Abstract

Cellulolytic bacteria can produce cellulase enzymes, which allow them to degrade cellulose into simpler chemicals while using it as a growth substrate. These bacteria have drawn a lot of interest due to their unique properties and potential uses, such as in agriculture and the food industry. Soil is one of the habitats of cellulolytic bacteria. This study aims to determine the potential and screening of cellulolytic bacterial isolates. Cellulolytic bacteria were isolated from soil samples and cultured using selective Carboxyl Methyl Cellulose (CMC) media. Five purified isolates were collected, and their cellulase enzyme activity was qualitatively assessed using the Congo red test. Isolated bacteria code Area Jalan Setapak (AJS1) produced a higher clear zone index and will be used for further testing. Quantitative analysis of isolate AJS1 revealed its highest enzyme activity after ten minutes of incubation, with 0.0049 U/mL unit activity and 0.018 U/mg specific activity. Based on morphological and biochemical characteristics, isolated AJS1 was identified as Corynebacterium spp.

Abstract

A review of water quality from the results of research on Java and Kalimantan has been carried out to get an illustration of water contamination level (refill drinking water, PDAM water, and well water) by Coliform bacteria. Data was obtained from secondary sources of research results from several regions in Java and Kalimantan. The results of water quality assessment of the two islands are presented as a percentage. Based on the results, the level of water contamination on Java island is higher than Kalimantan. The average percentage of Coliform contamination in water on Java is 61.42% and only 42.26% in Kalimantan. The high percentage of water contamination by Coliform bacteria in Java is due to the fact that Java has a higher source of contaminants. The existence of Coliform sources was influenced by the population, housing, and organic waste production.

Abstract

 Phosphorus compound in the critical land is relatively low that caused phosphorus is bonded to metals. The isolate BRC-AP and BRC-AK are a phosphate solubilizing bacteria (PSB) indigenous critical land that has the ability to dissolve phosphate. The used of phosphate solubilizing bacteria are able to improve plant growth on critical lands. The aimed of this study was to measure the growth rate of shoot length, root length and biomass of root, and leaf cuttings elephant grass (Pennisetum purpureum cv. King Grass) which inocu-lated with the consortium of phosphate solubilizing bacteria and IAA producing mold. Phosphate dissolving activity is determined by the formation of clear zone in solid Pikovskaya medium, synergism assay between bacteria and fungi observed on SDA medium then proceed testing the ability of bacteria and fungi consortium to promote plant growth. The result showed that the highest of phosphate solubility in-dex was found in isolate BRC-AP (27), addition of the combination of isolate BRC-AP, BRC-AK and Humicola sp. R.Dn have significant effect on shoot growth in the first week and leaf dry weight. The high-est length shoot of cutting elephant grass was found in treatment with the consortium of phosphate solubilizing bacteria and IAA producing mold (BRC-AP + BRC-AK + KP) (58.66 cm). The highest of leaf dry weight was found in treatment with BRC-AP + BRC-AK + KP (5.65 gr). The addition of consortium phosphate solubilizing bacteria and IAA producing fungi from South Kalimantan critical land proved can improve plant growth especially length shoot and leaf dry weight.

Abstract

Diseases in rice plants (paddy) caused by microorganisms such as Pyricularia oryzae lead to a decrease in rice production. Therefore, it is essential to find out biological agents for protecting paddy and plants in general, against plant diseases. Liquid smoke and endophytic fungi have been known as biological agents to enhance the protection of plants against disease. The purpose of this study was to determine the ability of liquid smoke, endophytic fungi and the concentrations combinations to suppress the growth of P. oryzae. The results showed that liquid smoke concentrations of 0.17% to 1.75% and endophytic fungi filtrate of 2% to 10% showed significant ability against pathogen P. oryzae. However, the combination of liquid smoke and endophytic fungi filtrate at selected concentrations (0.17% liquid smoke combined with 2% endophytic fungi filtrate and 0.34% liquid smoke combined with 2% endophytic fungi filtrate) showed no significant inhibition percentage against  P. oryzae compared to control. In conclusion, this study showed that the respective applications of liquid smoke and endophytic fungi filtrate inhibit the growth of P. oryzae.
