (diabetes + 15 g jamu), dan P5 (diabetes tanpa terapi). Tikus dieuthanasia pada hari ke-3, 10, dan 20, kemudian limpa diisolasi dan difiksasi. Preparat histologis dibuat menggunakan metode parafin dan pewarnaan hematoksilin-eosin. Parameter yang diamati meliputi bobot badan, bobot limpa, indeks organ, serta diameter dan luas germinal center serta pulpa putih. Data dianalisis dengan uji sidik ragam satu arah, dilanjutkan uji Duncan, sedangkan parameter non parametrik diuji dengan Kruskal-Wallis. Hasil menunjukkan terdapat perbedaan nyata pada bobot badan (p<0,05), namun bobot limpa, indeks organ, diameter, dan luas germinal center tidak berbeda nyata (p>0,05), begitu pula diameter dan luas pulpa putih tidak menunjukkan perbedaan signifikan. Disimpulkan bahwa jamu kencing manis madura tidak berdampak negatif pada struktur histologi limpa tikus diabetes melitus.
