Abstract

Abstract: The process of extracting secondary metabolite compounds from plants through extraction. One of factor that influences the success of extraction is the type of solvent used. This study aims to determine the effect of solvent type on the contents of secondary metabolite compounds and the antioxidant activity of Lino (G. koordersiana Burret) bark extract. This research began with taking lino bark samples, extraction, phytochemical screening, content analysis and antioxidant activity tests. The research results showed that the yields for ethanol, methanol, ethyl acetate and n-hexane extracts were 42.15%, 41.65%, 0.98% and 0.62%, respectively. The phytochemical test results of ethanol and methanol extracts contain alkaloids, flavonoids, phenolics, triterpenoids, saponins and tannins. Meanwhile, ethyl acetate extract contains phenolics, triterpenoids and tannins and n-hexane extract contains triterpenoids. The results of analysis of secondary metabolite on the contents for ethanol, methanol, ethyl acetate and n-hexane extracts showed different results. Antioxidant activity test results for each ethanol, methanol, ethyl acetate and n-hexane extract with different IC50 values. Antioxidant activity of ethanol, methanol and ethyl acetate extracts has antioxidant activity in the very strong category. Meanwhile, n-hexane extract has antioxidant activity in the inactive category. The results showed that the type of solvent affected the levels and antioxidant activity of secondary metabolite compounds from Lino (G. koordersiana Burret) extract.

Abstract

Exploration of the potential of alternative ingredients to replace black tea or green tea in making kombucha has begun to be widely carried out. On Timor Island, faloak stem bark is processed into a traditional drink by some locals. For this reason, the use of faloak stem bark as a basic ingredient for kombucha needs to be studied, especially in exploring its antibacterial potential, which is analyzed based on the concentration of sugar used. This study aims to determine the chemical characteristics and antibacterial activity of faloak stem bark kombucha with various concentrations of sugar. The methods involved making faloak stem bark kombucha with varying sugar concentrations of 6, 8, 10, and 12% (w/v), and testing chemical characteristics in the form of pH, total acid, total sugar, and total phenol tests. Meanwhile, the antibacterial activity testing used a good diffusion method against Escherichia coli ATCC 25922 bacteria. After 14 days of fermentation, faloak stem bark kombucha with a 12% sugar concentration showed the best chemical characteristics, with pH, total acid, total sugar, and total phenol values of 2.91, 1.19%, 7.33%, and 420.15 mg GAE/L, respectively. Furthermore, antibacterial activity also showed the highest inhibition (13.02 mm) at a sugar concentration of 12%. Thus, faloak bark kombucha made with a sugar concentration of 12% has the potential to be antibacterial.

Abstract

The goal of this study was to genetically link natural materials derived from Tinospora crispa L with Berberine to dihydrofolate reductase-thymidylate synthase (DHFR-TS). Method: The ligand berberine (CID: 2353) was obtained from Pubchem, while the protein DHFR-TS (PDB ID 2bl9) was obtained from Protein Data Bank. The ligands and proteins interacted with HEX 8.0.0.0 and were visualized with Discovery Studio. The researchers discovered a positive interaction between berberine and DHFR-TS, observed at four amino acid residues that bind to the protein TYR125, ILE121, LEU45, and MET54. Van der Waals interactions, hydrogen bonds, Pi-Sulfur, Pi-Alkyl, and Pi-Stalked interactions all contribute to strength and stability. In conclusion, berberine has the potential to act as a DHFR-TS inhibitor and thus prevent malaria.ABSTRAK: Tujuan penelitian ini adalah memanfaatkan bahan alam yang berasal dari Tinospora crispa L dengan kandungan utama Berberin dengan dihydrofolate reductase-thymidylate synthase (DHFR-TS) secara genetik. Metode, Ligan berberin (CID: 2353 ) diperoleh dari Pubchem sedangkan protein DHFR-TS (PDB ID 2bl9) diperoleh dari Protein Data Bank, ligan dan protein diinteraksikan menggunakan HEX 8.0.0.0 dan divisualisasikan menggunakan discovery studio. Ditemukan interaksi positif antara berberin dan DHFR-TS yang menunjukkan interaksi pada empat residu asam amino yang berikatan dengan protein. Mereka adalah TYR125, ILE121, LEU45 dan MET54. Beberapa interaksi yang dilakukan Van der Waals, ikatan hidrogen, Pi-Sulfur, Pi-Alkyl dan Pi-Stalked juga memberikan dukungan dalam rangka meningkatkan kekuatan dan stabilisasi. Kesimpulannya, berberin memiliki potensi fungsi sebagai penghambat DHFR-TS dan mengarah pada malaria.

Abstract

Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dari propolis sarang lebah Trigona spp asal Tenau Kupang. Senyawa metabolit sekunder dari propolis sarang lebah sampel diekstraksi dengan metode maserasi menggunakan etanol 70%. Identifikasi golongan senyawa dalam ekstrak kasar, dilakukan menggunakan metode skrining fitokimia. Uji aktivitas antibakteri dilakukan menggunakan metode sumur difusi terhadap dua jenis bakteri patogen dan dua jenis bakteri non patogen. Hasil penelitian, diperoleh bahwa ekstrak etanol propolis sarang lebah sampel berwarna merah tua dan bersifat engket. Hasil uji fitokimia, mengandung golongan terpenoid, alkaloid, dan flavonoid. Akrivitas antibakteri bersifat kuat atau bakterisidalt erhadap bakteri uji golongan patogen yakni E. Coli   dan S. tiphymurium  dengan Konsentrasi Hambat Tumbuh Minimum  masing-masing  0,52%, dan   0,52%, Aktivitas antibakteri bersifat lemah terhadap bakteri uji golongan non patogen yakni L. Casei dan Bacterroides sp, dengan Konsentrasi Hambat Tumbuh Minimum  masing-masing   2,08%, dan 2,08%. Disimpulkan bahwa propolis dari sampel sarang lebah Trigona spp asal Tenau Kupang mengandung golongan senyawa antibakteri dan bersifat bakterisidal terhadap bak terijenis patogen.
