Abstract

This research aims to determine the total chlorophyll of jabon merah leaves extract using acetone and ethanol solvent and to determine the difference in total chlorophyll of jabon merah leaves extract using acetone dan ethanol solvent. The result of the analysis of chlorophyll levels in the extraction of jabon merah leaves using the SPAD-502 method showed that the base and the middle of the jabon merah leaf were the part that had the highest chlorophyll content, and the tip of the jabon merah leaf has a lower chlorophyll content. In the spectrophotometric method using acetone solvent, the result of chlorophyll a is 34.543,9 mg/L, and chlorophyll b is 30.338,6 mg/L. The result of chlorophyll using ethanol solvent is 33.171,8 mg/L for chlorophyll a and 19.065,9 mg/L for chlorophyll b. The total chlorophyll content in jabon merah leaves with acetone solvent is 65,346,4 mg/L, and for ethanol solvent, ia 52.474 mg/L. The results show that acetone is a good solvent for extracting jabon merah leaves.

Abstract

This study investigated the diversity and density of rhizosphere fungi in the post-nickel mining land of PT. Vale Indonesia Tbk., to assess its potential in supporting revegetation and ecological recovery. By analyzing fungal populations at various dilution levels in PDA media, this study revealed the relationship between post-reclamation duration and increased soil microbiological activity. The results showed that longer reclaimed land had denser fungal populations, especially at lower dilutions, indicating the importance of rhizosphere fungi in soil health recovery. This study confirmed that the presence of plants significantly affected the abundance and diversity of rhizosphere fungi. These findings provide insight into the importance of selecting specific fungal species for post-mining land revegetation strategies, which support plant growth and accelerate ecological recovery. It can contribute to developing best practices in ecological engineering and environmental management in post-mining areas.

Abstract

Penghitungan jumlah kromosom dapat dilakukan dengan cara analisis kromosom dengan perlakuan pretretment. Merupakan suatu metode pembuatan preparate kromosom yang tepat dalam menghasilkan pemisahan kromosom yang jelas baik, sehinggah memudahkan dalam penghitungan kromosom. Penelitian ini bertujuan untuk mengetahui pengaruh lama pemberian hidroksiquinolin pada pembuatan preparate akar tanaman cabai rawit (Capsicum frutescens). Penelitian ini meliputi dua kegiatan yaitu persiapan bahan serta pembuatan preparat kromosom akar cabai. Ada dua hal yang diamati pada preparat kromosom akar cabai, yaitu kemampuan pemisahan kromosom dan ketajaman pewarnaan kromosom dibandingkan dengan sitoplasma sekelilingnya. Kesimpulan pada penelitian ini adalah adalah jumlah kromosom cabai rawit 2n=2x=24,  analisis kromosom akar maristematis pada tanaman cabai rawit dengan pemberian perlakuan lama perendaman hidroksiquinolin selama 4 jam setelah di fiksasi menggunakan larutan farmer dapat menghasilkan tampilan pemisahan kromosom terbaik dibawah mikroskop lebih jelas dan memudahkan dalam penghitungan jumlah kromosom.
