Abstract

Introduction: The use of traditional medicines derived from the active ingredients of several types of plants is preferred by the community in treating several types of diseases. One of the plants that has been used by the community as medicine is purple leaves with the Latin name Grapthophylum pictum (L.) Griff. These leaves are traditionally used to treat rheumatism, menstruation, hemorrhoids, urinary tract infections, scabies, swelling, wounds, dermatitis, ear diseases, laxatives, and cancer. The aim of this study was to determine the total levels of secondary metabolites from purple leaf extract ethanol (PLEE) and evaluate the effect of administering PLEE on acute toxicity (LD50) in Wistar rats. Methods: This research is a laboratory experiment with a post test only controlled group design. The research subjects were 20 male Wistar rats, divided into 1 control group and 3 treatment groups, each consisting of 5 rats. The control group only received distilled water, the Treatment I group (P1) was given a suspension of the purple leaf extract test preparation at a dose of 500 mg/kg. Treatment group II (P2), received a suspension of the test preparation at a dose of 2,000 mg/kg, while Treatment group III (P3) received a suspension of the test preparation at a high dose of 5,000 mg/kg of mice. The test preparation was given orally with only one administration at the beginning of the research period and observation was carried out for 14 days. Data was obtained if the rats died after being given treatment. Results: The results showed that the total secondary metabolite content of purple leaves was alkaloids (24,725.99 mg/g), saponins (89.191 mg/g), flavonoids (6.332 mg/g) and tannins (0.884 mg/g), while based on toxicity the PLEE classification was mildly toxic with an LD50 value of 3,890 mg/kg in accordance with BPOM RI's acute toxicity potential at toxicity level 4 (oral LD50 500 – 5,000 mg/kg). Conclusion: PLEE showed the highest total content of secondary metabolites in alkaloids, and consumption of PLEE in high doses caused the death of rats with a mild toxic classification.

Abstract

Self-medication is a common activity carried out by a person when he is sick. Proper self-medication by providing information provided by health workers will help the patient's healing process by optimizing therapy. Irrational management can result in various disadvantages such as medication errors due to inaccurate self-diagnosis; drug use is not appropriate because of the acquisition of incorrect information from the media; waste of time and costs; can cause unwanted drug reactions such as sensitivity, allergies, side effects or resistance. The use of drugs is said to be rational, if the patient receives the right drug for clinical needs, in doses that meet the needs for a sufficient period of time, and at affordable costs for both individuals and society. This activity was carried out with the aim to increase the understanding and knowledge of the community in Sejahtera Village, Palolo District, Sigi Regency, especially about rational medicine and the provision social assistance. Counseling is given by interactive lecture method through posters and distribution of leaflets. The effectiveness of the extension was tested by giving a partner satisfaction questionnaire to the activities carried out. The results of the activity observations showed that the Partners were satisfied with the outreach activities carried out.

Abstract

Traditional herbal medicine has become increasingly popular as an alternative to conventional medical treatments. Although the plant offers numerous health benefits, caution should be exercised regarding its long-term use. Long-term use of traditional herbal remedies can have detrimental effects on the kidneys. This study was designed to assess the histological changes in the kidneys of male rats following 28 days of purple sage leaf extract administration and to identify potential harmful effects. The study utilized 20 male Wistar rats and utilized a randomized group design for the experimental methodology. Rats were categorized into four distinct groups, consisting of one control group and three treatment groups. The control group received solely sodium chloride, whereas the treatment group 1 (P1) received 500 mg/kg BB of purple sage ethanol extract. Treatment group 2 received up to 2000 mg/kg BB of purple sage ethanol extract, while treatment group 3 received up to 5000 mg/kg BB. The oral examination was conducted for a duration of 28 days. The treatment groups showed an increase in weight and organ index of the right kidney. The findings suggest a potential for toxicity resulting from the administration of purple sage leaf extract over a duration of 28 days.
